    Microglia in the aging brain: relevance to neurodegeneration

    Microglia cells are the brain counterpart of macrophages and function as the first defense in the brain. Although they are neuroprotective in the young brain, microglia cells may be primed to react abnormally to stimuli in the aged brain and to become neurotoxic and destructive during neurodegeneration. Aging-induced immune senescence occurs in the brain as age-associated microglia senescence, which renders microglia to function abnormally and may eventually promote neurodegeneration. Microglia senescence is manifested by both morphological changes and alterations in immunophenotypic expression and inflammatory profile. These changes are likely caused by microinvironmental factors, but intrinsic factors cannot yet be completely excluded. Microglia senescence appears to underlie the switching of microglia from neuroprotective in the young brain to neurotoxic in the aged brain. The hypothesis of microglia senescence during aging offers a novel perspective on their roles in aging-related neurodegeneration. In Parkinson's disease and Alzheimer's disease, over-activation of microglia may play an active role in the pathogenesis because microglia senescence primes them to be neurotoxic during the development of the diseases

    Pixel-wise Graph Attention Networks for Person Re-identification

    Graph convolutional networks (GCN) is widely used to handle irregular data since it updates node features by using the structure information of graph. With the help of iterated GCN, high-order information can be obtained to further enhance the representation of nodes. However, how to apply GCN to structured data (such as pictures) has not been deeply studied. In this paper, we explore the application of graph attention networks (GAT) in image feature extraction. First of all, we propose a novel graph generation algorithm to convert images into graphs through matrix transformation. It is one magnitude faster than the algorithm based on K Nearest Neighbors (KNN). Then, GAT is used on the generated graph to update the node features. Thus, a more robust representation is obtained. These two steps are combined into a module called pixel-wise graph attention module (PGA). Since the graph obtained by our graph generation algorithm can still be transformed into a picture after processing, PGA can be well combined with CNN. Based on these two modules, we consulted the ResNet and design a pixel-wise graph attention network (PGANet). The PGANet is applied to the task of person re-identification in the datasets Market1501, DukeMTMC-reID and Occluded-DukeMTMC (outperforms state-of-the-art by 0.8\%, 1.1\% and 11\% respectively, in mAP scores).     OBJECTIVES: to investigate the quality of life and the associated factors on left behind elderly in rural China. METHOD: the research was conducted cluster sampling to select 456 elderly left behind when family members migrated out of rural China to participate in a cross-sectional study by completing a general data questionnaire and Quality of Life questionnaire. RESULTS: 91.5% of the elderly requested psychological counseling and education. For the elderly, scores for mental health (39.56±13.73) were significantly lower compared with Chinese standard data (61.6±13.7) (POBJETIVOS: investigar la calidad de vida y los factores asociados a los adultos mayores que se quedan en las zonas rurales de China. MÉTODO: la investigación se realizó por medio de muestreo por conglomerados para seleccionar 456 adultos mayores que se quedaron cuando los miembros de la familia emigraron de zonas rurales de China, para participar en un estudio de corte transversal, completando un cuestionario de datos generales y cuestionario de calidad de vida. RESULTADOS: el 91.5% de los adultos mayores solicitó asistencia psicológica y educación. Para los adultos mayores, las puntuaciones de salud mental (39.56±13.73) fueron significativamente más bajos en comparación con los datos estándar de China (61.6±13.7) (pOBJETIVOS: investigar a qualidade de vida e fatores associados de idosos deixados para trás na China rural.     Circular RNA Expression Profile and Analysis of Their Potential Function in Psoriasis

