    Comprehensive analysis of the membrane phosphoproteome regulated by oligogalacturonides in Arabidopsis thaliana

    Early changes in the Arabidopsis thaliana membrane phosphoproteome in response to oligogalacturonides (OGs), a class of plant damage-associated molecular patterns (DAMPs), were analyzed by two complementary proteomic approaches. Differentially phosphorylated sites were determined through phosphopeptide enrichment followed by LC-MS/MS using label-free quantification; differentially phosphorylated proteins were identified by 2D-DIGE combined with phospho-specific fluorescent staining (phospho-DIGE). This large-scale phosphoproteome analysis of early OG-signaling enabled us to determine 100 regulated phosphosites using LC-MS/MS and 46 differential spots corresponding to 34 pdhosphoproteins using phospho-DIGE. Functional classification showed that the OG-responsive phosphoproteins include kinases, phosphatases and receptor-like kinases, heat shock proteins (HSPs), reactive oxygen species (ROS) scavenging enzymes, proteins related to cellular trafficking, transport, defense and signaling as well as novel candidates for a role in immunity, for which elicitor-induced phosphorylation changes have not been shown before. A comparison with previously identified elicitor-regulated phosphosites shows only a very limited overlap, uncovering the immune-related regulation of 70 phosphorylation sites and revealing novel potential players in the regulation of elicitor-dependent immunity

    Jonestown, Mississippi has been known to be a place of poverty and in need of infrastructure. A nonprofit, Third Lens Ministries is developing housing for the people of Jonestown, Mississippi. Over the last 9 months, an architect Margy Maher, an engineer Autumn Wagner, as well as myself, a construction management major, have all collaborated in order to design, engineer and plan the perfect Dwelling for the people of Jonestown, Mississippi. While the scope of the project includes community centers and housing, our team’s scope was to focus on the residential development and site work. As the construction manager, my deliverables are the construction scope analysis, which describes each piece of the work performed, a hazard mitigation analysis showing how natural disasters, crime and insect infestations will be mitigated, a construction safety plan outlining safety concerns, preventative maintenance and procedures, a storm water pollution prevention plan outlining how large rainfall will be handled and maintained on site, a site logistics plan showing phasing, material laydown, trailers, path of travel and phasing, an estimate broken into subcategories based on CSI formatting and finally a schedule describing each phase of the project beginning with utilities, residential structures and then site work

    Changes in the microsomal proteome of tomato fruit during ripening

    The variations in the membrane proteome of tomato fruit pericarp during ripening have been investigated by mass spectrometry-based label-free proteomics. Mature green (MG30) and red ripe (R45) stages were chosen because they are pivotal in the ripening process: MG30 corresponds to the end of cellular expansion, when fruit growth has stopped and fruit starts ripening, whereas R45 corresponds to the mature fruit. Protein patterns were markedly different: among the 1315 proteins identified with at least two unique peptides, 145 significantly varied in abundance in the process of fruit ripening. The subcellular and biochemical fractionation resulted in GO term enrichment for organelle proteins in our dataset, and allowed the detection of low-abundance proteins that were not detected in previous proteomic studies on tomato fruits. Functional annotation showed that the largest proportion of identified proteins were involved in cell wall metabolism, vesicle-mediated transport, hormone biosynthesis, secondary metabolism, lipid metabolism, protein synthesis and degradation, carbohydrate metabolic processes, signalling and response to stress

    The results of a diagnostic survey on the materials of representative Art Nouveau buildings in Italy, Portugal and Poland are here presented and compared, as a contribution to their understanding and, hence, to support compatible restoration. In particular, the facade decorations were investigated for the appraisal of their materials and technologies, often neglected in current maintenance/restoration works and so cancelled, leading to a severe loss in architectural image. The ongoing diagnostic campaign, in collaboration among different universities, is aimed to set up a database on materials and technologies of Art Nouveau facade decorations at a European scale, as a technical-scientific background for the highlighting of preservation guidelines

    O programa nacional de fortalecimento da agricultura familiar no Brasil: uma análise sobre a distribuição regional e setorial dos recursos.

    No Brasil, as políticas públicas para o espaço rural sempre tenderam a priorizar a agricultura patronal, em detrimento dos agricultores familiares. Todavia, os estudos realizados pelos órgãos FAO - INCRA deram subsídio para a criação do Programa Nacional de Fortalecimento da Agricultura Familiar (PRONAF), resultando em um novo direcionamento dos investimentos públicos, os quais passaram a contemplar o segmento dos agricultores familiares. Entende-se o PRONAF como uma política não-compensatória, que, apesar de seus problemas, tem contribuído de fato para mudanças e melhorias no espaço agrário brasileiro. Desde sua criação no final da década de 1990, o PRONAF passou por várias mudanças em sua estrutura administrativa e operacional, a fim de alcançar seus objetivos e adequar-se face a complexa realidade social agrária brasileira. Sendo assim, o presente estudo visa discutir as ações do Estado por meio desse Programa, a partir de suas linhas de atuação, bem como analisar a distribuição de suas concessões de crédito regional e setorialmente. Assim, os procedimentos metodológicos utilizados para a realização deste trabalho compreendem pesquisa bibliográfica e documental, além de pesquisa em fontes secundárias, no intuito de obter dados e informações relevantes para a análise das relações sociais estabelecidas em meio a esse processo de concretização e espacialização desse Programa. Dentre as implicações do PRONAF pode-se notar em âmbito nacional, uma diminuição da disparidade regional brasileira, bem como a preocupação que o Programa tem demonstrado com os aspectos socioculturais locais e regionais, como forma de garantir que seus investimentos perpassem a dimensão econômica, mas valorize outras dimensões, a exemplo dos elementos culturais

    Human Perception in Natural Language Generation

    We ask subjects whether they perceive as human-produced a bunch of texts, some of which are actually human-written, while others are automatically generated. We use this data to fine-tune a GPT-2 model to push it to generate more human-like texts, and observe that this fine-tuned model produces texts that are indeed perceived more human-like than the original model. Contextually, we show that our automatic evaluation strategy well correlates with human judgements. We also run a linguistic analysis to unveil the characteristics of human- vs machine-perceived language
