    The Effects of Invasive Earthworm Species on Salamanders in the Grand Valley State University Ravine Ecosystem

    Earthworms are an invasive species that are causing ecological damage to northern forest ecosystems. The disruption to soil nutrient cycling and litter decomposition can negatively impact organisms that live within the leaf litter, such as salamanders. To test this hypothesis, we sampled earthworms within three ravines at thirty-six sites using the mustard extraction method. We surveyed salamander populations on two dates in 2015 at each site using cover boards. We also collected data on slope aspect, altitude, soil moisture, leaf litter coverage, canopy cover, and coarse woody debris at each site to determine their effects on earthworm and salamander populations. Unexpectedly, our results show that total earthworm populations did not decrease salamander abundance. However, epigeic earthworms in north facing, low elevation sites did have a negative effect on salamanders. We also found that anecic earthworm species had a negative impact on leaf litter in south facing, low elevation sites. Using a GLIMMIX model, we found that epigeic earthworms had a negative effect on salamander populations, while anecic earthworms had a positive effect on salamander populations

    Ce(OH)2Cl and lanthanide-substituted variants as precursors to redox-active CeO2 materials

    The cerium(III) hydroxide chloride Ce(OH)2Cl crystallises directly as a polycrystalline powder from a solution of CeCl3·7H2O in poly(ethylene) glycol (Mn = 400) heated at 240 °C and is found to be isostructural with La(OH)2Cl, as determined from high-resolution synchrotron powder X-ray diffraction (P21/m, a = 6.2868(2) Å, b = 3.94950(3) Å, c = 6.8740(3) Å, β = 113.5120(5)°). Replacement of a proportion of the cerium chloride in synthesis by a second lanthanide chloride yields a set of materials Ce1−xLnx(OH)2Cl for Ln = La, Pr, Gd, Tb. For La the maximum value of x is 0.2, with an isotropic expansion of the unit cell, but for the other lanthanides a wider composition range is possible, and the lattice parameters show an isotropic contraction with increasing x. Thermal decomposition of the hydroxide chlorides at 700 °C yields mixed-oxides Ce1−xLnxO2−δ that all have cubic fluorite structures with either expanded (Ln = La, Gd) or contracted (Ln = Pr, Tb) unit cells compared to CeO2. Scanning electron microscopy shows a shape memory effect in crystal morphology upon decomposition, with clusters of anisotropic sub-micron crystallites being seen in the precursor and oxide products. The Pr- and Tb-substituted oxides contain the substituent in a mixture of +3 and +4 oxidation states, as seen by X-ray absorption near edge structure spectroscopy at the lanthanide LIII edges. The mixed oxide materials are examined using temperature programmed reduction in 10%H2 in N2, which reveals redox properties suitable for heterogeneous catalysis, with the Pr-substituted materials showing the greatest reducibility at lower temperature

    BACKGROUND & AIMS: Plasma concentrations of bilirubin, a product of heme catabolism formed by biliverdin reductase A (BVRA), inversely associate with the risk of metabolic diseases including hepatic steatosis and diabetes mellitus in humans. Bilirubin has antioxidant and anti-inflammatory activities and may also regulate insulin signaling and peroxisome proliferator-activated receptor alpha (PPARα) activity. However, a causal link between bilirubin and metabolic diseases remains to be established. Here, we used the global Bvra gene knockout (Bvra-/-) mouse as a model of deficiency in bilirubin to assess its role in metabolic diseases. APPROACH & RESULTS: We fed mice fat-rich diets to induce hepatic steatosis and insulin resistance. Bile pigments were measured by LC-MS/MS, and hepatic lipids by LC-MS/MS (non-targeted lipidomics), HPLC-UV and Oil-Red-O staining. Oxidative stress was evaluated measuring F2-isoprostanes by GC-MS. Glucose metabolism and insulin sensitivity were verified by glucose and insulin tolerance tests, ex vivo and in vivo glucose uptake, and Western blotting for insulin signaling. Compared with wild type littermates, Bvra-/- mice contained negligible bilirubin in plasma and liver, and they had comparable glucose metabolism and insulin sensitivity. However, Bvra-/- mice exhibited an inflamed and fatty liver phenotype, accompanied by hepatic accumulation of oxidized triacylglycerols and F2-isoprostanes, in association with depletion of α-tocopherol. α-Tocopherol supplementation reversed the hepatic phenotype and observed biochemical changes in Bvra-/- mice. CONCLUSIONS: Our data suggests that BVRA deficiency renders mice susceptible to oxidative stress-induced hepatic steatosis in the absence of insulin resistance

    Heat Shock Protein-90 Inhibitors Enhance Antigen Expression on Melanomas and Increase T Cell Recognition of Tumor Cells

    In an effort to enhance antigen-specific T cell recognition of cancer cells, we have examined numerous modulators of antigen-expression. In this report we demonstrate that twelve different Hsp90 inhibitors (iHsp90) share the ability to increase the expression of differentiation antigens and MHC Class I antigens. These iHsp90 are active in several molecular and cellular assays on a series of tumor cell lines, including eleven human melanomas, a murine B16 melanoma, and two human glioma-derived cell lines. Intra-cytoplasmic antibody staining showed that all of the tested iHsp90 increased expression of the melanocyte differentiation antigens Melan-A/MART-1, gp100, and TRP-2, as well as MHC Class I. The gliomas showed enhanced gp100 and MHC staining. Quantitative analysis of mRNA levels showed a parallel increase in message transcription, and a reporter assay shows induction of promoter activity for Melan-A/MART-1 gene. In addition, iHsp90 increased recognition of tumor cells by T cells specific for Melan-A/MART-1. In contrast to direct Hsp90 client proteins, the increased levels of full-length differentiation antigens that result from iHsp90 treatment are most likely the result of transcriptional activation of their encoding genes. In combination, these results suggest that iHsp90 improve recognition of tumor cells by T cells specific for a melanoma-associated antigen as a result of increasing the expressed intracellular antigen pool available for processing and presentation by MHC Class I, along with increased levels of MHC Class I itself. As these Hsp90 inhibitors do not interfere with T cell function, they could have potential for use in immunotherapy of cancer

    Pharmacologically blocking p53-dependent apoptosis protects intestinal stem cells and mice from radiation.

    Exposure to high levels of ionizing radiation (IR) leads to debilitating and dose-limiting gastrointestinal (GI) toxicity. Using three-dimensional mouse crypt culture, we demonstrated that p53 target PUMA mediates radiation-induced apoptosis via a cell-intrinsic mechanism, and identified the GSK-3 inhibitor CHIR99021 as a potent radioprotector. CHIR99021 treatment improved Lgr5+ cell survival and crypt regeneration after radiation in culture and mice. CHIR99021 treatment specifically blocked apoptosis and PUMA induction and K120 acetylation of p53 mediated by acetyl-transferase Tip60, while it had no effect on p53 stabilization, phosphorylation or p21 induction. CHIR99021 also protected human intestinal cultures from radiation by PUMA but not p21 suppression. These results demonstrate that p53 posttranslational modifications play a key role in the pathological and apoptotic response of the intestinal stem cells to radiation and can be targeted pharmacologically
