    CCR2 and coronary artery disease: a woscops substudy

    Background Several lines of evidence support a role for CCL2 (monocyte chemotactic protein-1) and its receptor CCR2 in the development of atherosclerosis. The aim of the present study was to determine the association of the CCR2 Val64Ile polymorphism with the development of coronary artery disease in the WOSCOPS study sample set. Findings A total of 443 cases and 1003 controls from the West of Scotland Coronary Prevention Study (WOSCOPS) were genotyped for the Val64Ile polymorphism in the CCR2 gene. Genotype frequencies were compared between cases and controls. The CCR2 Val64Ile polymorphism was found not to be associated with coronary events in this study population (odds ratio 1.15, 95% CI 0.82-1.61, p = 0.41). Conclusions This case-control study does not support an association of the CCR2 Val64Ile polymorphism with coronary artery disease in the WOSCOPS sample set and does not confirm a possible protective role for CCR2 Val64Ile in the development of coronary artery disease

    Personalising laboratory medicine in the ‘real world’:assessing clinical utility, by clinical indication, of serum total B12 and Active-B12® (holotranscobalamin) in the diagnosis of vitamin B12 deficiency

    BACKGROUND: Assessing the pre- and post-test probability of disease in the context of routine health care is challenging. We wished to study how test performance parameters relating to clinical utility vary by clinical indication in a ‘real-world’ setting. METHODS: The diagnostic accuracy of serum total B(12) and Active-B(12)® (holotranscobalamin) was evaluated in a primary care population, using serum methylmalonic acid as the reference standard. We used electronic requesting to establish the clinical indication for each request. Routine requests from primary care for serum total B(12) were included if creatinine was also measured and estimated glomerular filtration rate was at least 60 mL/min/1.73 m(2). RESULTS: Clinical indications included peripheral neuropathy (n = 168), anaemia (n = 168), cognitive decline (n = 125), suspected dietary deficiency (n = 76), other (n = 362). For peripheral neuropathy, the area under the receiver operator curve ± 95% confidence interval (AUC ± CI) was 0.63 (0.54–0.71) (P = 0.002) for total B(12) and 0.68 (0.60–0.77) (P < 0.0001) for Active-B(12)®. For anaemia, AUC ± CI was 0.56 (0.47–0.66) (P = 0.10) for total B(12) and 0.69 (0.59–0.78) (P < 0.0001) for Active-B(12)®. For cognitive decline, AUC ± CI was 0.54 (0.43–0.65) (P = 0.26) for total B(12) and 0.69 (0.58–0.80) (P = 0.0002) for Active-B(12)®. The pre–post-test change in probability of disease varied by clinical indication. CONCLUSION: Combining diagnostic accuracy studies and electronic testing in a ‘real-world’ setting allows clinical utility to be assessed by clinical indication. Wider application of this would permit more personalised laboratory medicine. In this study, diagnostic performance of total B(12) and Active-B(12)® varied across all indications. Active-B(12)® provided better discrimination, but this may have reflected the cut-offs used

    Comparison of Glucose Monitoring Methods during Steady-State Exercise in Women

    Data from Continuous Glucose Monitoring (CGM) systems may help improve overall daily glycemia; however, the accuracy of CGM during exercise remains questionable. The objective of this single group experimental study was to compare CGM-estimated values to venous plasma glucose (VPG) and capillary plasma glucose (CPG) during steady-state exercise. Twelve recreationally active females without diabetes (aged 21.8 ± 2.4 years), from Central Washington University completed the study. CGM is used by individuals with diabetes, however the purpose of this study was to first validate the use of this device during exercise for anyone. Data were collected between November 2009 and April 2010. Participants performed two identical 45-min steady-state cycling trials (~60% Pmax) on non-consecutive days. Glucose concentrations (CGM-estimated, VPG, and CPG values) were measured every 5 min. Two carbohydrate gel supplements along with 360 mL of water were consumed 15 min into exercise. A product-moment correlation was used to assess the relationship and a Bland-Altman analysis determined error between the three glucose measurement methods. It was found that the CGM system overestimated mean VPG (mean absolute difference 17.4 mg/dL (0.97 mmol/L)) and mean CPG (mean absolute difference 15.5 mg/dL (0.86 mmol/L)). Bland-Altman analysis displayed wide limits of agreement (95% confidence interval) of 44.3 mg/dL (2.46 mmol/L) (VPG compared with CGM) and 41.2 mg/dL (2.29 mmol/L) (CPG compared with CGM). Results from the current study support that data from CGM did not meet accuracy standards from the 15197 International Organization for Standardization (ISO)

    State-by-State Report on Permanent Public Access to Electronic Government Information

