    Comparative study of Cl₂, Cl₂/O₂, and Cl₂/N₂ inductively coupled plasma processes for etching of high-aspect-ratio photonic-crystal holes in InP

    An extensive investigation has been performed on inductively coupled plasmaetching of InP. An important motivation for this work is the fabrication of high-aspect-ratio holes for photonic crystals. The essential chemistry is based on Cl₂ with the addition of N₂ or O₂ for sidewall passivation. The influence of different process parameters such as gas flows, temperature,pressure, ion energy, and inductively coupled plasma power on the hole geometry is presented. It is concluded that photonic crystals can be etched with Cl₂ only; however, temperature and pressure control is critical. Adding passivation gases largely broadens the window in the parameter space for hole etching. Most importantly, etching of narrow holes can be carried out at higher temperatures where the etching is mass limited and spontaneous etching of InP by Cl₂ occurs.Part of this research is supported by NanoNed, a technology program of the Dutch Ministry of Economic Affairs

    Rehabilitation patients: undernourished and obese?

    BACKGROUND: The aim of this study was to assess the prevalence of undernutrition in patients in Dutch rehabilitation centres and to measure the diagnostic accuracy of available screening tools. METHODS: This cross-sectional multicentre study was conducted in 11 rehabilitation centres in The Netherlands. Patient's nutritional status was determined by the amount of weight loss during the last 1, 3 and 6 months and body mass index (BMI). Diagnostic accuracy was assessed for 5 screening tools: Short Nutritional Assessment Questionnaire (SNAQ), Short Nutritional Assessment Questionnaire Residential Care (SNAQRC), SNAQ65+, Malnutrition Universal Screening Tool and Mini nutrition Assessment-short form. RESULTS: Twenty-eight percent of the patients were severely undernourished and 10% were moderately undernourished. In the undernourished group, 28% were overweight (BMI 25-30) and 19% were obese (BMI > 30). The SNAQ65+ is the recommended screening tool due to its high diagnostic accuracy (sensitivity 96%, specificity 77%, positive predictive value 62%, negative predictive value 90%) and quick and easy use. The MNA had the worst diagnostic accuracy, with a sensitivity of 44%. CONCLUSION: The prevalence of undernutrition in patients in Dutch rehabilitation centres is high. Almost half of the undernourished patients were overweight or obese. Therefore, it is important not only to screen for undernutrition, but also carefully to assess possible overweight/obesity in every undernourished rehabilitation patient

    Integrability of a conducting elastic rod in a magnetic field

    We consider the equilibrium equations for a conducting elastic rod placed in a uniform magnetic field, motivated by the problem of electrodynamic space tethers. When expressed in body coordinates the equations are found to sit in a hierarchy of non-canonical Hamiltonian systems involving an increasing number of vector fields. These systems, which include the classical Euler and Kirchhoff rods, are shown to be completely integrable in the case of a transversely isotropic rod; they are in fact generated by a Lax pair. For the magnetic rod this gives a physical interpretation to a previously proposed abstract nine-dimensional integrable system. We use the conserved quantities to reduce the equations to a four-dimensional canonical Hamiltonian system, allowing the geometry of the phase space to be investigated through Poincar\'e sections. In the special case where the force in the rod is aligned with the magnetic field the system turns out to be superintegrable, meaning that the phase space breaks down completely into periodic orbits, corresponding to straight twisted rods.Comment: 19 pages, 1 figur

