Ontogenetic changes in DNA and RNA content of laboratory-reared Prochilodus lineatus larvae: use of RNA/DNA ratios as indicators of nutritional condition
The aims of the present study were to describe the ontogenetic performance of DNA and RNA content, as well as to assess the nutritional condition of Prochilodus lineatus larvae reared under different feeding treatments. The experiment consisted of two parts, the first 6-14 days after hatching (DAH) and the second 15-21 DAH. In the first part of the experiment, larvae were kept under one of two feeding treatments: fed or starved. During the second part of the experiment, larvae were kept under one of four treatments: fed, fed until 14 DAH and later starved, starved until 14 DAH and fed later or starved. It is of note that the size and weight reached of larvae that were starved during early development and later received food were higher than those of than larvae in the other feeding treatments. In addition, instantaneous growth rates were higher and nutritional condition was better in larvae that were initially starved and fed later. The availability of food following a period of starvation seems to trigger larval metabolism and cellular division. Consequently, we recommend maintaining appropriate food provision in the aquaculture of P. lineatus larvae, especially in advanced developmental stages. We verified that RNA/DNA ratio is affected by the feeding condition and that it is sensitive enough to promptly reflect changes in feeding conditions of P. lineatus larvae. Thus, the RNA/DNA ratio could be used to assess nutritional condition of both wild and cultured P. lineatus larvae.Fil: Diaz, Marina Vera. Intracellular trafficking and cellular uptake mechanism of PHBV nanoparticles for targeted delivery in epithelial cell lines
Indexación: Web of Science; Scopus; Scielo.Background: Nanotechnology is a science that involves imaging, measurement, modeling and a manipulation of matter at the nanometric scale. One application of this technology is drug delivery systems based on nanoparticles obtained from natural or synthetic sources. An example of these systems is synthetized from poly(3-hydroxybutyrate-co-3-hydroxyvalerate), which is a biodegradable, biocompatible and a low production cost polymer. The aim of this work was to investigate the uptake mechanism of PHBV nanoparticles in two different epithelial cell lines (HeLa and SKOV-3).
Results: As a first step, we characterized size, shape and surface charge of nanoparticles using dynamic light scattering and transmission electron microscopy. Intracellular incorporation was evaluated through flow cytometry and fluorescence microscopy using intracellular markers. We concluded that cellular uptake mechanism is carried out in a time, concentration and energy dependent way. Our results showed that nanoparticle uptake displays a cell-specific pattern, since we have observed different colocalization in two different cell lines. In HeLa (Cervical cancer cells) this process may occur via classical endocytosis pathway and some internalization via caveolin-dependent was also observed, whereas in SKOV-3 (Ovarian cancer cells) these patterns were not observed. Rearrangement of actin filaments showed differential nanoparticle internalization patterns for HeLa and SKOV-3. Additionally, final fate of nanoparticles was also determined, showing that in both cell lines, nanoparticles ended up in lysosomes but at different times, where they are finally degraded, thereby releasing their contents.
Conclusions: Our results, provide novel insight about PHBV nanoparticles internalization suggesting that for develop a proper drug delivery system is critical understand the uptake mechanism.https://jnanobiotechnology.biomedcentral.com/articles/10.1186/s12951-016-0241-
Modeling Ovoproduct Spoilage with Red Led Light
Heat-treatment of ovoproducts is often required to ensure microbial safety. However, it has been shown that in most microbial species slow heating, or heat shocks may induce a higher heat resistance, that means that it is not possible to remove the microbial flora completely. These microorganisms produce on ovoproducts spoilage especially when the cold chain is broken along the transportation and/or storage. As a result, the life span for the product is shortened. The microbial activity inside the product causes changes in several physical properties, which can be supervised using optical methods. The aim of this work is to monitor the sigmoid behaviour underlying the ovoproduct evolution and spoilage by means of red LED light. For two commercial types of liquid and pasteurized egg white, storage at 9°C, an average correlation of r=0.94 has been found between microorganism growth and mean grey level of LED light passing though the sample. The results show that it is possible to develop very simple predictive models taking into account only one optical parameter corre¬sponding to a single LED

La extrema sensibilidad de los ovoproductos al deterioro microbiano hace necesaria la aplicación de un tratamiento térmico que asegure su inocuidad. El hecho de que el tratamiento térmico no asegure la completa erradicación de la flora microbiana junto con la falta de mantenimiento de la cadena de frío a lo largo de la distribución y/o conservación del mismo limita y acorta la vida útil del producto. La degradación microbiana del ovoproducto afecta a ciertas propiedades físicas del mismo cuya alteración
puede ser objeto de supervisión mediante técnicas ópticas siendo el objetivo de este trabajo la realización de un estudio preliminar para la supervisión de la degradación de ovoproducto mediante LED con el objeto de hacer una propuesta de implantación en laboratorios de calidad en la industria. Los resultados muestran que en clara líquida pasteurizada el aumento del contenido microbiano a lo largo del tiempo de almacenamiento se correlaciona (con valores de hasta r = 0.97) con el incremento en el nivel de
