    Polyhexamethylene Biguanide and Nadifloxacin Self-Assembled Nanoparticles: Antimicrobial Effects against Intracellular Methicillin-Resistant Staphylococcus aureus

    The treatment of skin and soft tissue infections caused by methicillin-resistant Staphylococcus aureus (MRSA) remains a challenge, partly due to localization of the bacteria inside the host’s cells, where antimicrobial penetration and efficacy is limited. We formulated the cationic polymer polyhexamethylene biguanide (PHMB) with the topical antibiotic nadifloxacin and tested the activities against intracellular MRSA in infected keratinocytes. The PHMB/nadifloxacin nanoparticles displayed a size of 291.3 ± 89.6 nm, polydispersity index of 0.35 ± 0.04, zeta potential of +20.2 ± 4.8 mV, and drug encapsulation efficiency of 58.25 ± 3.4%. The nanoparticles killed intracellular MRSA, and relative to free polymer or drugs used separately or together, the nanoparticles displayed reduced toxicity and improved host cell recovery. Together, these findings show that PHMB/nadifloxacin nanoparticles are effective against intracellular bacteria and could be further developed for the treatment of skin and soft tissue infections

    Dislocation Dynamics in an Anisotropic Stripe Pattern

    The dynamics of dislocations confined to grain boundaries in a striped system are studied using electroconvection in the nematic liquid crystal N4. In electroconvection, a striped pattern of convection rolls forms for sufficiently high driving voltages. We consider the case of a rapid change in the voltage that takes the system from a uniform state to a state consisting of striped domains with two different wavevectors. The domains are separated by domain walls along one axis and a grain boundary of dislocations in the perpendicular direction. The pattern evolves through dislocation motion parallel to the domain walls. We report on features of the dislocation dynamics. The kinetics of the domain motion are quantified using three measures: dislocation density, average domain wall length, and the total domain wall length per area. All three quantities exhibit behavior consistent with power law evolution in time, with the defect density decaying as t−1/3t^{-1/3}, the average domain wall length growing as t1/3t^{1/3}, and the total domain wall length decaying as t−1/5t^{-1/5}.     Recent studies directed to the design of compounds targeting the bc(1) protein complex of Plasmodium falciparum, the parasite responsible for most lethal cases of malaria, identified quinolones (4-oxo-quinolines) with low nanomolar inhibitory activity against both the enzyme and infected erythrocytes. The 4-oxo-quinoline 3-ester chemotype emerged as a possible source of potent bc(1) inhibitors, prompting us to expand the library of available analogs for SAR studies and subsequent lead optimization. We now report the synthesis and structural characterization of unexpected ethyl 6-methyl-7-iodo-4-(3-iodo-4-methylphenoxy)quinoline-3-carboxylate, a 4-aryloxy-quinoline 3-ester formed during attempted preparation of 6-methyl-7-iodo-4-oxo-quinoline-3-carboxylate (4-oxo-quinoline 3-ester). We propose that the 4-aryloxy-quinoline 3-ester derives from 6-methyl-7-iodo-4-hydroxy-quinoline-3-carboxylate (4-hydroxy-quinoline 3-ester), the enol form of 6-methyl-7-iodo-4-oxo-quinoline-3-carboxylate. Formation of the 4-aryloxy-quinoline 3-ester confirms the impact of quinolone/hydroxyquinoline tautomerism, both on the efficiency of synthetic routes to quinolones and on pharmacologic profiles. Tautomers exhibit different cLogP values and interact differently with the enzyme active site.     Cervical nodular fasciitis in a 10-year-old girl: a case report of a rare condition

