    Use of Urban Tree Canopy Assessments by Localities in the Chesapeake Bay Watershed

    Urban tree canopy (UTC) in the Chesapeake Bay watershed (CBW) provides numerous environmental, economic, and societal benefits. UTC assessments use remote sensing technology to deliver a comprehensive spatial snapshot of a locality’s existing UTC. Because UTC assessments delineate the extent and location of tree canopy cover in the context of other land covers (including plantable space), they are important for establishing tree canopy goals, creating and implementing strategies to achieve those goals, and monitoring progress. Over the past decade, UTC assessments have been completed for numerous localities in the CBW as a result of the Chesapeake Bay Program identifying UTC as a key strategy for Bay restoration. Our research investigated the prevalence of UTC assessments within the CBW and studied how localities are using them. We conducted two surveys: 1) a pilot survey of Virginia localities that received UTC assessments as part of the Virginia UTC project; and 2) a comprehensive survey of all 101 localities in the CBW with populations over 2,500 for which a UTC assessment existed as of May 2013. Surprisingly, 33% of localities in the CBW reported being unaware that a UTC assessment had been performed for their jurisdiction. In general, counties and cities were more likely to be aware of the assessments than were towns (or their jurisdictional equivalent). Most localities that were aware of their assessment were using it in some manner for urban forest planning and management; however, the most frequent activities were also the most basic uses, including: educating officials or citizens about the importance of tree canopy (57%), providing a baseline for evaluating progress toward UTC goals (49%), creating a locality-wide tree canopy goal (47%), planning and prioritizing tree plantings (45%), and informing larger initiatives (43%). All other uses of the assessments (i.e., specialized uses) were reported by 33% or fewer of the CBW localities. Our findings point to the need for outreach to local governments about UTC assessments and their potential uses, particularly in light of increasing emphasis in the CBW on managing urban forests and optimizing UTC as a Bay restoration strategy

    Pine sawdust biochar as a potential amendment for establishing trees in Appalachian mine spoils

    Early growth and survival of tree seedlings is often poor on reclaimed coal surface mines in Appalachia. Biochar produced in bioenergy generation has potential for use as an amendment to improve seedling performance. Mine soil was collected from a recently reclaimed coal surface mine in Wise County, Virginia and mixed with loblolly pine (Pinus taeda L.) sawdust biochar, simulating application rates of 2.3, 11.2 and 22.5 Mg ha-1. Unplanted leaching columns and 4 L tree planting pots were filled with these biochar-soil mixtures, plus controls of pure mine soil and pure biochar. For the tree planting pots, additional pots were created where the biochar was applied as a topdressing at the same application rates as in the mixtures. One-year-old seedlings of both American sycamore (Platanus occidentalis L.) and black locust (Robinia pseudoacacia L.) were planted. Unplanted leaching columns were leached with collected rainwater for six months to simulate weathering. Trees were grown for one growing season. Black locust had higher average above-ground dry woody biomass (24.4 g) than American sycamore (17.0 g), and also higher below-ground biomass (61.0 g compared to 30.2 g). The pure biochar produced greater average below-ground biomass (99.9 g) than the pure mine soil (46.9 g). All of the biochar treatments produced greater average above-ground woody biomass (19.1 g – 33.4 g) than the pure mine soil (10.9 g). After weathering, biochar provided less available soil phosphorus, calcium and iron than the mine soil itself while increasing soil carbon and organic matter. High (22.5 Mg ha-1) biochar applications increased soil volumetric water holding capacity to 18.6% compared to 13.4% for pure mine soil. Naturally-occurring herbaceous biomass in the pots was negatively correlated with above-ground woody biomass at r = -0.483. Topdressing and full incorporation of biochar were not significantly different in their effects on biomass. Results suggest that pine biochar either broadcast at 2.3 - 22.5 Mg ha‑1, or mixed in planting holes with backfill soil, will promote faster above-ground growth and larger root systems in seedlings in mine soils. Further studies should test these methods in the field over multiple years and further refine recommendations of the rate of biochar to use and how best to apply it. New systems are being developed in Appalachia to produce biofuels and biochar from local biomass and to recycle biochar into the land base to enhance future biomass productivity.     The unexplored role of sedentary time and physical activity in glucose and lipid metabolism-related placental mRNAs in pregnant women who are obese: the DALI lifestyle randomised controlled trial

    Objective: We aimed to explore: (i) the association of sedentary time (ST) and physical activity (PA) during pregnancy with the placental expression of genes related to glucose and lipid metabolism in pregnant women who are obese; (ii) maternal metabolic factors mediating changes in these placental transcripts; and (iii) cord blood markers related to the mRNAs mediating neonatal adiposity. Design: Multicentre randomised controlled trial. Setting: Hospitals in nine European countries. Population: A cohort of 112 pregnant women with placental tissue. Methods: Both ST and moderate-to-vigorous PA (MVPA) levels were measured objectively using accelerometry at three time periods during pregnancy. Main outcome measures: Placental mRNAs (FATP2, FATP3, FABP4, GLUT1 and PPAR-γ) were measured with NanoString technology. Maternal and fetal metabolic markers and neonatal adiposity were assessed. Results: Longer periods of ST, especially in early to middle pregnancy, was associated with lower placental FATP2 and FATP3 expression (P \u3c 0.05), whereas MVPA at baseline was inversely associated with GLUT1 mRNA (P = 0.02). Although placental FATP2 and FATP3 expression were regulated by the insulin–glucose axis (P \u3c 0.05), no maternal metabolic marker mediated the association of ST/MVPA with placental mRNAs (P \u3e 0.05). Additionally, placental FATP2 expression was inversely associated with cord blood triglycerides and free fatty acids (FFAs; P \u3c 0.01). No cord blood marker mediated neonatal adiposity except for cord blood leptin, which mediated the effects of PPAR-γ on neonatal sum of skinfolds (P \u3c 0.05). Conclusions: In early to middle pregnancy, ST is associated with the expression of placental genes linked to lipid transport. PA is hardly related to transporter mRNAs. Strategies aimed at reducing sedentary behaviour during pregnancy could modulate placental gene expression, which may help to prevent unfavourable fetal and maternal pregnancy outcomes.     People of the British Isles: preliminary analysis of genotypes and surnames in a UK control population

    There is a great deal of interest in fine scale population structure in the UK, both as a signature of historical immigration events and because of the effect population structure may have on disease association studies. Although population structure appears to have a minor impact on the current generation of genome-wide association studies, it is likely to play a significant part in the next generation of studies designed to search for rare variants. A powerful way of detecting such structure is to control and document carefully the provenance of the samples involved. Here we describe the collection of a cohort of rural UK samples (The People of the British Isles), aimed at providing a well-characterised UK control population that can be used as a resource by the research community as well as providing fine scale genetic information on the British population. So far, some 4,000 samples have been collected, the majority of which fit the criteria of coming from a rural area and having all four grandparents from approximately the same area. Analysis of the first 3,865 samples that have been geocoded indicates that 75% have a mean distance between grandparental places of birth of 37.3km, and that about 70% of grandparental places of birth can be classed as rural. Preliminary genotyping of 1,057 samples demonstrates the value of these samples for investigating fine scale population structure within the UK, and shows how this can be enhanced by the use of surnames
