    Wind tunnel measurements of surface pressure fluctuations at Mach numbers of 1.6, 2.0, and 2.5 using 12 different transducers

    The turbulent boundary layer on the wall of a 9 by 7 foot wind tunnel was measured with 12 different transducers at Mach numbers of 1.6, 2.0, and 2.5. The results indicated that the wall surface-pressure-fluctuation field was more homogeneous at a Mach number of 2.5 than at Mach numbers of 1.6 or 2.0. A comparison of power-spectral-density data at Mach 2.5 with a summary of similar data (Mach 0.1 to 3.45) showed good agreement. The measurement uncertainty was greatest when frequencies were low and the surface-pressure-fluctuation field was homogeneous. The uncertainty at higher frequencies increased as the surface-pressure-fluctuation field became more inhomogeneous. Since transducer mounting effects and system noise levels were determined not to have contributed appreciably to measurement uncertainties, the result was attributed to an interaction between the surface-pressure-fluctuation field and the transducers. Corcos' correction for size effects improved the comparison between transducers at the high frequencies, but did not eliminate an apparent size effect at the lower frequencies

    Effect of gaseous and solid simulated jet plumes on an 040A space shuttle launch configuration at m=1.6 to 2.2

    The effect of plume-induced flow separation and aspiration effects due to operation of both orbiter and the solid rocket motors on a 0.019-scale model of the launch configuration of the Space Shuttle Vehicle is determined. Longitudinal and lateral-directional stability data were obtained at Mach numbers of 1.6, 2.0, and 2.2 with and without the engines operating. The plumes exiting from the engines were simulated by a cold-gas jet supplied by an auxiliary 200-atm air supply system and solid-body plume simulators. The aerodynamic effects produced by these two simulation procedures are compared. The parameters most significantly affected by the jet plumes are pitching moment, elevon control effectiveness, axial force, and orbiter wing loads. The solid rocket motor (SRM) plumes have the largest effect on the aerodynamic characteristics. The effect of the orbiter plumes in combination with the SRM plumes is also significant. Variations in the nozzle design parameters and configuration changes can reduce the jet plume-induced aerodynamic effects

    De novo design of protein logic gates

    The design of modular protein logic for regulating protein function at the posttranscriptional level is a challenge for synthetic biology. Here, we describe the design of two-input AND, OR, NAND, NOR, XNOR, and NOT gates built from de novo–designed proteins. These gates regulate the association of arbitrary protein units ranging from split enzymes to transcriptional machinery in vitro, in yeast and in primary human T cells, where they control the expression of the TIM3 gene related to T cell exhaustion. Designed binding interaction cooperativity, confirmed by native mass spectrometry, makes the gates largely insensitive to stoichiometric imbalances in the inputs, and the modularity of the approach enables ready extension to three-input OR, AND, and disjunctive normal form gates. The modularity and cooperativity of the control elements, coupled with the ability to de novo design an essentially unlimited number of protein components, should enable the design of sophisticated posttranslational control logic over a wide range of biological functions

    The peptide agonist-binding site of the glucagon-like peptide-1 (GLP-1) receptor based on site-directed mutagenesis and knowledge-based modelling

    Glucagon-like peptide-1 (7–36)amide (GLP-1) plays a central role in regulating blood sugar levels and its receptor, GLP-1R, is a target for anti-diabetic agents such as the peptide agonist drugs exenatide and liraglutide. In order to understand the molecular nature of the peptide–receptor interaction, we used site-directed mutagenesis and pharmacological profiling to highlight nine sites as being important for peptide agonist binding and/or activation. Using a knowledge-based approach, we constructed a 3D model of agonist-bound GLP-1R, basing the conformation of the N-terminal region on that of the receptor-bound NMR structure of the related peptide pituitary adenylate cyclase-activating protein (PACAP21). The relative position of the extracellular to the transmembrane (TM) domain, as well as the molecular details of the agonist-binding site itself, were found to be different from the model that was published alongside the crystal structure of the TM domain of the glucagon receptor, but were nevertheless more compatible with published mutagenesis data. Furthermore, the NMR-determined structure of a high-potency cyclic conformationally-constrained 11-residue analogue of GLP-1 was also docked into the receptor-binding site. Despite having a different main chain conformation to that seen in the PACAP21 structure, four conserved residues (equivalent to His-7, Glu-9, Ser-14 and Asp-15 in GLP-1) could be structurally aligned and made similar interactions with the receptor as their equivalents in the GLP-1-docked model, suggesting the basis of a pharmacophore for GLP-1R peptide agonists. In this way, the model not only explains current mutagenesis and molecular pharmacological data but also provides a basis for further experimental design

    Properties of Blood, Porphyrins, and Exposure to Legacy and Emerging Persistent Organic Pollutants in Surf Scoters (Melanitta perspicillata) Overwintering on the South Coast of British Columbia, Canada

    The surf scoter (Melanitta perspicillata) is a little-studied species of North American sea duck. Estimates suggest it has experienced a precipitous decline in breeding numbers over the latter half of the past century. To investigate the potential role of contaminant uptake and toxicity in the population decline, this study undertook to measure blood chemistry, porphyrin concentrations, EROD, and organic contaminants in mature surf scoters wintering in the Strait of Georgia, BC, Canada. Hepatic organochlorine pesticide, polychlorinated dibenzo-p-dioxin, polychlorinated dibenzofuran, polychlorinated biphenyl (PCB), polybrominated diphenyl ether, and nonylphenol concentrations were relatively low; for example, ΣTEQs (toxic equivalents) for PCBs, dioxins, and furans combined ranged from 4.7 ng/kg wet weight in reference-site (Baynes Sound) birds to 11.4 ng/kg wet weight in birds from Vancouver Harbour. Nonetheless, elevated EROD activity indicated that birds in Howe Sound were responding to an Ah-receptor-mediated stressor, which was also affecting hematocrit values and possibly vitamin A status. In addition, a low proportion of lymphocytes in individuals across locations in early spring samples was associated with poor body condition. The apparent loss of fitness just prior to the onset of northerly migrations to breeding grounds is of particular concern. Compromised health of mature birds at this point in the season might impact negatively on the productivity and survival of some individuals, particularly those overwintering in Howe Sound

    A Blockchain-Assisted Hash-Based Signature Scheme

    We present a server-supported, hash-based digital signature scheme. To achieve greater efficiency than current state of the art, we relax the security model somewhat. We postulate a set of design requirements, discuss some approaches and their practicality, and finally reach a forward-secure scheme with only modest trust assumptions, achieved by employing the concepts of authenticated data structures and blockchains. The concepts of blockchain authenticated data structures and the presented blockchain design could have independent value and are worth further research