    Nodular fasciitis (NF) is a benign fibroblastic proliferation that usually involves  superficial fascia in the subcutaneous tissue or muscle. The rapid growth, high  cellularity and mitotic activity can simulate sarcoma. Thus, as clinical and morphologic characteristics are similar to those of malignant tumours, this entity is often  misdiagnosed, with a few cases described in the literature. We present a case of nodular fasciitis involving the retropharynx and hypopharynx of a 10-year-old girl and discuss the importance of considering this rare diagnosis in different soft tissue masses.Keywords: excisional biopsy, histology, nodular fasciiti

    Turbulent fluxes in atmospheric boundary layer of a semi-arid region of N-E Brazil

    The preliminary results of the experiment “Experimento de Microfisica de Nuvens-EmfiN” (Experiment of microphysics of clouds) conducted by Universidade Estadual de Ceara-UECE at Fortaleza, a semi-arid region of N-E Brazil, are presented. The mean kinematic fluxes of sensible heat and water vapor of the surface boundary layer are estimated by the thermodynamic energy and water vapor conservation equations; and by the Monin-Obukhov similarity theory. The results of the two methods are in good agreement. It is shown that in the absence of sophisticated fast-response turbulence instrumentation and wind data the conservations equations methods are better option for estimation of heat and water vapor fluxes. Further they are useful to study the turbulent fluxes in inhomogeneous condition in time like early morning and late evening boundary layer transitions

    The preliminary results of an analysis based on the experiment “Experimento de Microfisica de Nuvens—EmfiN” (Experiment of Microphysics of Clouds), conducted by Universidade Estadual de Ceara—UECE at Fortaleza (3.77 S and 38.60 W), a semi-arid tropical region of N-E Brazil, are presented. The kinematic stresses are computed by the layer integration of the Planetary Boundary Layer(PBL) momentum equation forhor izontally homogeneous turbulence above the viscous sublayer condition and considering that the inertial acceleration terms could be neglected compared to the Coriolis force and pressure gradient force. The computed stress profiles are in good agreement with the results reported in the literature

    Extraction, quantification and degree of polymerization of yacon (Smallanthus sonchifolia) fructans

    Yacon tubers have been a distinguished alternative of fructans, drawing the attention of researchers and food industries. Since fructans are carbohydrate reserves storage can reduce their contents. Additionally, the type of extraction used can provide a higher yield of fructans. Therefore, it was necessary to study yacon storage and its influence on the extraction and quantification of fructans. Thus, the objective of this study was to evaluate three fructan extractions (water 75°C, water 95°C, ethanol 90°C) in yacon with 3 sizes (large, medium, small), stored for 20 days, at room temperature as well as to compare two quantification techniques. The three extractions can be used when fructans are quantified by high performance liquid chromatography (HPLC). For quantification by spectrometry, the best extraction method was ethanol at 90°C. Medium and small-sized tubers presented the highest contents of fructans that large tubers, and storage negatively influenced these contents. Fructan quantification by HPLC was higher than the spectrophotometric technique. All treatments showed a degree of polymerization in the range from 3 to 7, allowing numerous technological applications for fructans present in yacon.Key words: Fructooligosaccharides, storage, tuber size

    Bioconversion of potato-processing wastes into an industrially-important chemical lactic acid

    Lactic acid (LA) is an important biomolecule applied in food, pharmaceutical and chemical areas, mainly to produce biodegradable polymers, such as poly–lactic acid (PLA). In this work, an efficient fermentative process for LA production was developed using potato processing waste (PPW) hydrolysate with Lactobacillus pentosus. After optimization and kinetics studies, LA production reached 150 g/L with a productivity of 1.6 g/L.h in Erlenmeyer flasks. LA production was also conducted in STR where 110 g/L were reached with a productivity of 2.4 g/L.h. LA recovery consisted of a clarification step, with powdered activated carbon, with further precipi- tation at low temperature and acidification of calcium lactate for conversion to LA. The process was effective for contaminants’ removal and clarification, and LA concentration to 416 g/L. Good perspectives for LA production, recovery and clarification were observed. Future studies will be carried out for LA purification and polymeri- zation for PLA synthesis
