    Cryptocapsinepoxide-type Carotenoids from Red Mamey, Pouteria sapota

    Three new carotenoids, cryptocapsin-5,6-epoxide, 3ʹ-deoxycapsanthin-5,6-epoxide, and cryptocapsin-5,8-epoxides, have been isolated from the ripe fruits of red mamey (Pouteria sapota). Cryptocapsin-5,6-epoxide was prepared by partial synthesis via epoxidation of cryptocapsin and the (5R,6S)- and (5S,6R)-stereoisomers were identified by HPLC-ECD analysis. Spectroscopic data of the natural (anti) and semisynthetic (syn) derivatives obtained by acid-catalyzed rearrangement of cryptocapsin-5,8-epoxide stereoisomers were compared for structural elucidation. Chiral HPLC separation of natural and semisynthetic samples of cryptocapsin-5,8-epoxides was performed and HPLC-ECD analysis allowed configurational assignment of the separated stereoisomers

    Development of electrocardiogram intervals during growth of FVB/N neonate mice

    <p>Abstract</p> <p>Background</p> <p>Electrocardiography remains the best diagnostic tool and therapeutic biomarker for a spectrum of pediatric diseases involving cardiac or autonomic nervous system defects. As genetic links to these disorders are established and transgenic mouse models produced in efforts to understand and treat them, there is a surprising lack of information on electrocardiograms (ECGs) and ECG abnormalities in neonate mice. This is likely due to the trauma and anaesthesia required of many legacy approaches to ECG recording in mice, exacerbated by the fragility of many mutant neonates. Here, we use a non-invasive system to characterize development of the heart rate and electrocardiogram throughout the growth of conscious neonate FVB/N mice.</p> <p>Results</p> <p>We examine ECG waveforms as early as two days after birth. At this point males and females demonstrate comparable heart rates that are 50% lower than adult mice. Neonatal mice exhibit very low heart rate variability. Within 12 days of birth PR, QRS and QTc interval durations are near adult values while heart rate continues to increase until weaning. Upon weaning FVB/N females quickly develop slower heart rates than males, though PR intervals are comparable between sexes until a later age. This suggests separate developmental events may contribute to these gender differences in electrocardiography.</p> <p>Conclusions</p> <p>We provide insight with a new level of detail to the natural course of heart rate establishment in neonate mice. ECG can now be conveniently and repeatedly used in neonatal mice. This should serve to be of broad utility, facilitating further investigations into development of a diverse group of diseases and therapeutics in preclinical mouse studies.</p

    Improving Ecological Restoration to Curb Biotic Invasion—A Practical Guide

    Common practices for invasive species control and management include physical, chemical, and biological approaches. The first two approaches have clear limitations and may lead to unintended (negative) consequences, unless carefully planned and implemented. For example, physical removal rarely completely eradicates the targeted invasive species and can cause disturbances that facilitate new invasions by nonnative species from nearby habitats. Chemical treatments can harm native, and especially rare, species through unanticipated side effects. Biological methods may be classified as biocontrol and the ecological approach. Similar to physical and chemical methods, biocontrol also has limitations and sometimes leads to unintended consequences. Therefore, a relatively safer and more practical choice may be the ecological approach, which has two major components: (1) restoration of native species and (2) biomass manipulation of the restored community, such as selective grazing or prescribed burning (to achieve and maintain viable population sizes). Restoration requires well-planned and implemented planting designs that consider alpha-, beta-, and gamma-diversity and the abundance of native and invasive component species at local, landscape, and regional levels. Given the extensive destruction or degradation of natural habitats around the world, restoration could be most effective for enhancing ecosystem resilience and resistance to biotic invasions. At the same time, ecosystems in human-dominated landscapes, especially those newly restored, require close monitoring and careful intervention (e.g., through biomass manipulation), especially when successional trajectories are not moving as intended. Biomass management frequently uses prescribed burning, grazing, harvesting, and thinning to maintain overall ecosystem health and sustainability. Thus, the resulting optimal, balanced, and relatively stable ecological conditions could more effectively limit the spread and establishment of invasive species. Here we review the literature (especially within the last decade) on ecological approaches that involve biodiversity, biomass, and productivity, three key community/ecosystem variables that reciprocally influence one another. We focus on the common and most feasible ecological practices that can aid in resisting new invasions and/or suppressing the dominance of existing invasive species. We contend that, because of the strong influences from neighboring areas (i.e., as exotic species pools), local restoration and management efforts in the future need to consider the regional context and projected climate changes

