    An Algorithm for the Real Interval Eigenvalue Problem

    In this paper we present an algorithm for approximating the range of the real eigenvalues of interval matrices. Such matrices could be used to model real-life problems, where data sets suffer from bounded variations such as uncertainties (e.g. tolerances on parameters, measurement errors), or to study problems for given states. The algorithm that we propose is a subdivision algorithm that exploits so- phisticated techniques from interval analysis. The quality of the computed approximation, as well as the running time of the algorithm depend on a given input accuracy. We also present an efficient C++ implementation and illustrate its efficiency on various data sets. In most of the cases we manage to compute efficiently the exact boundary points (limited by floating point representation)

    A filtering method for the interval eigenvalue problem

    We consider the general problem of computing intervals that contain the real eigenvalues of interval matrices. Given an outer estimation of the real eigenvalue set of an interval matrix, we propose a filtering method that improves the estimation. Even though our method is based on an sufficient regularity condition, it is very efficient in practice, and our experimental results suggest that, in general, improves significantly the input estimation. The proposed method works for general, as well as for symmetric matrices

    How to realise a homogeneous dipolar Bose gas in the roton regime

    Homogeneous quantum gases open up new possibilities for studying many-body phenomena and have now been realised for a variety of systems. For gases with short-range interactions the way to make the cloud homogeneous is, predictably, to trap it in an ideal (homogeneous) box potential. We show that creating a close to homogeneous dipolar gas in the roton regime, when long-range interactions are important, actually requires trapping particles in soft-walled (inhomogeneous) box-like potentials. In particular, we numerically explore a dipolar gas confined in a pancake trap which is harmonic along the polarisation axis and a cylindrically symmetric power-law potential rpr^p radially. We find that intermediate pp's maximise the proportion of the sample that can be brought close to the critical density required to reach the roton regime, whereas higher pp's trigger density oscillations near the wall even when the bulk of the system is not in the roton regime. We characterise how the optimum density distribution depends on the shape of the trapping potential and find it is controlled by the trap wall steepness.Comment: 7 pages, 3 figure

    Constraints on the Geological History of the Karst System in Southern Missouri, U.S.A. Provided by Radiogenic, Cosmogenic and Physical/Chemical Characteristics of Doline Fill

    Območje Ozarkov v južnem Missouriju gradijo predvsem karbonatne, morske, platformne kamnine paleozojske starosti. Teren, ki vsebuje obširen kras, predstavlja kopno že od poznega Paleozojka. Da bi bolje razumeli geološko zgodovino tega kraškega sistema, smo raziskali stratigrafske podatke ohranjene v zapolnitvi velike vrtače pri največjem izviru v okolici. Vzorci zapolnitve iz naravnih izdankov in iz vrtine so bili analizirani s termoluminiscenco (TL) in 10Be kozmogeno metodo. Fizikalno-kemijske značilnosti zapolnitve so bile določene vizualno, z rentgensko metodo in merjenjem velikosti delcev. Podatki vrtine kažejo, da je debelina alohtonega materiala, ki zapolnjuje vrtačo 36,3 m. Ta material prekriva podorne bloke in sedimente debeline vsaj 15,6 m. Glede na teksturo, strukturo in barvo delimo material, ki zapolnjuje vrtačo na 7 con. Analize 10Be koncentracij kažejo, da material za celotni stolpec zapolnitve izvira iz rezidualnega materiala iz srednjega (Illinoian) in zgornjega Pleistocena (Wisconsian). Rentgenske analize glin kažejo, da zapolnitev vrtače vsebuje enake količine kaolinita in illita, kar je skladno z zemeljskim preperevanjem.The Ozark Plateaus region of southern Missouri is underlain by dominantly carbonate marine platform rocks of Paleozoic age. The region has been sub-aerially exposed since the late Paleozoic and is characterized by extensive karst. To better understand the geologic history of this regional karst system, we examined the stratigraphic record preserved in the fill of a large doline near the largest spring in the region. Samples of fill from natural exposures and drill core were analyzed using thermoluminescence (TL) and 10Be cosmogenic techniques, and the physical/chemical characteristics of the fill material were determined by visual inspection, X-ray analyses, and grain-size measurements. Drill-hole data indicate that the allochthonous doline fill is 36.3 m thick and rests on at least 15.6 m of cave breakdown and sediment. The doline fill is divisible into 7 zones. Analysis of 10Be concentrations suggest that the entire doline fill was derived from local residuum during the middle (Illinoian) to late Pleistocene (Wisconsinan). X-ray diffraction analyses of clays throughout the doline fill indicate that they consist of nearly equal amounts of kaolinite and illite, consistent with terrestrial weathering.

