    Overexpression of hIGF-1 exclusively in skeletal muscle increases the number of dihydropyridine receptors in adult transgenic mice

    AbstractThe number of dihydropyridine receptors (DHPR) and sarcoplasmic reticulum (SR) Ca2+ release channels (RyR1) and their interaction determine the efficacy of the sarcolemmal excitation-SR Ca2+ release-contraction coupling (ECC). Both receptors play a central role in ECC as demonstrated in various animal species and muscle subtypes. In the present work we studied the effect of transgenic overexpression of human insulin-like growth factor 1 (hIGF-1) on the levels of these two Ca2+ channels in extensor digitorum longus (EDL) (fast-twitch), soleus (slow-twitch) and pool of fast- and slow-twitch muscles from adult C57BL/6 mice. Muscles from hIGF-1 transgenic mice showed a significant increase in IGF-1 concentration (20–30-fold) and in the number of DHPR (52% increase) whereas no significant change in RyR1 binding sites was detected. The differential effect on DHPR and RyR1 resulted in a 30% increase in DHPR/RyR1 ratio. Fast- and slow-twitch muscles showed 50 and 70% increase in the number of DHPR and 30 and 80% increase in DHPR/RyR1, respectively. These results support the concept that the increased autocrine/paracrine secretion of hIGF-1 exerts potent stimulatory effects on DHPR α1 subunit expression in adult skeletal muscle

    Radioactivity concentrations and their radiological significance in sediments of the Tema Harbour (Greater Accra, Ghana)

    Abstract Studies on environmental radioactivity in tropical Africa are scarce. Therefore, a baseline study of natural ( 238 U, 210 Pb, 226 Ra, 232 Th, 228 Ra, 228 Th, 40 K) and anthropogenic ( 137 Cs) radionuclides was carried out on Tema Harbour (Greater Accra, Ghana) surface sediments and on their radiological significance. Grab surface sediment samples were collected from 21 stations within the Tema Harbour and their radioactivity concentrations measured by gamma spectrometry. The mean sediment radioactivity concentrations (Bq kg −1 dw) were 34 for 238 U, 210 for 210 Pb, 14 for 226 Ra, 30 for 232 Th, 29 for 228 Ra, 31 for 228 Th, 320 for 40 K, and 1.5 for 137 Cs. Large 238 U/ 226 Ra disequilibria were observed in the harbour sediments and a complex dynamics of several mixed sources of sediments within the Tema Harbour can be inferred from the spatial variations in the radioactivity concentrations. The estimated total absorbed dose rate in air ( D ), radium equivalent activity (Ra eq ), external hazard index ( H ex ), annual gonadal dose equivalent (AGDE) and annual effective dose equivalent (AEDE) indicated no significant radiological risks from the sediment radioactivity concentrations. Application of the Environmental Risk from Ionising Contaminants Assessment and Management tool (ERICA) confirmed that the potential dose rates to biota from the sediment radioactivity concentrations are unlikely to pose appreciable ecological risks.     Settling fluxes and sediment accumulation rates by the combined use of sediment traps and sediment cores in Tema Harbour (Ghana)

    Settling fluxes and sediment accumulation rates in coastal Tema Harbour (Ghana) were investigated by the combined analyses of results in sediment traps and sediment cores. Sediment traps were deployed at 5 stations within the Tema Harbour at two sampling depths and were retrieved every two weeks till the end of 12 weeks to estimate the Settling Fluxes (SFs). Four sediment cores from the harbour were analysed for their radioactivity (7Be, 234Th, 210Pb, 212Pb, 226Ra, 40K and 137Cs) profiles to quantify Sediment Accumulation Rates (SARs). The sediment cores exhibited variable bulk density profiles, indicating highly dynamic and non-steady sedimentation conditions. 7Be-derived gross-estimates of very recent SARs using the constant flux-constant sedimentation (CF-CS) model were in the range of 2.5–9.0 g·cm− 2·y− 1. These values were much lower than the estimated average SFs (15.2–53.8 g·cm− 2·y− 1), indicating sediment resuspension plays an important role. On a decadal time scale, conventional 210Pb sediment dating models did not allow any estimation of SARs in the Tema Harbour. Thus, the 210Pb-based TERESA model was applied to depict a reliable scenario for sedimentation with time-averaged SARs in the range of 1.4–3.0 g·cm− 2·y− 1 and fluxes of matter contributed by the marine inflow and local sources. Sediment accretion rates of 1.7–3 cm·y− 1 were also inferred, which may pose a moderate problem of sustainability for the Tema Harbour. This study reveals how the geochemical behaviour of different radionuclides with Gamma spectrometry in the marine environment can be used to obtain reliable information on the complex dynamics of Suspended Particulate Matter (SPM), even in a very disturbed and anthropic environment as a coastal harbour area where (1) conventional 210Pb-based dating methods fail and (2) the use of sediment traps and 234Th and 7Be profiles in sediment cores show serious constraints.University of Ghana UGFD/7/2012-2013/00

    BDA-410 Treatment Reduces Body Weight and Fat Content by Enhancing Lipolysis in Sedentary Senescent Mice

