    Equine strangles: An update on disease control and prevention

    Streptococcus equi spp. equi (SEE) causes a disease in horses commonly referred to as strangles. Carrier or reservoir equids are important for the maintenance of the bacteria between epizootics and the initiation of outbreaks on premises, they also make the control and prevention of the disease more difficult. Disease outbreaks are common in many countries, affecting negatively equine health and causing major economic losses to the equine industry. This review describes general aspects of the disease caused by SEE in horses (clinical signs, pathogenesis, epidemiology, treatment, complications) and then focuses on prevention, control and eradication mechanisms

    Working equids presented at a veterinary reference center in southern Chile (2015-2021)

    In Chile many families still rely on working equids as a fundamental source of income. Their use is particularly important for harnessing and transport of people and goods, in agriculture as well as in the leisure industry. Information on common emergency and clinical conditions in working equids is important to identify foci for owner education and to guide action towards disease prevention and animal welfare. The aim of the report was to highlight details about the clinical presentation of working equids at a veterinary reference center in southern Chile.Hospital records from working equids presented pro bono from 2015 to 2021 at the Equine Clinic of the Veterinary Teaching Hospital of the Universidad Austral de Chile were evaluated and included.A total of 107 working equid cases were recorded. The age range went from 1 day of age to 19 years (8.2±5.19 years). Most cases were not hospitalized and discharged on the same day of presentation (n=52). Horses were presented for evaluation of reproductive status (23%), lameness (15%) and castration (13%). Common reproductive diagnoses were uncomplicated castration (13%), barren (9%) and pregnant (8%) mares. Causes of lameness included laceration and wounds (40%), distal limb fractures (15%) and foot abscesses and/or poor foot care (10%). Only 4 cases of colic (3.7%) were presented over the years. Transrectal ultrasonography was the most used imaging tool, followed by radiography and endoscopy. Laboratory analysis was only performed in 15% of all cases.This is the first report on clinical presentation of working equids at a veterinary care reference center in southern Chile. Most working equids seek veterinary care to aid reproduction and treat conditions related to their use. Findings suggest that animal use, husbandry and welfare can be improved

    A partir de biomasa de algas en forma de pasta y procedente de su centrifugación (Nanochloropsis gaditana, Ascophyllum Nodosum y Laminaria Digitata) se elabora por procedimiento de solvólisis un biodiesel susceptible de hidrogenación en bioquioqueroseno y para su posterior uso como combustible de aviones. Este trabajo investiga la composición de las algas en ácido algínico y en sus unidades constituyentes o ácidos manurónico y gulurónico, y el contenido en alginato de sodio sobre algas secas. La valorización de residuos procedentes de centrifugados de algas con bajo contenido en aceite, se ha mostrado útil para la extracción de los mismos, y posterior solvolisis ácida y extracción con disolventes (acetilacetona y el etilenglicol). Especialmente la espectroscopía 1 HNMR y 13 CNMR se ha mostrado útil para identificar en los aceites, ácidos grasos próximos al ácido cis-cis-9,12- octadecadienoico.     Comparison of the performance of synthetic maize varieties created based on either genetic distance or general combining ability of the parents

    Synthetics varieties are grown by farmers and used by breeders to select new inbred lines. In countries unable to market hybrids, use of synthetics leads to yield improvements over landraces. Synthetics are derived from intercrossing inbred lines known to possess high general combining ability (GCA) as measured via crossing with testers and phenotyping for yield in multiple environments. Genetic similarity (GS) between lines measured by molecular markers may efficiently estimate GCA. Although the prediction of specific combining ability (SCA) of lines via GS has not been successful, it may have potential to predict the suitability of lines to form a synthetic variety. As this has not been reported, the objective of this research was to compare the performance of four synthetic maize varieties developed using GS calculated between parents using SSR markers with the performance of synthetics developed using GCA based on yield. Synthetics were phenotyped for yield and other agronomic traits in replicated field trials in several environments. The two synthetics formed based on low GS (0.34 and 0.33) performed better than all other synthetics in yield and most agronomic traits. The synthetics formed based on high GS (0.77 and 0.53), performed worst for nearly all traits. The GCA-based synthetics were generally intermediate for all traits.     Isolation and characterization of endosymbiotic bacteria from copper contaminated soils in Chile

