    Intestinal model of Inflammation in primary cells

    The gastrointestinal tract contains an enormous mucosal surface, which is continuously exposed to antigens hence making it susceptible to an inflammatory response. Such response targets potential pathogens by direct activation of the mucosal immune cells, however, in newborns the continuous inflammation attacks the intestine which leads to induction of necrotising enterocolitis. The present study aims at developing an “in-vitro” intestinal model of inflammation to assist in understanding the complex interplay of pro-inflammatory mediators during the immune response in neonates. Segments (1.5cm length) from the ileum were obtained from SD rat neonates (1-4 days old) and exposed to 0.25% trypsin/EDTA for 30min. Following trituration and subsequent centrifugation for 5min at 450xg, cells were suspended in DMEM-Hepes supplemented with 10% FCS, 2.5% Penicillin/Streptomycin, 2.5% L-Glutamine, and 0.2% Amphotericin B. Cell suspension were transferred to culture flaks and incubated at 37°C. Once confluent, the cell preparation media was replaced by FCS-free media, and treated with 0, 10, 50, and 100'g/ml of LPS. IL-8 and nitric oxide (NO) response were subsequently measured. In separate studies cell proliferation, cell viability, and cell adhesion were analysed. Additionally, the phenotypic properties of the intestinal muscle cells were also investigated via immunocytochemistry. Initial studies demonstrated that LPS treatments induced a significant increase in the release of IL-8 and NO compared to controls. The effect of LPS treatments on cell dynamics demonstrated small changes in cell viability and adhesion, whereas an increase in cell proliferation was observed. Immunocytochemistry studies indicated that LPS treatment caused a decrease in the expression of actin fibers with impaired distribution compared to controls. In the present model key aspects of intestinal inflammation were replicated “in-vitro” including the activation of pro-inflammatory mediators, the loss in enteric innervations and subsequent tissue hyperplasia. Thus, this model may be used as a tool to investigate the anti-inflammatory properties of candidate drugs targeting functional GI diseases

    Late Cretaceous and Paleogene radiolaria from the Nicoya Peninsula, Costa Rica: A tectonostratigraphic application

    Detailed field mapping and paleontological dating in the central and southeastern Nicoya Peninsula has revealed Late Cretaceous and Paleogene radiolarian-bearing siliceous mudstones. These rocks belong to two terranes (Matambú and Manzanillo) that are partially contemporaneous with the Nicoya Complex, but are genetically different. While the Nicoya Complex is formed exclusively by intraplate igneous rocks with associated radiolarites, the studied sections include variable amounts of arc-derived volcanic and terrigenous materials. These fore-arc terranes include mafic to intermediate volcaniclastics and associated pelagic and hemipelagic rocks rich in biogenic silica. Radiolarian preservation in these sediments is often enhanced by the presence of silica-saturated volcanic tuffs and debris. Seven out of 29 samples from different outcrops yielded relatively well-preserved radiolarian faunas. In total, 60 species belonging to 34 genera were present in these faunas, ranging in age from middle Turonian-Santonian to late Thanetian-Ypresian

    Formation of the Stomatal Outer Cuticular Ledge Requires a Guard Cell Wall Proline-Rich Protein

    Stomata are formed by a pair of guard cells which have thickened, elastic cell walls to withstand the large increases in turgor pressure that have to be generated to open the pore that they surround. We have characterised FOCL1, a guard cell-expressed, secreted protein with homology to hydroxyproline-rich cell wall proteins. FOCL1-GFP localises to the guard cell outer cuticular ledge and plants lacking FOCL1 produce stomata without a cuticular ledge. Instead the majority of stomatal pores are entirely covered-over by a continuous fusion of the cuticle, and consequently plants have decreased levels of transpiration and display drought tolerance. The focl1 guard cells are larger and less able to reduce the aperture of their stomatal pore in response to closure signals suggesting that the flexibility of guard cell walls is impaired. FOCL1 is also expressed in lateral root initials where it aids lateral root emergence. We propose that FOCL1 acts in these highly specialised cells of the stomata and root to impart cell wall strength at high turgor and/or to facilitate interactions between the cell wall and the cuticle

    Transcriptomic Analyses of Inner Ear Sensory Epithelia in Zebrafish

    Analysis of gene expression has the potential to assist in the understanding of multiple cellular processes including proliferation, cell-fate specification, senesence, and activity in both healthy and disease states. Zebrafish model has been increasingly used to understand the process of hearing and the development of the vertebrate auditory system. Within the zebrafish inner ear, there are three otolith organs, each containing a sensory macula of hair cells. The saccular macula is primarily involved in hearing, the utricular macula is primarily involved in balance and the function of the lagenar macula is not completely understood. The goal of this study is to understand the transcriptional differences in the sensory macula associated with different otolith organs with the intention of understanding the genetic mechanisms responsible for the distinct role each organ plays in sensory perception. The sensory maculae of the saccule, utricle, and lagena were dissected out of adult Et(krt4:GFP) zebrafish expressing green fluorescent protein in hair cells for transcriptional analysis. The total RNAs of the maculae were isolated and analyzed by RNA GeneChip microarray. Several of the differentially expressed genes are known to be involved in deafness, otolith development and balance. Gene expression among these otolith organs was very well conserved with less than 10% of genes showing differential expression. Data from this study will help to elucidate which genes are involved in hearing and balance. Furthermore, the findings of this study will assist in the development of the zebrafish model for human hearing and balance disorders.     More than sense of place? Exploring the emotional dimension of rural tourism experiences