    Exercise and redox status responses following alpha-lipoic acid supplementation in G6PD deficient individuals

    G6PD deficiency renders cells more susceptible to oxidative insults, while antioxidant dietary supplementation could restore redox balance and ameliorate exercise-induced oxidative stress. To examine the effects of alpha-lipoic acid (ALA) supplementation on redox status indices in G6PD deficient individuals, eight male adults with G6PD deficiency (D) participated in this randomized double-blind placebo-controlled crossover trial. Participants were randomly assigned to receive ALA (600 mg/day) or placebo for 4 weeks separated by a 4-week washout period. Before and at the end of each treatment period, participants exercised following an exhaustive treadmill exercise protocol. Blood samples were obtained before (at rest), immediately after and 1h after exercise for later analysis of total antioxidant capacity (TAC), uric acid, bilirubin, thiobarbituric acid reactive substances (TBARS) and protein carbonyls (PC). ALA resulted in significantly increased resting TAC and bilirubin concentrations. Moreover, TAC increased immediately and 1h after exercise following both treatment periods, whereas bilirubin increased immediately after and 1h after exercise following only ALA. No significant change in uric acid, TBARS or PC was observed at any time point. ALA supplementation for 4 weeks may enhance antioxidant status in G6PD individuals; however, it does not affect redox responses to acute exercise until exhaustion or exercise performance

    Iron Supplementation Effects on Redox Status following Aseptic Skeletal Muscle Trauma in Adults and Children.

    Exercise-induced skeletal muscle microtrauma is characterized by loss of muscle cell integrity, marked aseptic inflammatory response, and oxidative stress. We examined if iron supplementation would alter redox status after eccentric exercise. In a randomized, double blind crossover study, that was conducted in two cycles, healthy adults (n = 14) and children (n = 11) received daily either 37 mg of elemental iron or placebo for 3 weeks prior to and up to 72 h after an acute eccentric exercise bout. Blood was drawn at baseline, before exercise, and 72 h after exercise for the assessment of iron status, creatine kinase activity (CK), and redox status. Iron supplementation at rest increased iron concentration and transferrin saturation (p < 0.01). In adults, CK activity increased at 72 h after exercise, while no changes occurred in children. Iron supplementation increased TBARS at 72 h after exercise in both adults and children; no changes occurred under placebo condition. Eccentric exercise decreased bilirubin concentration at 72 h in all groups. Iron supplementation can alter redox responses after muscle-damaging exercise in both adults and children. This could be of great importance not only for healthy exercising individuals, but also in clinical conditions which are characterized by skeletal muscle injury and inflammation, yet iron supplementation is crucial for maintaining iron homeostasis.     French responses to the Prague Spring: connections, (mis)perception and appropriation

    Looking at the vast literature on the events of 1968 in various European countries, it is striking that the histories of '1968' of the Western and Eastern halves of the continent are largely still written separately.1 Nevertheless, despite the very different political and socio-economic contexts, the protest movements on both sides of the Iron Curtain shared a number of characteristics. The 1968 events in Czechoslovakia and Western Europe were, reduced to the basics, investigations into the possibility of marrying social justice with liberty, and thus reflected a tension within European Marxism. This essay provides an analysis specifically of the responses by the French left—the Communist Party, the student movements and the gauchistes—to the Prague Spring, characterised by misunderstandings and strategic appropriation. The Prague Spring was seen by both the reformist and the radical left in France as a moderate movement. This limited interpretation of the Prague Spring as a liberal democratic project continues to inform our memory of it