    Crop growth models for the -omics era: the EU-SPICY project

    The prediction of phenotypic responses from genetic and environmental information is an area of active research in genetics, physiology and statistics. Rapidly increasing amounts of phenotypic information become available as a consequence of high throughput phenotyping techniques, while more and cheaper genotypic data follow from the development of new genotyping platforms. , A wide array of -omics data can be generated linking genotype and phenotype. Continuous monitoring of environmental conditions has become an accessible option. This wealth of data requires a drastic rethinking of the traditional quantitative genetic approach to modeling phenotypic variation in terms of genetic and environmental differences. Where in the past a single phenotypic trait was partitioned in a genetic and environmental component by analysis of variance techniques, nowadays we desire to model multiple, interrelated and often time dependent, phenotypic traits as a function of genes (QTLs) and environmental inputs, while we would like to include transcription information as well. The EU project 'Smart tools for Prediction and Improvement of Crop Yield' (KBBE-2008-211347), or SPICY, aims at the development of genotype-to-phenotype models that fully integrate genetic, genomic, physiological and environmental information to achieve accurate phenotypic predictions across a wide variety of genetic and environmental configurations. Pepper (Capsicum annuum) is chosen as the model crop, because of the availability of genetically characterized populations and of generic models for continuous crop growth and greenhouse production.     InP-based two-dimensional photonic crystals filled with polymers

    Polymer filling of the air holes of Indium Phosphide based two-dimensional photonic crystals is reported. After infiltration of the holes with a liquid monomer and solidification of the infill in situ by thermal polymerization, complete filling is proven using scanning electron microscopy. Optical transmission measurements of a filled photonic crystal structure exhibit a redshift of the air band, confirming the complete filling.Comment: To be published in Appl. Phys. Let

    Single-molecule studies of the stringency factors and rates governing the polymerization of RecA on double-stranded DNA

    RecA is a key protein in homologous recombination. During recombination, one single-stranded DNA (ssDNA) bound to site I in RecA exchanges Watson–Crick pairing with a sequence-matched ssDNA that was part of a double-stranded DNA molecule (dsDNA) bound to site II in RecA. After strand exchange, heteroduplex dsDNA is bound to site I. In vivo, direct polymerization of RecA on dsDNA through site I does not occur, though it does in vitro. The mechanisms underlying the difference have been unclear. We use single-molecule experiments to decouple the two steps involved in polymerization: nucleation and elongation. We find that elongation is governed by a fundamental clock that is insensitive to force and RecA concentration from 0.2 and 6 µM, though rates depend on ionic conditions. Thus, we can probe nucleation site stability by creating nucleation sites at high force and then measuring elongation as a function of applied force. We find that in the presence of ATP hydrolysis a minimum force is required for polymerization. The minimum force decreases with increasing RecA or ATP concentrations. We propose that force reduces the off-rate for nucleation site binding and that nucleation site stability is the stringency factor that prevents in vivo polymerization

    The Role of Interleukin-4 and Interleukin-10 in Osteoarthritic Joint Disease: A Systematic Narrative Review

    OBJECTIVE: A fusion protein of interleukin-4 and interleukin-10 (IL4-10 FP) was developed as a disease-modifying osteoarthritis drug (DMOAD), and chondroprotection, anti-inflammation, and analgesia have been suggested. To better understand the mechanisms behind its potential as DMOAD, this systematic narrative review aims to assess the potential of IL-4, IL-10 and the combination of IL-4 and IL-10 for the treatment of osteoarthritis. It describes the chondroprotective, anti-inflammatory, and analgesic effects of IL-4, IL-10, and IL4-10 FP. DESIGN: PubMed and Embase were searched for publications that were published from 1990 until May 21, 2021 (moment of search). Key search terms were: Osteoarthritis, Interleukin-4, and Interleukin-10. This yielded 2,479 hits, of which 43 were included in this review. RESULTS: IL-4 and IL-10 showed mainly protective effects on osteoarthritic cartilage in vitro and in vivo, as did IL4-10 FP. Both cytokines showed anti-inflammatory effects, but also proinflammatory effects. Only in vitro IL4-10 FP showed purely anti-inflammatory effects, indicating that proinflammatory effects of one cytokine can be counteracted by the other when given as a combination. Only a few studies investigated the analgesic effects of IL-4, IL-10 or IL4-10 FP. In vitro, IL-4 and IL4-10 FP were able to decrease pain mediators. In vivo, IL-4, IL-10, and IL4-10 FP were able to reduce pain. CONCLUSIONS: In conclusion, this review describes overlapping, but also different modes of action for the DMOAD effects of IL-4 and IL-10, giving an explanation for the synergistic effects found when applied as combination, as is the case for IL4-10 FP