    It is widely suggested that participation in rural tourism is underpinned by a sense of rural place or “rurality”. However, although nature and the countryside have long been recognised as a source of spiritual or emotional fulfilment, few have explored the extent to which tourism, itself often claimed to be a sacred experience, offers an emotional/spiritual dimension in the rural context. This paper addresses that literature gap. Using in-depth interviews with rural tourists in the English Lake District, it explores the extent to which, within respondents’ individual understanding of spirituality, a relationship exists between sense of place and deeper, emotional experiences and, especially, whether participation in rural tourism may induce spiritual or emotional responses. The research revealed that all respondents felt a strong attachment to the Lake District; similarly, and irrespective of their openness to spirituality, engaging in rural tourism activities resulted in highly emotive experiences for all respondents, the description/interpretation of such experiences being determined by individual “beliefs”. However, sense of place was not a prerequisite to emotional or spiritual experiences. Being in and engaging with the landscape � effectively becoming part of it � especially through physical activity is fundamental to emotional responses

    Creating university spin-offs: A science-based design perspective

    Academic entrepreneurship by means of university spin-offs commercializes technological breakthroughs, which may otherwise remain unexploited. However, many universities face difficulties in creating spin-offs. This article adopts a science-based design approach to connect scholarly research with the pragmatics of effectively creating university spin-offs. This approach serves to link the practice of university spin-off creation, via design principles, to the scholarly knowledge in this area. As such, science-based design promotes the interplay between emergent and deliberate design processes. This framework is used to develop a set of design principles that are practice based as well as grounded in the existing body of research on university spin-offs. A case-study of spin-off creation at a Dutch university illustrates the interplay between initial processes characterized by emergent design and the subsequent process that was more deliberate in nature. This case study also suggests there are two fundamentally different phases in building capacity for university spin-off creation. First, an infrastructure for spin-off creation (including a collaborative network of investors, managers and advisors) is developed that then enables support activities to individual spin-off ventures. This study concludes that to build and increase capacity for creating spin-offs, universities should do the following: (1) create university-wide awareness of entrepreneurship opportunities, stimulate the development of entrepreneurial ideas, and subsequently screen entrepreneurs and ideas by programs targeted at students and academic staff; (2) support start-up teams in composing and learning the right mix of venturing skills and knowledge by providing access to advice, coaching, and training; (3) help starters in obtaining access to resources and developing their social capital by creating a collaborative network organization of investors, managers, and advisors; (4) set clear and supportive rules and procedures that regulate the university spin-off process, enhance fair treatment of involved parties, and separate spin-off processes from academic research and teaching; and (5) shape a university culture that reinforces academic entrepreneurship by creating norms and exemplars that motivate entrepreneurial behavior. These and other results of this study illustrate how science-based design can connect scholarly research to the pragmatics of actually creating spin-offs in academic institutions

    Motivation and Knowledge Sharing through Social Media within Danish Organizations

    Part 3: Creating Value through ApplicationsInternational audienceBased on an empirical quantitative study, this article investigates employee motivation in Danish companies and aims at determining which factors affect employees’ knowledge sharing through social media in a working environment. Our findings pinpoint towards the potential social media have for enhancing internal communication, knowledge sharing and collaboration in organizations, but the adoption is low, at this point, due to mainly organizational and individual factors. Technological factors do not seem to affect employees’ motivation for knowledge sharing as much as previous research has found, but it is the influence from the combination of individual and organizational factors, which affect the adoption of the platforms. A key finding in the study is that knowledge sharing is not a ‘social dilemma’ as previous studies have found. The study shows a positive development in employees’ willingness to share knowledge, because knowledge sharing is considered more beneficial than to hoard it

    Leptin Matures Aspects of Lung Structure and Function in the Ovine Fetus

    In human and ovine fetuses, glucocorticoids stimulate leptin secretion, although the extent to which leptin mediates the maturational effects of glucocorticoids on pulmonary development is unclear. This study investigated the effects of leptin administration on indices of lung structure and function before birth. Chronically catheterized singleton sheep fetuses were infused iv for 5 days with either saline or recombinant ovine leptin (0.5 mg/kg · d leptin (LEP), 0.5 LEP or 1.0 mg/kg · d, 1.0 LEP) from 125 days of gestation (term ∼145 d). Over the infusion, leptin administration increased plasma leptin, but not cortisol, concentrations. On the fifth day of infusion, 0.5 LEP reduced alveolar wall thickness and increased the volume at closing pressure of the pressure-volume deflation curve, interalveolar septal elastin content, secondary septal crest density, and the mRNA abundance of the leptin receptor (Ob-R) and surfactant protein (SP) B. Neither treatment influenced static lung compliance, maximal lung volume at 40 cmH2O, lung compartment volumes, alveolar surface area, pulmonary glycogen, protein content of the long form signaling Ob-Rb or phosphorylated signal transducers and activators of transcription-3, or mRNA levels of SP-A, C, or D, elastin, vascular endothelial growth factor-A, the vascular endothelial growth factor receptor 2, angiotensin-converting enzyme, peroxisome proliferator-activated receptor γ, or parathyroid hormone-related peptide. Leptin administration in the ovine fetus during late gestation promotes aspects of lung maturation, including up-regulation of SP-B
