    Characterization of the free energy dependence of an interprotein electron transfer reaction by variation of pH and site-directed mutagenesis

    AbstractThe interprotein electron transfer (ET) reactions of the cupredoxin amicyanin, which mediates ET from the tryptophan tryptophylquinone (TTQ) cofactor of methylamine dehydrogenase to cytochrome c-551i have been extensively studied. However, it was not possible to perform certain key experiments in that native system. This study examines the ET reaction from reduced amicyanin to an alternative electron acceptor, the diheme protein MauG. It was possible to vary the ΔG° for this ET reaction by simply changing pH to determine the dependence of kET on ΔG°. A P94A mutation of amicyanin significantly altered its oxidation–reduction midpoint potential value. It was not possible to study the ET from reduced P94A amicyanin to cytochrome c-551i in the native system because that reaction was kinetically coupled. However, the reaction from reduced P94A amicyanin to MauG was a true ET reaction and it was possible to determine values of reorganization energy (λ) and electronic coupling for the reactions of this variant as well as native amicyanin. Comparison of the λ values associated with the ET reactions between amicyanin and the TTQ of methylamine dehydrogenase, the diheme center of MauG and the single heme of cytochrome c-551i, provides insight into the factors that dictate the λ values for the respective reactions. These results demonstrate how study of ET reactions with alternative redox partner proteins can complement and enhance our understanding of the reactions with the natural redox partners, and further our understanding of mechanisms of protein ET reactions

    Taking Snapshots, Living the Picture: The Kodak Company's Making of Photographic Biography

    In this article I explore how George Eastman and the Eastman Kodak Company encouraged early twentieth-century camera users to think of snapshots as pictorial biographies. Analysing a wide selection of articles from the Kodakery, one of Kodak’s most popular magazines in the first half of the twentieth century, I demonstrate that the company endeavoured to secure its prominence in the photographic market by encouraging members of the public to integrate picture-taking into everyday life, and regard photographs as self-contained repositories of biographical details. To this end, Kodak framed the speedy pace of life that characterised the practice of being in the industrial world as a reality that allegedly weakened the human eye and mind’s ability to process the experience of life itself. Introducing the idea of the camera and picturetaking as the ultimate cures for this purported human deficiency, Kodak provided camera users with advice that helped to cement an understanding of photographs as surrogates of both the changing human body and individual subjects’ experiences in time and space. As in popular culture, and sometimes also in academia, photographs are still widely regarded as pictorial biographies, I argue that considering the popular photographic industry’s role in shaping photographic practices and photographs’ perceived meanings can help clarify the relationship between photography and life-writing

    Expanding the search for galaxies at z ~7-10 with new NICMOS Parallel Fields

    We have carried out a search for galaxies at z ~ 7-10 in ~14.4 sq. arcmin of new NICMOS parallel imaging taken in the Great Observatories Origins Deep Survey (GOODS, 5.9 sq. arcmin), the Cosmic Origins Survey (COSMOS, 7.2 sq. arcmin), and SSA22 (1.3 sq. arcmin). These images reach 5 sigma sensitivities of J110 = 26.0-27.5 (AB), and combined they increase the amount of deep near-infrared data by more than 60% in fields where the investment in deep optical data has already been made. We find no z>7 candidates in our survey area, consistent with the Bouwens et al. (2008) measurements at z~7 and 9 (over 23 sq. arcmin), which predict 0.7 galaxies at z~7 and <0.03 galaxies at z~9. We estimate that 10-20% of z>7 galaxies are missed by this survey, due to incompleteness from foreground contamination by faint sources. For the case of luminosity evolution, assuming a Schecter parameterization with a typical phi* = 10^-3 Mpc^-3, we find M* > -20.0 for z~7 and M* > -20.7 for z~9 (68% confidence). This suggests that the downward luminosity evolution of LBGs continues to z~7, although our result is marginally consistent with the z~6 LF of Bouwens et al.(2006, 2007). In addition we present newly-acquired deep MMT/Megacam imaging of the z~9 candidate JD2325+1433, first presented in Henry et al. (2008). The resulting weak but significant detection at i' indicates that this galaxy is most likely an interloper at z~2.7.Comment: Accepted to ApJ.     Hard Two-Photon Contribution to Elastic Lepton-Proton Scattering: Determined by the OLYMPUS Experiment

    The OLYMPUS collaboration reports on a precision measurement of the positron-proton to electron-proton elastic cross section ratio, R2γR_{2\gamma}, a direct measure of the contribution of hard two-photon exchange to the elastic cross section. In the OLYMPUS measurement, 2.01~GeV electron and positron beams were directed through a hydrogen gas target internal to the DORIS storage ring at DESY. A toroidal magnetic spectrometer instrumented with drift chambers and time-of-flight scintillators detected elastically scattered leptons in coincidence with recoiling protons over a scattering angle range of 20°\approx 20\degree to 80°80\degree. The relative luminosity between the two beam species was monitored using tracking telescopes of interleaved GEM and MWPC detectors at 12°12\degree, as well as symmetric M{\o}ller/Bhabha calorimeters at 1.29°1.29\degree. A total integrated luminosity of 4.5~fb1^{-1} was collected. In the extraction of R2γR_{2\gamma}, radiative effects were taken into account using a Monte Carlo generator to simulate the convolutions of internal bremsstrahlung with experiment-specific conditions such as detector acceptance and reconstruction efficiency. The resulting values of R2γR_{2\gamma}, presented here for a wide range of virtual photon polarization 0.456<ϵ<0.9780.456<\epsilon<0.978, are smaller than some hadronic two-photon exchange calculations predict, but are in reasonable agreement with a subtracted dispersion model and a phenomenological fit to the form factor data.Comment: 5 pages, 3 figures, 2 table