    Loss of muscle mass and force with age leads to fall risk, mobility impairment, and reduced quality of life. This article shows that BDA-410, a calpain inhibitor, induced loss of body weight and fat but not lean mass or skeletal muscle proteins in a cohort of sedentary 23-month-old mice. Food and water intake and locomotor activity were not modified, whereas BDA-410 treatment decreased intramyocellular lipid and perigonadal fat, increased serum nonesterified fatty acids, and upregulated the genes mediating lipolysis and oxidation, lean phenotype, muscle contraction, muscle transcription regulation, and oxidative stress response. This finding is consistent with our recent report that lipid accumulation in skeletal myofibers is significantly correlated with slower fiber-contraction kinetics and diminished power in obese older adult mice. A proteomic analysis and immunoblot showed downregulation of the phosphatase PPP1R12B, which increases phosphorylated myosin half-life and modulates the calcium sensitivity of the contractile apparatus. This study demonstrates that BDA-410 exerts a beneficial effect on skeletal muscle contractility through new, alternative mechanisms, including enhanced lipolysis, upregulation of "lean phenotype-related genes," downregulation of the PP1R12B phosphatase, and enhanced excitation- contraction coupling.     The sympathetic nervous system regulates skeletal muscle motor innervation and acetylcholine receptor stability

    Aim: Symptoms of autonomic failure are frequently the presentation of advanced age and neurodegenerative diseases that impair adaptation to common physiologic stressors. The aim of this work was to examine the interaction between the sympathetic and motor nervous system, the involvement of the sympathetic nervous system (SNS) in neuromuscular junction (NMJ) presynaptic motor function, the stability of postsynaptic molecular organization, and the skeletal muscle composition and function. Methods: Since muscle weakness is a symptom of diseases characterized by autonomic dysfunction, we studied the impact of regional sympathetic ablation on muscle motor innervation by using transcriptome analysis, retrograde tracing of the sympathetic outflow to the skeletal muscle, confocal and electron microscopy, NMJ transmission by electrophysiological methods, protein analysis, and state of the art microsurgical techniques, in C57BL6, MuRF1KO and Thy-1 mice. Results: We found that the SNS regulates motor nerve synaptic vesicle release, skeletal muscle transcriptome, muscle force generated by motor nerve activity, axonal neurofilament phosphorylation, myelin thickness, and myofibre subtype composition and CSA. The SNS also modulates the levels of postsynaptic membrane acetylcholine receptor by regulating the Gα i2 -Hdac4-Myogenin-MuRF1pathway, which is prevented by the overexpression of the guanine nucleotide-binding protein Gα i2 (Q205L), a constitutively active mutant G protein subunit. Conclusion: The SNS regulates NMJ transmission, maintains optimal Gα i2 expression, and prevents any increase in Hdac4, myogenin, MuRF1, and miR-206. SNS ablation leads to upregulation of MuRF1, muscle atrophy, and downregulation of postsynaptic AChR.     Expression of the α7 nicotinic acetylcholine receptor in human lung cells

    BACKGROUND: We and others have shown that one of the mechanisms of growth regulation of small cell lung cancer cell lines and cultured pulmonary neuroendocrine cells is by the binding of agonists to the α7 neuronal nicotinic acetylcholine receptor. In addition, we have shown that the nicotine-derived carcinogenic nitrosamine, 4(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), is a high affinity agonist for the α7 nicotinic acetylcholine receptor. In the present study, our goal was to determine the extent of α7 mRNA and protein expression in the human lung. METHODS: Experiments were done using reverse transcription polymerase chain reaction (RT-PCR), a nuclease protection assay and western blotting using membrane proteins. RESULTS: We detected mRNA for the neuronal nicotinic acetylcholine receptor α7 receptor in seven small cell lung cancer (SCLC) cell lines, in two pulmonary adenocarcinoma cell lines, in cultured normal human small airway epithelial cells (SAEC), one carcinoid cell line, three squamous cell lines and tissue samples from nine patients with various types of lung cancer. A nuclease protection assay showed prominent levels of α7 in the NCI-H82 SCLC cell line while α7 was not detected in SAEC, suggesting that α7 mRNA levels may be higher in SCLC compared to normal cells. Using a specific antibody to the α7 nicotinic receptor, protein expression of α7 was determined. All SCLC cell lines except NCI-H187 expressed protein for the α7 receptor. In the non-SCLC cells and normal cells that express the α7 nAChR mRNA, only in SAEC, A549 and NCI-H226 was expression of the α7 nicotinic receptor protein shown. When NCI-H69 SCLC cell line was exposed to 100 pm NNK, protein expression of the α7 receptor was increased at 60 and 150 min. CONCLUSION: Expression of mRNA for the neuronal nicotinic acetylcholine receptor α7 seems to be ubiquitously expressed in all human lung cancer cell lines tested (except for NCI-H441) as well as normal lung cells. The α7 nicotinic receptor protein is expressed in fewer cell lines, and the tobacco carcinogen NNK increases α7 nicotinic receptor protein levels
