Mitotic Exit Network Controls the Localization of Cdc14 to the Spindle Pole Body in Saccharomyces cerevisiae
AbstractBudding yeast Cdc14 phosphatase plays essential roles in mitotic exit. Cdc14 is sequestered in the nucleolus by its inhibitor Net1/Cfi1 and is only released from the nucleolus during anaphase to inactivate mitotic CDK. It is believed that the mitotic exit network (MEN) is required for the release of Cdc14 from the nucleolus because liberation of Cdc14 by net1/cfi1 mutations bypasses the essential role of the MEN. But how the MEN residing at the spindle pole body (SPB) controls the association of Cdc14 with Net1/Cfi1 in the nucleolus is not yet understood [1, 2]. We found that Cdc14-5GFP was released from the nucleolus in the MEN mutants (tem1, cdc15, dbf2, and nud1), but not in the cdc5 cells during early anaphase. The Cdc14 liberation from the nucleolus was inhibited by the Mad2 checkpoint and by the Bub2 checkpoint in a different manner when microtubule organization was disrupted. We observed Cdc14-5GFP at the SPB in addition to the nucleolus. The SPB localization of Cdc14 was significantly affected by the MEN mutations and the bub2 mutation. We conclude that Cdc14 is released from the nucleolus at the onset of anaphase in a CDC5-dependent manner and that MEN factors possibly regulate Cdc14 release from the SPB
The Lyman Continuum Escape Survey: Ionizing Radiation from [O III]-Strong Sources at a Redshift of 3.1
We present results from the LymAn Continuum Escape Survey (LACES), a Hubble
Space Telescope (HST) program designed to characterize the ionizing radiation
emerging from a sample of Lyman alpha emitting galaxies at redshift . As many show intense [O III] emission characteristic of
star-forming galaxies, they may represent valuable low redshift analogs of
galaxies in the reionization era. Using HST Wide Field Camera 3 / UVIS
to image Lyman continuum emission, we investigate the escape fraction of
ionizing photons in this sample. For 61 sources, of which 77% are
spectroscopically confirmed and 53 have measures of [O III] emission, we detect
Lyman continuum leakage in 20%, a rate significantly higher than is seen in
individual continuum-selected Lyman break galaxies. We estimate there is a 98%
probability that of our detections could be affected by foreground
contamination. Fitting multi-band spectral energy distributions (SEDs) to take
account of the varying stellar populations, dust extinctions and metallicities,
we derive individual Lyman continuum escape fractions corrected for foreground
intergalactic absorption. We find escape fractions of 15 to 60% for individual
objects, and infer an average 20% escape fraction by fitting composite SEDs for
our detected samples. Surprisingly however, even a deep stack of those sources
with no individual detections provides a stringent upper limit on the
average escape fraction of less than 0.5%. We examine various correlations with
source properties and discuss the implications in the context of the popular
picture that cosmic reionization is driven by such compact, low metallicity

Observation of a Highly Spin Polarized Topological Surface State in GeBiTe
Spin polarization of a topological surface state for GeBiTe, the
newly discovered three-dimensional topological insulator, has been studied by
means of the state of the art spin- and angle-resolved photoemission
spectroscopy. It has been revealed that the disorder in the crystal has a minor
effect on the surface state spin polarization and it exceeds 75% near the Dirac
point in the bulk energy gap region (180 meV). This new finding for
GeBiTe promises not only to realize a highly spin polarized surface
isolated transport but to add new functionality to its thermoelectric and
thermomagnetic properties.Comment: 5 pages, 4 figure
A hard ionizing spectrum in z=3-4 Lyα emitters with intense [OIII] emission: Analogs of galaxies in the reionization era?
We present Keck/MOSFIRE spectra of the diagnostic nebular emission lines [O iii]λλ5007,4959, [O ii]λ3727, and Hβ for a sample of 15 redshift z≃3.1\mbox-3.7 Lyα emitters (LAEs) and Lyman break galaxies (LBGs). In conjunction with spectra from other surveys, we confirm earlier indications that LAEs have a much higher [O iii]/[O ii] line ratio than is seen in similar redshift LBGs. By comparing their distributions on a [O iii]/[O ii] versus R23 diagram, we demonstrate that this difference cannot arise solely because of their lower metallicities but most likely is due to a harder ionizing spectrum. Using measures of Hβ and recombination theory, we demonstrate, for a subset of our LAEs, that ξion—the number of Lyman continuum photons per UV luminosity—is indeed 0.2–0.5 dex larger than for typical LBGs at similar redshifts. Using photoionization models, we estimate the effect this would have on both [O iii]/[O ii] and R23 and conclude such a hard spectrum can only partially explain such intense line emission. The additional possibility is that such a large [O iii]/[O ii] ratio is in part due to density rather than ionization bound nebular regions, which would imply a high escape fraction of ionizing photons. We discuss how further observations could confirm this possibility. Clearly LAEs with intense [O iii] emission represent a promising analog of those z > 7 sources with similarly strong lines that are thought to be an important contributor to cosmic reionization