    The purpose of this study was to research what, if anything, state governments are doing to meet the enormous challenges of ensuring permanent public access to state electronic government information. A comprehensive survey was created and distributed to AALL authors in each of the fifty states, the District of Columbia and Puerto Rico. State authors completed the survey by December 2002 and, in addition, submitted a short executive summary based on their survey results. The survey reveals that only one state—Colorado—has enacted legislation that explicitly addresses permanent public access (effective August 15, 2003). No state, including Colorado, comprehensively addresses the challenges of permanent public access to and preservation of electronic government information. State records boards, state archives and state libraries are often aware of permanent public access issues and have often taken steps to preserve electronic information. They have sometimes taken steps to provide continuous public access or have developed guidelines for state agencies to provide such access. These efforts of state records boards, state archives and state libraries are often ineffective, however, because they lack a solid statutory foundation. Without comprehensive statutes supporting a system to coordinate and centralize permanent public access, state agencies thwart the positive efforts of state records boards, state archives and state libraries. The agencies fail to appreciate the need to ensure the full lifecycle of electronic government information, particularly Web-based publications and records. And any guidelines for permanent public access that target them do not solve the problem of agencies’ lack the expertise, personnel and funding. We envisioned that this project would be the first step in the advocacy process necessary to enact state laws that will prevent the loss of important state government information in electronic format. Toward this end, we sought to create a document that could be provided to legislators and other policymakers to educate them about the responsibility of state governments to ensure permanent public access to electronic information. An additional objective was to strengthen the GRC and WAO’s ties to AALL members at the local level, thereby forming a base of activists who could advocate for improved laws mandating permanent public access to state government information. Because AALL and other library organizations lack the manpower to tackle the problem of disappearing electronic government information in all states simultaneously, the Grant Team has identified key states to target for legislative activity

    Using Field Data to Assess Model Predictions of Surface and Ground Fuel Consumption by Wildfire in Coniferous Forests of California

    Inventories of greenhouse gas (GHG) emissions from wildfire provide essential information to the state of California, USA, and other governments that have enacted emission reductions. Wildfires can release a substantial amount of GHGs and other compounds to the atmosphere, so recent increases in fire activity may be increasing GHG emissions. Quantifying wildfire emissions however can be difficult due to inherent variability in fuel loads and consumption and a lack of field data of fuel consumption by wildfire. We compare a unique set of fuel data collected immediately before and after six wildfires in coniferous forests of California to fuel consumption predictions of the first-order fire effects model (FOFEM), based on two different available fuel characterizations. We found strong regional differences in the performance of different fuel characterizations, with FOFEM overestimating the fuel consumption to a greater extent in the Klamath Mountains than in the Sierra Nevada. Inaccurate fuel load inputs caused the largest differences between predicted and observed fuel consumption. Fuel classifications tended to overestimate duff load and underestimate litter load, leading to differences in predicted emissions for some pollutants. When considering total ground and surface fuels, modeled consumption was fairly accurate on average, although the range of error in estimates of plot level consumption was very large. These results highlight the importance of fuel load input to the accuracy of modeled fuel consumption and GHG emissions from wildfires in coniferous forests

    An exponential decline at the bright end of the z=6 galaxy luminosity function

    We present the results of a search for the most luminous star-forming galaxies at redshifts z~6 based on CFHT Legacy Survey data. We identify a sample of 40 Lyman break galaxies brighter than magnitude z'=25.3 across an area of almost 4 square degrees. Sensitive spectroscopic observations of seven galaxies provide redshifts for four, of which only two have moderate to strong Lyman alpha emission lines. All four have clear continuum breaks in their spectra. Approximately half of the Lyman break galaxies are spatially resolved in 0.7 arcsec seeing images, indicating larger sizes than lower luminosity galaxies discovered with the Hubble Space Telescope, possibly due to on-going mergers. The stacked optical and infrared photometry is consistent with a galaxy model with stellar mass ~ 10^{10} solar masses. There is strong evidence for substantial dust reddening with a best-fit A_V=0.7 and A_V>0.48 at 2 sigma confidence, in contrast to the typical dust-free galaxies of lower luminosity at this epoch. The spatial extent and spectral energy distribution suggest that the most luminous z~6 galaxies are undergoing merger-induced starbursts. The luminosity function of z=5.9 star-forming galaxies is derived. This agrees well with previous work and shows strong evidence for an exponential decline at the bright end, indicating that the feedback processes which govern the shape of the bright end are occurring effectively at this epoch.Comment: 14 pages, 11 figures, AJ in press, revised to address referee comment

    High-resolution transcriptional analysis of the regulatory influence of cell-to-cell signalling reveals novel genes that contribute to Xanthomonas phytopathogenesis

    The bacterium Xanthomonas campestris is an economically important pathogen of many crop species and a model for the study of bacterial phytopathogenesis. In X.campestris, a regulatory system mediated by the signal molecule DSF controls virulence to plants. The synthesis and recognition of the DSF signal depends upon different Rpf proteins. DSF signal generation requires RpfF whereas signal perception and transduction depends upon a system comprising the sensor RpfC and regulator RpfG. Here we have addressed the action and role of Rpf/DSF signalling in phytopathogenesis by high-resolution transcriptional analysis coupled to functional genomics. We detected transcripts for many genes that were unidentified by previous computational analysis of the genome sequence. Novel transcribed regions included intergenic transcripts predicted as coding or non-coding as well as those that were antisense to coding sequences. In total, mutation of rpfF, rpfG and rpfC led to alteration in transcript levels (more than fourfold) of approximately 480 genes. The regulatory influence of RpfF and RpfC demonstrated considerable overlap. Contrary to expectation, the regulatory influence of RpfC and RpfG had limited overlap, indicating complexities of the Rpf signalling system. Importantly, functional analysis revealed over 160 new virulence factors within the group of Rpf-regulated genes.</p