    Background/Aims: Circular RNAs (circRNAs) are evolutionary conserved circular non-coding RNAs that play a role in several diseases by sequestering (sponging) microRNAs (miRNAs). However, their role in psoriasis remains unclear. In the present study, we investigated the expression of circRNAs and analyzed their potential functions in psoriasis. Methods: The SBC human ceRNA array V1.0 was used to analyze circRNA expression in psoriatic lesions and normal healthy skin tissues. Functional analyses were performed using Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Putative miRNA response elements (MREs) were identified using miRNA target prediction software. Six upregulated circRNAs were verified by quantitative real-time reverse transcription polymerase chain reaction in psoriatic lesions and healthy skin tissues. Results: A total of 4956 circRNAs (3016 upregulated and 1940 downregulated; fold change ≥2 and p< 0.05) were identified as differentially expressed in psoriasis. Furthermore, 4405 MREs were identified among the differentially expressed circRNAs. hsa_circ_0061012 was upregulated in psoriatic lesions compared with normal healthy skin tissues. The top five MREs of hsa_circ_0061012 were hsa-miR-7157-5p, hsa-miR-4769-3p, hsa-miR-6817-5p, hsa-miR-4310, and hsa-miR-6882-3p. GO analysis was carried out to investigate the biological functions enriched among the upregulated targets of five miRNAs in psoriasis. The GO analysis identified that most of top 30 of GO enrichment are related to psoriasis. Conclusion: hsa_circ_0061012 might be a candidate biomarker for psoriasis. The results provide a new perspective for a better understanding of ceRNA-mediated gene regulation in psoriasis, and provide a novel theoretical basis for further studies on the function of circRNA in psoriasis

    Effect of polysaccharide from the root of Bupleurum Chinese DC and Bupleurum scorzonerifolium Willd on hydrogen peroxide-induced myocardial apoptosis

    Purpose: To investigate the protective effect of polysaccharide (BRP) from the root of Bupleurum Chinese DC. and Bupleurum scorzonerifolium Willd. on cardiomyocyte cells. Methods: Response surface methodology (RSM) based on Box-Behnken Design (BBD) was performed to optimize the extraction conditions for BRP. The effect of BRP on cardiomyocyte cell apoptosis was evaluated in H9c2 cells treated with hydrogen peroxide (H2O2). Cell viability was determined by CCK-8 assay, while oxidative stress levels in H9c2 cells, including lactate dehydrogenase (LDH), superoxide dismutase (SOD), catalase (CAT) and creatine kinase (CK) were determined using commercial kits following the manufacture’s instruction. mRNA expressions (caspase-3, caspase-8, caspase-9 and Fas) were determined by quantitative real time-polymerase chain reaction (RT-qPCR). Results: The obtained optimal extraction conditions for BRP was as follows: extraction time (1.43 h), ratio of water to the raw material (30 mL/g) and extraction times (2 times). BRP (200, 400, 600 and 800 μg/mL) significantly increased the cell viability of H2O2 induced H9c2 cells (p < 0.05, p < 0.01, p < 0.01, p < 0.01, respectively). BRP (200, 400 and 800 μg/mL) significantly decreased LDH and CK levels (p < 0.01, p < 0.01, p < 0.01, respectively). However, BRP increased levels of SOD (200, 400 and 800 μg/mL, p < 0.05) and CAT (400 and 800 μg/mL, p < 0.05) in H9c2 cells. BRP significantly downregulated mRNA expressions of Caspase-3, Caspase-8, Caspase-9 and Fas (200, 400 and 800 μg/mL, p < 0.01) in H9c2 cells induced by H2O2. Conclusion: BRP protects cardiomyocyte against apoptosis via inhibition of oxidative stress and mitochondria-mediated intrinsic apoptosis, and thus, may be potential therapeutic agent for the management of cardiovascular diseases.     Insect pest management with sex pheromone precursors from engineered oilseed plants

    Pheromones have become an environmentally friendly alternative to conventional insecticides for pest control. Most current pheromone-based pest control products target lepidopteran pests of high-value crops, as today’s manufacturing processes cannot yet produce pheromones at low enough costs to enable their use for lower-value crops, especially commodity crops. Camelina sativa seeds genetically modified to express (Z)-11-hexadecenoic acid, a sex pheromone precursor of several moth species, provided the oil from which the precursor was isolated, purified and transformed into the final pheromone. Trap lures containing this pheromone were then assessed for their capacity to manage moth pests in the field. Plant-derived pheromone lures proved equally effective as synthetic pheromone lures in monitoring the diamondback moth, Plutella xylostella, in cabbage and disrupting mating of cotton bollworm, Helicoverpa armigera, in common bean fields.