    Legume endosymbiotic bacteria indigenous of copper (Cu)-contaminated soils from Chile have been isolated using pea (Pisum sativum), bean (Phaseolus vulgaris) and alfalfa (Medicago sativa) as trap host plants. Highly contaminated soils only produced nodules in certain legume hosts, whereas nodulation was observed in the three legume hosts when inoculated with soils containing a low Cu concentration. A collection of 362 strains was isolated, and their levels of Cu resistance were tested in media supplemented with increasing metal concentrations and in disk diffusion assays. By these two approaches, 84 strains displaying levels of Cu resistance higher than those exhibited by the corresponding reference strains were selected. The most resistant strains isolated from alfalfa and bean nodules grew normally at 3 mM and 2.5 mM CuSO4 and were obtained from two different highly contaminated soils. Strains nodulating pea plants showed similar levels of resistance to Cu (2-2.5 mM CuSO4) and were isolated from low-contaminated soils.     Photo-reverberation mapping of a protoplanetary accretion disk around a T Tauri star

    Theoretical models and spectroscopic observations of newborn stars suggest that protoplantary disks have an inner 'wall' at a distance set by the disk interaction with the star. Around T Tauri stars, the size of this disk hole is expected to be on a 0.1 au scale that is unresolved by current adaptive optics imaging, though some model-dependent constraints have been obtained by near-infrared interferometry. Here we report the first measurement of the inner disk wall around a solar-mass young stellar object, YLW 16B in the ρ Ophiuchi star-forming region, by detecting the light-travel time of the variable radiation from the stellar surface to the disk. Consistent time lags were detected on two nights, when the time series in H (1.6 μm) and K (2.2 μm) bands were synchronized while the 4.5 μm emission lagged by 74.5 ± 3.2 s. Considering the nearly edge-on geometry of the disk, the inner rim should be 0.084 au from the protostar on average, with an error of order 0.01 au. This size is likely larger than the range of magnetospheric truncations and consistent with an optically and geometrically thick disk front at the dust sublimation radius at ∼1500 K. The widths of the cross-correlation functions between the data in different wavebands place possible new constraints on the geometry of the disk

    Designing and Engineering Methylorubrum extorquens AM1 for Itaconic Acid Production

    Methylorubrum extorquens (formerly Methylobacterium extorquens) AM1 is a methylotrophic bacterium with a versatile lifestyle. Various carbon sources including acetate, succinate and methanol are utilized by M. extorquens AM1 with the latter being a promising inexpensive substrate for use in the biotechnology industry. Itaconic acid (ITA) is a high-value building block widely used in various industries. Given that no wildtype methylotrophic bacteria are able to utilize methanol to produce ITA, we tested the potential of M. extorquens AM1 as an engineered host for this purpose. In this study, we successfully engineered M. extorquens AM1 to express a heterologous codon-optimized gene encoding cis-aconitic acid decarboxylase. The engineered strain produced ITA using acetate, succinate and methanol as the carbon feedstock. The highest ITA titer in batch culture with methanol as the carbon source was 31.6 ± 5.5 mg/L, while the titer and productivity were 5.4 ± 0.2 mg/L and 0.056 ± 0.002 mg/L/h, respectively, in a scaled-up fed-batch bioreactor under 60% dissolved oxygen saturation. We attempted to enhance the carbon flux toward ITA production by impeding poly-β-hydroxybutyrate accumulation, which is used as carbon and energy storage, via mutation of the regulator gene phaR. Unexpectedly, ITA production by the phaR mutant strain was not higher even though poly-β-hydroxybutyrate concentration was lower. Genome-wide transcriptomic analysis revealed that phaR mutation in the ITA-producing strain led to complex rewiring of gene transcription, which might result in a reduced carbon flux toward ITA production. Besides poly-β-hydroxybutyrate metabolism, we found evidence that PhaR might regulate the transcription of many other genes including those encoding other regulatory proteins, methanol dehydrogenases, formate dehydrogenases, malate:quinone oxidoreductase, and those synthesizing pyrroloquinoline quinone and thiamine co-factors. Overall, M. extorquens AM1 was successfully engineered to produce ITA using acetate, succinate and methanol as feedstock, further supporting this bacterium as a feasible host for use in the biotechnology industry. This study showed that PhaR could have a broader regulatory role than previously anticipated, and increased our knowledge of this regulator and its influence on the physiology of M. extorquens AM1

