    Missing Phases of Deliberation Dialogue for Real Applications

    Models of deliberative dialogue are fundamental for developing autonomous systems that support human practical reasoning. However, we must consider whether these existing models are able to capture the complexity and richness of natural deliberation for developing real applications. In real contexts, circumstances relevant to the decision can change rapidly. In this paper, we introduce an extension to today\u27s leading model of deliberation dialogue to capture dynamic changes of circumstances during dialogue. Moreover, in natural deliberation, a dialogue may be successful even if a decision on what to do has not been made. A set of criteria is proposed to address the problem of when to close o the practical reasoning phase of dialogue. We then discuss some initial efforts to introduce those characteristics within an existing model of deliberation for agent collaboration.     Inactivation of cloned Na channels expressed in Xenopus oocytes

    This study investigates the inactivation properties of Na channels expressed in Xenopus oocytes from two rat IIA Na channel cDNA clones differing by a single amino acid residue. Although the two cDNAs encode Na channels with substantially different activation properties (Auld, V. J., A. L. Goldin, D. S. Krafte, J. Marshall, J. M. Dunn, W. A. Catterall, H. A. Lester, N. Davidson, and R. J. Dunn. 1988. Neuron. 1:449-461), their inactivation properties resemble each other strongly but differ markedly from channels induced by poly(A+) rat brain RNA. Rat IIA currents inactivate more slowly, recover from inactivation more slowly, and display a steady-state voltage dependence that is shifted to more positive potentials. The macroscopic inactivation process for poly(A+) Na channels is defined by a single exponential time course; that for rat IIA channels displays two exponential components. At the single-channel level these differences in inactivation occur because rat IIA channels reopen several times during a depolarizing pulse; poly(A+) channels do not. Repetitive stimulation (greater than 1 Hz) produces a marked decrement in the rat IIA peak current and changes the waveform of the currents. When low molecular weight RNA is coinjected with rat IIA RNA, these inactivation properties are restored to those that characterize poly(A+) channels. Slow inactivation is similar for rat IIA and poly(A+) channels, however. The data suggest that activation and inactivation involve at least partially distinct regions of the channel protein

    We argue that burden of proof (BoP) of the kind present in persuasion does not apply to deliberation. We analyze existing computational models showing that in deliberation agents may answer a critique but there is no violation of the protocol if they choose not to. We propose a norm-­‐‑governed dialogue where BoP in persuasion is modeled as an obligation to respond, and permissions capture the different types of constraint observed in deliberation

We reflect on today's leading model of deliberation dialogue and we propose an extension to capture how newly exchanged information about changing circumstances may shape the dialogue.Moreover, in natural deliberation, a dialogue may be successful even if a decision on what to do has not been made. A set of criteria is proposed to address the problem of when to close off the practical reasoning phase of dialogue. We discuss some measures for evaluating the success of a dialogue after closure and we present some initial efforts to introduce the new deliberation features within an existing model of agent dialogue.     The initiator methionine tRNA drives cell migration and invasion leading to increased metastatic potential in melanoma

    The cell's repertoire of transfer RNAs (tRNAs) has been linked to cancer. Recently, levels of the initiator methionine tRNA (tRNAiMet) in stromal fibroblasts have been shown to influence extracellular matrix (ECM) secretion to drive tumour growth and angiogenesis. Here we show that increased tRNAiMet within cancer cells does not influence tumour growth, but drives cell migration and invasion via a mechanism that is independent from ECM synthesis and dependent on α5ÎČ1 integrin and levels of the translation initiation ternary complex. In vivo and ex vivo migration (but not proliferation) of melanoblasts is significantly enhanced in transgenic mice which express additional copies of the tRNAiMet gene. We show that increased tRNAiMet in melanoma drives migratory, invasive behaviour and metastatic potential without affecting cell proliferation and primary tumour growth, and that expression of RNA polymerase III-associated genes (which drive tRNA expression) are elevated in metastases by comparison with primary tumours. Thus specific alterations to the cancer cell tRNA repertoire drive a migration/invasion programme that may lead to metastasis

    Together, yet still not equal? Sex integration in equestrian sport

    Sex segregation is a core organising principle of most modern sports and is a key element in the marginalisation and subordination of girls and women in sport and beyond. In this article I explore the only Olympic-level sport which is not organised around sex segregation – equestrian sport – in order to consider the implications of sex integration for female participants. I draw on a study conducted on elite riders that found that although sex integration in equestrian sport does not lead to female participants being excluded from high-level competition, men continue to perform disproportionately well. This suggests that although sex integration may be an important step towards breaking down gender hierarchies in sport, without accompanying wider changes in gender norms and expectations, sex integration alone will not be enough to achieve greater gender equality in equestrian sport

    Radiogenomics map-based molecular and imaging phenotypical characterization in localised prostate cancer using pre-biopsy biparametric MR imaging

    Objective: To create a radiogenomics map and evaluate the correlation between molecular and imaging phenotypes in localized prostate cancer (PCa), using radical prostatectomy histopathology as a reference standard.Methods: Radiomic features were extracted from T2-weighted (T2WI) and Apparent Diffusion Coefficient (ADC) images of clinically localized PCa patients (n=15) across different Gleason scorebased risk categories. DNA extraction was performed on formalin-fixed, paraffin-embedded (FFPE) samples. Gene expression analysis of androgen receptor expression, apoptosis, and hypoxia was conducted using the Chromosome Analysis Suite (ChAS) application and OSCHIP files. The relationship between gene expression alterations and textural features was assessed using Pearson's correlation analysis. Receiver operating characteristic (ROC) analysis was utilized to evaluate the predictive accuracy of the model.Results: A significant correlation was observed between radiomic texture features and copy number variation (CNV) of genes associated with apoptosis, hypoxia, and androgen receptor (p-value= ≀ 0.05). The identified radiomic features, including Sum Entropy ADC, Inverse Difference ADC, Sum Variance T2WI, Entropy T2WI, Difference Variance T2WI, and Angular Secondary Moment T2WI, exhibited potential for predicting cancer grade and biological processes such as apoptosis and hypoxia. Incorporating radiomics and genomics into a prediction model significantly improved the prediction of prostate cancer grade (clinically significant prostate cancer), yielding an AUC of 0.95.Conclusion: Radiomic texture features significantly correlate with genotypes for apoptosis, hypoxia, and androgen receptor expression in localised prostate cancer.