intensidad lumínica detectado en las muestras. En la yema a partir de niveles altos de concentración de microorganismos los fenómenos de turbidez y gelificación se identifican con una disminución en los valores de los parámetros ópticos considerados

La extrema sensibilidad de los ovoproductos al deterioro microbiano hace necesaria la aplicacion de un tratamiento termico que asegure su inocuidad. El hecho de que el tratamiento termico no asegure la completa erradicacion de la flora microbiana junto con la falta de mantenimiento de la cadena de fn'o a lo largo de la distribucion y/o conservacion del mismo limita y acorta la vida util del producto. La degradacion microbiana del ovoproducto afecta a ciertas propiedades ffsicas del mismo cuya alteration puede ser objeto de supervision mediante tecnicas opticas siendo el objetivo de este trabajo la realization de un estudio preliminar para la supervision de la degradacion de ovoproducto mediante LED con el objeto de hacer una propuesta de implantation en laboratories de calidad en la industria. Unsuspected pulmonary alveolar proteinosis in a patient with acquired immunodeficiency syndrome: a case report
<p>Abstract</p> <p>Introduction</p> <p>Diffuse lung infiltrates are a common finding in patients with acquired immunodeficiency syndrome and causes range from infectious processes to malignancies or interstitial lung diseases. Pulmonary alveolar proteinosis is a rare pulmonary disorder rarely reported in patients infected with human immunodeficiency virus. Secondary pulmonary alveolar proteinosis is associated with conditions involving functional impairment or reduced numbers of alveolar macrophages. It can be caused by hematologic malignancies, inhalation of toxic dust, fumes or gases, infectious or pharmacologic immunosuppression, or lysinuric protein intolerance.</p> <p>Case presentation</p> <p>A 42-year-old African American man infected with human immunodeficiency virus was admitted with chronic respiratory symptoms and diffuse pulmonary infiltrates. Chest computed tomography revealed bilateral spontaneous pneumothoraces, for which he required bilateral chest tubes. Initial laboratory investigations did not reveal any contributory conditions. Histological examination of a lung biopsy taken during video-assisted thoracoscopy showed pulmonary alveolar proteinosis concurrent with cytomegalovirus pneumonitis. After ganciclovir treatment, our patient showed radiologic and clinical improvement.</p> <p>Conclusion</p> <p>The differential diagnosis for patients with immunosuppression and lung infiltrates requires extensive investigations. As pulmonary alveolar proteinosis is rare, the diagnosis can be easily missed. Our case highlights the importance of invasive investigations and histology in the management of patients infected with human immunodeficiency virus and pulmonary disease who do not respond to empiric therapy.</p
Looking for Crumbs in the Obesity Forest: Anti-obesity Interventions and Obesity-Associated Cardiometabolic Traits in the Mexican Population. History and Systematic Review With Meta-Analyses
Mexicans and Mexican Americans share culture, genetic background, and predisposition for chronic complications associated with obesity and diabetes making imperative efficacious treatments and prevention. Obesity has been treated for centuries focused-on weight loss while other treatments on associated conditions like gout, diabetes (T2D), and hypertriglyceridemia. To date, there is no systematic review that synthesizes the origin of obesity clinics in Mexico and the efforts to investigate treatments for obesity tested by randomized clinical trials (RCT). We conducted systematic searches in Pubmed, Scopus, and Web of Science to retrieve anti-obesity RCT through 2019 and without an inferior temporal limit. The systematic review included RCT of anti-obesity treatments in the Mexican adult population, covering alternative medicine, pharmacological, nutritional, behavioral, and surgical interventions reporting metabolism-associated traits such as BMI, weight, waist circumference, triglycerides, glucose, among others. Only the studies with at least 3 months of treatment were included in the meta-analyses in order to reduce placebo effects. We found 634 entries, after removal of duplicates and screening the studies based on eligibility criteria, we analyzed 43 national, and 2 multinational-collaborative studies. Most of the national studies had small sample sizes, and the implemented strategies do not have replications in the population. The nutrition/behavioral interventions were difficult to blind, and most studies have medium-to-high risk of bias. Nutritional/behavioral interventions and medications showed effects on BMI, waist circumference, and blood pressure. Simple measures like pure water instead of sweet beverages decrease triglycerides and systolic blood pressure. Dark chocolate showed the highest effect for BMI and high blood pressure, and treatment with insulin increased weight in those with T2D. The study of obesity in Mexico has been on-going for more than four decades, the interest on RCT just increased until this millennium, but with small sample sizes and lack of replication. The interventions affect different cardiometabolic associated traits, which should be analyzed in detail in the population living near the Mexico-U.S. border; therefore, bi-national collaboration is desirable to disentangle the cultural effects on this population\u27s treatment response