    Diagnostic value of a heart-type fatty acid-binding protein (H-FABP) bedside test in suspected acute coronary syndrome in primary care

    AbstractBackgroundTo determine the diagnostic accuracy of a rapid heart-type fatty acid-binding protein (H-FABP) test in patients suspected of acute coronary syndrome (ACS) in primary care.MethodsGeneral practitioners included 298 patients suspected of ACS. In all patients, whether referred to hospital or not, ECG and cardiac biomarker testing was performed. ACS was determined in accordance with international guidelines. Multivariate analysis was used to determine the value of H-FABP in addition to clinical findings.ResultsMean patient age was 66years (SD 14), 52% was female and 66 patients (22%) were diagnosed with ACS. The H-FABP bedside test was performed within 24h (median 3.1, IQR 1.5 to 7.1) after symptom onset. The positive predictive value (PPV) of H-FABP was 65% (95% confidence interval (CI) 50–78). The negative predictive value (NPV) was 85% (95% CI 80–88). Sensitivity was 39% (29–51%) and specificity 94% (90–96%). Within 6h after symptom onset, the PPV was 72% (55–84) and the NPV was 83% (77–88), sensitivity 43% (31–57%) and specificity 94% (89–97%). Adding the H-FABP test to a diagnostic model for ACS led to an increase in the area under the receiver operating curve from 0.66 (95% CI 0.58–0.73) to 0.75 (95% CI 0.68–0.82).ConclusionThe H-FABP rapid test provides modest additional diagnostic certainty in primary care. It cannot be used to safely exclude rule out ACS. The test can only be used safely in patients otherwise NOT referred to hospital by the GP, as an extra precaution not to miss ACS (‘rule in’)

    Immune response profiles of calves following vaccination with live BCG and inactivated Mycobacterium bovis vaccine candidates

    <div><p>Conventional control and eradication strategies for bovine tuberculosis (BTB) face tremendous difficulties in developing countries; countries with wildlife reservoirs, a complex wildlife-livestock-human interface or a lack of veterinary and veterinary public health surveillance. Vaccination of cattle and other species might in some cases provide the only suitable control strategy for BTB, while in others it may supplement existing test-and-slaughter schemes. However, the use of live BCG has several limitations and the global rise of HIV/AIDS infections has furthermore warranted the exploration of inactivated vaccine preparations. The aim of this study was to compare the immune response profiles in response to parenteral vaccination with live BCG and two inactivated vaccine candidates in cattle.</p><p>Twenty-four mixed breed calves (<i>Bos taurus</i>) aged 4–6 months, were allocated to one of four groups and vaccinated sub-cutaneously with live <i>M</i>. <i>bovis</i> BCG (Danish 1331), formalin-inactivated <i>M</i>. <i>bovis</i> BCG, heat-killed <i>M</i>. <i>bovis</i> or PBS/Montanide™ (control). Interferon-γ responsiveness and antibody production were measured prior to vaccination and at weekly intervals thereafter for twelve weeks. At nine weeks post-priming, animals were skin tested using tuberculins and MTBC specific protein cocktails and subsequently challenged through intranodular injection of live <i>M</i>. <i>bovis</i> BCG.</p><p>The animals in the heat-killed <i>M</i>. <i>bovis</i> group demonstrated strong and sustained cell-mediated and humoral immune responses, significantly higher than the control group in response to vaccination, which may indicate a protective immune profile. Animals in this group showed reactivity to the skin test reagents, confirming good vaccine take. Lastly, although not statistically significant, recovery of BCG after challenge was lowest in the heat-killed <i>M</i>. <i>bovis</i> group.</p><p>In conclusion, the parenteral heat-killed <i>M</i>. <i>bovis</i> vaccine proved to be clearly immunogenic in cattle in the present study, urging further evaluation of the vaccine in challenge studies using virulent <i>M</i>. <i>bovis</i> and assessment of vaccine efficacy in field conditions.</p></div