    Infection with feline immunodeficiency virus (FIV) leads to the development of a disease state similar to AIDS in man. Recent studies have identified the chemokine receptor CXCR4 as the major receptor for cell culture-adapted strains of FIV, suggesting that FIV and human immunodeficiency virus (HIV) share a common mechanism of infection involving an interaction between the virus and a member of the seven transmembrane domain superfamily of molecules. This article reviews the evidence for the involvement of chemokine receptors in FIV infection and contrasts these findings with similar studies on the primate lentiviruses HIV and SIV (simian immunodeficiency virus)

    Radical stereotactic radiosurgery with real-time tumor motion tracking in the treatment of small peripheral lung tumors

    <p>Abstract</p> <p>Background</p> <p>Recent developments in radiotherapeutic technology have resulted in a new approach to treating patients with localized lung cancer. We report preliminary clinical outcomes using stereotactic radiosurgery with real-time tumor motion tracking to treat small peripheral lung tumors.</p> <p>Methods</p> <p>Eligible patients were treated over a 24-month period and followed for a minimum of 6 months. Fiducials (3–5) were placed in or near tumors under CT-guidance. Non-isocentric treatment plans with 5-mm margins were generated. Patients received 45–60 Gy in 3 equal fractions delivered in less than 2 weeks. CT imaging and routine pulmonary function tests were completed at 3, 6, 12, 18, 24 and 30 months.</p> <p>Results</p> <p>Twenty-four consecutive patients were treated, 15 with stage I lung cancer and 9 with single lung metastases. Pneumothorax was a complication of fiducial placement in 7 patients, requiring tube thoracostomy in 4. All patients completed radiation treatment with minimal discomfort, few acute side effects and no procedure-related mortalities. Following treatment transient chest wall discomfort, typically lasting several weeks, developed in 7 of 11 patients with lesions within 5 mm of the pleura. Grade III pneumonitis was seen in 2 patients, one with prior conventional thoracic irradiation and the other treated with concurrent Gefitinib. A small statistically significant decline in the mean % predicted DLCO was observed at 6 and 12 months. All tumors responded to treatment at 3 months and local failure was seen in only 2 single metastases. There have been no regional lymph node recurrences. At a median follow-up of 12 months, the crude survival rate is 83%, with 3 deaths due to co-morbidities and 1 secondary to metastatic disease.</p> <p>Conclusion</p> <p>Radical stereotactic radiosurgery with real-time tumor motion tracking is a promising well-tolerated treatment option for small peripheral lung tumors.</p

    Keeping the Board in the Dark: CEO Compensation and Entrenchment

    We study a model in which a CEO can entrench himself by hiding information from the board that would allow the board to conclude that he should be replaced. Assuming that even diligent monitoring by the board cannot fully overcome the information asymmetry visà- vis the CEO, we ask if there is a role for CEO compensation to mitigate the inefficiency. Our analysis points to a novel argument for high-powered, non-linear CEO compensation such as bonus pay or stock options. By shifting the CEO’s compensation into states where the firm’s value is highest, a high-powered compensation scheme makes it as unattractive as possible for the CEO to entrench himself when he expects that the firm’s future value under his management and strategy is low. This, in turn, minimizes the severance pay needed to induce the CEO not to entrench himself, thereby minimizing the CEO’s informational rents. Amongst other things, our model suggests how deregulation and technological changes in the 1980s and 1990s might have contributed to the rise in CEO pay and turnover over the same period

    FIV establishes a latent infection in feline peripheral blood CD4+ T lymphocytes in vivo during the asymptomatic phase of infection

    <p>Abstract</p> <p>Background</p> <p>Feline immunodeficiency virus (FIV) is a lentivirus of cats that establishes a lifelong persistent infection with immunologic impairment.</p> <p>Results</p> <p>In an approximately 2 year-long experimental infection study, cats infected with a biological isolate of FIV clade C demonstrated undetectable plasma viral loads from 10 months post-infection onward. Viral DNA was detected in CD4+CD25+ and CD4+CD25- T cells isolated from infected cats whereas viral RNA was not detected at multiple time points during the early chronic phase of infection. Viral transcription could be reactivated in latently infected CD4+ T cells <it>ex vivo </it>as demonstrated by detectable FIV <it>gag </it>RNA and 2-long terminal repeat (LTR) circle junctions. Viral LTR and <it>gag </it>sequences amplified from peripheral blood mononuclear cells during early and chronic stages of infection demonstrated minimal to no viral sequence variation.</p> <p>Conclusions</p> <p>Collectively, these findings are consistent with FIV latency in peripheral blood CD4+ T cells isolated from chronically infected cats. The ability to isolate latently FIV-infected CD4+ T lymphocytes from FIV-infected cats provides a platform for the study of <it>in vivo </it>mechanisms of lentiviral latency.</p

    Effects of antiplatelet therapy on stroke risk by brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases: subgroup analyses of the RESTART randomised, open-label trial

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage.