    PA-6 inhibits inward rectifier currents carried by V93I and D172N gain-of-function KIR2.1 channels, but increases channel protein expression

    Background: The inward rectifier potassium current IK1 contributes to a stable resting membrane potential and phase 3 repolarization of the cardiac action potential. KCNJ2 gain-of function mutations V93I and D172N associate with increased IK1, short QT syndrome type 3 and congenital atrial fibrillation. Pentamidine-Analogue 6 (PA-6) is an efficient (IC50 = 14 nM with inside-out patch clamp methodology) and specific IK1 inhibitor that interacts with the cytoplasmic pore region of the KIR2.1 ion channel, encoded by KCNJ2. At 10 μM, PA-6 increases wild-type (WT) KIR2. 1 expression in HEK293T cells upon chronic treatment. We hypothesized that PA-6 will interact with and inhibit V93I and D172N KIR2.1 channels, whereas impact on channel expression at the plasma membrane requires higher concentrations. Methods: Molecular modelling was performed with the human KIR2.1 closed state homology model using FlexX. WT and mutant KIR2.1 channels were expressed in HEK293 cells. Patch clamp single cell electrophysiology measurements were performed in the whole cell and inside-out mode of the patch clamp method. KIR2.1 expression level and localization were determined by western blot analysis and immunofluorescence microscopy, respectively. Results: PA-6 docking in the V93I/D172N double mutant homology model of KIR2.1 demonstrated that mutations and drug-binding site are >30 Å apart. PA-6 inhibited WT and V93I outward currents with similar potency (IC50 = 35.5 and 43.6 nM at +50 mV for WT and V93I), whereas D172N currents were less sensitive (IC50 = 128.9 nM at +50 mV) using inside-out patch-clamp electrophysiology. In whole cell mode, 1 μM of PA-6 inhibited outward IK1 at −50 mV by 28 ± 36%, 18 ± 20% and 10 ± 6%, for WT, V93I and D172N channels respectively. Western blot analysis demonstrated that PA-6 (5 μM, 24 h) increased KIR2.1 expression levels of WT (6.3 ± 1.5 fold), and V93I (3.9 ± 0.9) and D172N (4.8 ± 2.0) mutants. Immunofluorescent microscopy demonstrated dose-dependent intracellular KIR2.1 accumulation following chronic PA-6 application (24 h, 1 and 5 μM). Conclusions: 1) KCNJ2 gain-of-function mutations V93I and D172N in the KIR2.1 ion channel do not impair PA-6 mediated inhibition of IK1, 2) PA-6 elevates KIR2.1 protein expression and induces intracellular KIR2.1 accumulation, 3) PA-6 is a strong candidate for further preclinical evaluation in treatment of congenital SQT3 and AF

    Magnetobiochronology of lower Pliocene marine sediments from the lower Guadalquivir Basin: insights into the tectonic evolution of the Strait of Gibraltar area

    The Gibraltar Arc is a complex tectonic region, and several competing models have been proposed to explain its evolution. We studied the sedimentary fill of the Guadalquivir Basin to identify tectonic processes that were occurring when the reopening of the Strait of Gibraltar led to the reestablishment of Mediterranean outflow. We present a chronostratigraphic framework for the Lower Pliocene sediments from the lower Guadalquivir Basin (SW Spain). The updated chronology is based on magnetobiostratigraphic data from several boreholes. Our results show that the studied interval in the La Matilla core is in the early Pliocene section, providing better constraints on the sedimentary evolution of the basin during that period. Migrating depositional facies led to a younger onset of sandy deposition basinward. At the northwestern passive margin, a 0.7 m.y. period of sedimentary bypass related to a sharp decrease in sedimentation rates and lower sea levels resulted from the tectonic uplift of the forebulge. In contrast, high sedimentation rates with continuous deep-marine sedimentation are recorded at the basin center due to continuous tectonic subsidence and west-southwestward progradation of axial depositional systems. The marginal forebulge uplift, continuous tectonic basinal subsidence, and southward progradation of clinoforms in the early Pliocene can be explained by the pull of a lithospheric slab beneath the Gibraltar Arc as the Strait of Gibraltar opened. These findings are, to our knowledge, the first reported sedimentary expression of slab pull beneath the Betics related to the opening of the Strait of Gibraltar after the Messinian salinity crisis