Modification of a loop sequence between α-helices 6 and 7 of virus capsid (CA) protein in a human immunodeficiency virus type 1 (HIV-1) derivative that has simian immunodeficiency virus (SIVmac239) vif and CA α-helices 4 and 5 loop improves replication in cynomolgus monkey cells
<p>Abstract</p> <p>Background</p> <p>Human immunodeficiency virus type 1 (HIV-1) productively infects only humans and chimpanzees but not cynomolgus or rhesus monkeys while simian immunodeficiency virus isolated from macaque (SIVmac) readily establishes infection in those monkeys. Several HIV-1 and SIVmac chimeric viruses have been constructed in order to develop an animal model for HIV-1 infection. Construction of an HIV-1 derivative which contains sequences of a SIVmac239 loop between α-helices 4 and 5 (L4/5) of capsid protein (CA) and the entire SIVmac239 <it>vif </it>gene was previously reported. Although this chimeric virus could grow in cynomolgus monkey cells, it did so much more slowly than did SIVmac. It was also reported that intrinsic TRIM5α restricts the post-entry step of HIV-1 replication in rhesus and cynomolgus monkey cells, and we previously demonstrated that a single amino acid in a loop between α-helices 6 and 7 (L6/7) of HIV type 2 (HIV-2) CA determines the susceptibility of HIV-2 to cynomolgus monkey TRIM5α.</p> <p>Results</p> <p>In the study presented here, we replaced L6/7 of HIV-1 CA in addition to L4/5 and <it>vif </it>with the corresponding segments of SIVmac. The resultant HIV-1 derivatives showed enhanced replication capability in established T cell lines as well as in CD8+ cell-depleted primary peripheral blood mononuclear cells from cynomolgus monkey. Compared with the wild type HIV-1 particles, the viral particles produced from a chimeric HIV-1 genome with those two SIVmac loops were less able to saturate the intrinsic restriction in rhesus monkey cells.</p> <p>Conclusion</p> <p>We have succeeded in making the replication of simian-tropic HIV-1 in cynomolgus monkey cells more efficient by introducing into HIV-1 the L6/7 CA loop from SIVmac. It would be of interest to determine whether HIV-1 derivatives with SIVmac CA L4/5 and L6/7 can establish infection of cynomolgus monkeys <it>in vivo</it>.</p
Ferromagnetism in a Hubbard model for an atomic quantum wire: a realization of flat-band magnetism from even-membered rings
We have examined a Hubbard model on a chain of squares, which was proposed by
Yajima et al as a model of an atomic quantum wire As/Si(100), to show that the
flat-band ferromagnetism according to a kind of Mielke-Tasaki mechanism should
be realized for an appropriate band filling in such a non-frustrated lattice.
Reflecting the fact that the flat band is not a bottom one, the ferromagnetism
vanishes, rather than intensified, as the Hubbard U is increased. The exact
diagonalization method is used to show that the critical value of U is in a
realistic range. We also discussed the robustness of the magnetism against the

Effect of Dust Extinction on Estimating Star Formation Rate of Galaxies: Lyman Continuum Extinction
We re-examine the effect of Lyman continuum ( \AA)
extinction (LCE) by dust in H {\sc ii} regions in detail and discuss how it
affects the estimation of the global star formation rate (SFR) of galaxies. To
clarify the first issue, we establish two independent methods for estimating a
parameter of LCE (), which is defined as the fraction of Lyman continuum
photons contributing to hydrogen ionization in an H {\sc ii} region. One of
those methods determines from the set of Lyman continuum flux, electron
density and metallicity. In the framework of this method, as the metallicity
and/or the Lyman photon flux increase, is found to decrease. The other
method determines from the ratio of infrared flux to Lyman continuum flux.
Importantly, we show that f \la 0.5 via both methods in many H {\sc ii}
regions of the Galaxy. Thus, it establishes that dust in such H {\sc ii}
regions absorbs significant amount of Lyman continuum photons directly. To
examine the second issue, we approximate to a function of only the
dust-to-gas mass ratio (i.e., metallicity), assuming a parameter fit for the
Galactic H {\sc ii} regions. We find that a characteristic , which is
defined as averaged over a galaxy-wide scale, is 0.3 for the nearby spiral
galaxies. This relatively small indicates that a typical increment
factor due to LCE for estimating the global SFR () is large () for the nearby spiral galaxies. Therefore, we conclude that the effect of
LCE is not negligible relative to other uncertainties of estimating the SFR of
