    Angiotensin-converting enzyme 2 (ACE2) from raccoon dog can serve as an efficient receptor for the spike protein of severe acute respiratory syndrome coronavirus

    Raccoon dog is one of the suspected intermediate hosts of severe acute respiratory syndrome coronavirus (SARS-CoV). In this study, the angiotensin-converting enzyme 2 (ACE2) gene of raccoon dog (rdACE2) was cloned and sequenced. The amino acid sequence of rdACE2 has identities of 99.3, 89.2, 83.9 and 80.4% to ACE2 proteins from dog, masked palm civet (pcACE2), human (huACE2) and bat, respectively. There are six amino acid changes in rdACE2 compared with huACE2, and four changes compared with pcACE2, within the 18 residues of ACE2 known to make direct contact with the SARS-CoV S protein. A HeLa cell line stably expressing rdACE2 was established; Western blot analyses and an enzyme-activity assay indicated that the cell line expressed ACE2 at a similar level to two previously established cell lines that express ACE2 from human and masked palm civet, respectively. Human immunodeficiency virus-backboned pseudoviruses expressing spike proteins derived from human SARS-CoV or SARS-CoV-like viruses of masked palm civets and raccoon dogs were tested for their entry efficiency into these cell lines. The results showed that rdACE2 is a more efficient receptor for human SARS-CoV, but not for SARS-CoV-like viruses of masked palm civets and raccoon dogs, than huACE2 or pcACE2.     Helicoverpa armigera nucleopolyhedrovirus occlusion-derived virus-associated protein, HA100, affects oral infectivity in vivo but not virus replication in vitro

    ORF100 (ha100) of Helicoverpa armigera nucleopolyhedrovirus (HearNPV) has been reported as one of the unique genes of group II alphabaculoviruses encoding a protein located in the occlusion-derived virus (ODV) envelope and nucleocapsid. The protein consists of 510 aa with a predicted mass of 58.1 kDa and is a homologue of poly(ADP–ribose) glycohydrolase in eukaryotes. Western blot analysis detected a 60 kDa band in HearNPV-infected HzAM1 cells starting at 18 h post-infection. Transient expression of GFP-fused HA100 in HzAM1 cells resulted in cytoplasmic localization of the protein, but after superinfection with HearNPV, GFPfused HA100 was localized in the nucleus. To study the function of HA100 further, an ha100-null virus was constructed using bacmid technology. Viral one-step growth curve analyses showed that the ha100-null virus had similar budded virus production kinetics to that of the parental virus. Electron microscopy revealed that deletion of HA100 did not alter the morphology of ODVs or occlusion bodies (OBs). However, bioassays in larvae showed that the 50% lethal concentration (LC50) value of HA100-null OBs was significantly higher than that of parental OBs; the median lethal time (LT50) of ha100-null OBs was about 24 h later than control virus. These results indicate that HA100 is not essential for virus replication in vitro. However, it significantly affects the oral infectivity of OBs in host insects, suggesting that the association HA100 with the ODV contributes to the infectivity of OBs in vivo

    Spike Effects on Drag Reduction for Hypersonic Lifting Body

    A high lift-to-drag ratio is considered crucial for high-altitude and long-endurance hypersonic vehicles. One of the simplest and most useful methods is to install an aerospike in front of the vehicle’s nose. In this paper, the flight aerodynamic characteristics are investigated by simulating and comparing the lifting body with or without the aerospikes at Ma=8. The flowfields around aerospikes using different spike lengths and a hemispherical disk along with the lifting body are analyzed. The results of aerodynamic characteristics indicate that L/D=2 is the best ratio of the spike length to the nose diameter. By comparing with the baseline model, the maximum drag reduction of the nose’s part is 49.3% at α=8  deg using a hemispherical disk. In addition, three shapes of aerospike disks are compared to search for the best disk for hypersonic drag reduction. The best drag reduction is found for the double flat-faced disk aerospike, which gives a pressure drag reduction of 60.5% of the nose’s part at α=8  deg. Furthermore, when the flight angle of attack increases, the drag increases significantly. Employing a certain installation angle is shown to effectively improve the drag reduction around the angle of attack and results in improving the lift-to-drag ratio. At the end, the lift-to-drag ratio of the final optimized design is 9.1% better than that of the baseline model. The pressure center is moved forward by 1.6%, barely influencing the vertical static stability of the vehicle

    Precision Enhancement of Pavement Roughness Localization with Connected Vehicles

    Raj Bridgelall is the program director for the Upper Great Plains Transportation Institute (UGPTI) Center for Surface Mobility Applications & Real-time Simulation environments (SMARTSeSM).Transportation agencies rely on the accurate localization and reporting of roadway anomalies that could pose serious hazards to the traveling public. However, the cost and technical limitations of present methods prevent their scaling to all roadways. Connected vehicles with on-board accelerometers and conventional geospatial position receivers offer an attractive alternative because of their potential to monitor all roadways in real-time. The conventional global positioning system is ubiquitous and essentially free to use but it produces impractically large position errors. This study evaluated the improvement in precision achievable by augmenting the conventional geofence system with a standard speed bump or an existing anomaly at a pre-determined position to establish a reference inertial marker. The speed sensor subsequently generates position tags for the remaining inertial samples by computing their path distances relative to the reference position. The error model and a case study using smartphones to emulate connected vehicles revealed that the precision in localization improves from tens of metres to sub-centimetre levels, and the accuracy of measuring localized roughness more than doubles. The research results demonstrate that transportation agencies will benefit from using the connected vehicle method to achieve precision and accuracy levels that are comparable to existing laser-based inertial profilers.Mountain Plains Consortium (MPC)https://www.ugpti.org/about/staff/viewbio.php?id=7

    Efficient symmetric multiparty quantum state sharing of an arbitrary m-qubit state

    We present a scheme for symmetric multiparty quantum state sharing of an arbitrary mm-qubit state with mm Greenberger-Horne-Zeilinger states following some ideas from the controlled teleportation [Phys. Rev. A \textbf{72}, 02338 (2005)]. The sender Alice performs mm Bell-state measurements on her 2m2m particles and the controllers need only to take some single-photon product measurements on their photons independently, not Bell-state measurements, which makes this scheme more convenient than the latter. Also it does not require the parties to perform a controlled-NOT gate on the photons for reconstructing the unknown mm-qubit state and it is an optimal one as its efficiency for qubits approaches the maximal value.Comment: 6 pages, no figures; It simplifies the process for sharing an arbitrary m-qubit state in Phys.