    Repeated inoculations with the lung and heartworm nematode <i>Angiostrongylus vasorum</i> result in increasing larval excretion and worm burden in the red fox (<i>Vulpes vulpes</i>)

    The French heartworm Angiostongylus vasorum is found in European red fox (Vulpes vulpes) and dog populations, where it appears to be spreading geographically. Once introduced into new areas, it establishes in local fox populations, typically to over 50% prevalence in a few years. High susceptibility and constant excretion of first stage larvae (L1) by the definitive hosts are prerequisites for sustaining high parasite biomass in a particular habitat. The present study explores the hypothesis that repeated ingestion of gastropods in nature will result in accumulation of adult worms and elevated excretion of L1 in feces. Experimentally infected foxes were subsequently inoculated via stomach tube once (9 weeks post initial inoculation) or twice (9 and 13 weeks post inoculation (wpi)) with 100 third stage A. vasorum larvae (L3) previously isolated from aquatic snails infected with L1 from a naturally infected dog. Despite large variation in fecal larval excretion for the individual animals within the groups, excretion of L1 was significantly higher in foxes twice inoculated as compared to foxes inoculated only once. With an outlier in the once inoculated group removed, excretion became significantly higher in the three times inoculated group. Establishment of adult worms varied and only a trend to higher worm burdens was found in the group of foxes inoculated three times. However, this became significant with the same single outlier removed. Overall, it appears that protective immunity to A. vasorum does not appear to occur in V. vulpes with animals exhibiting high infection intensities without obvious clinical signs. The increasing larval excretion in foxes being repeatedly exposed to A. vasorum L3 support the hypothesis that foxes under natural conditions may repeatedly ingest infected gastropods and remain a source of environmental contamination for several months, potentially contributing to the establishment of endemic foci through increasing L1 excretion.     Dual PI3K and Wnt pathway inhibition is a synergistic combination against triple negative breast cancer

    Triple negative breast cancer accounts for 15-20% of all breast cancer cases, but despite its lower incidence, contributes to a disproportionately higher rate of mortality. As there are currently no Food and Drug Administration-approved targeted agents for triple negative breast cancer, we embarked on a genomic-guided effort to identify novel targeted modalities. Analyses by our group and The Cancer Genome Atlas have identified activation of the PI3K-pathway in the majority of triple negative breast cancers. As single agent therapy is commonly subject to resistance, we investigated the use of combination therapy against compensatory pathways. Herein, we demonstrate that pan-PI3K inhibition in triple negative breast cancers results in marked activation of the Wnt-pathway. Using the combination of two inhibitors currently in clinical trial as single agents, buparlisib(pan-PI3K) and WNT974(WNT-pathway), we demonstrate significant in vitro and in vivo synergy against triple negative breast cancer cell lines and xenografts. Taken together, these observations provide a strong rationale for testing dual targeting of the PI3K and WNT-pathways in clinical trials

    In a previous study, it was shown that the lipid fraction, which is occasionally observed in red blood cell hemolysates, represents cholesterol (Ch) associated with phospholipid firmly bound to haemoglobin (termed Hb-Ch). The current study was conducted to investigate whether Hb-Ch could affect the primary anti-oxidant enzyme defence system in human erythrocytes. Sixty healthy volunteers were used for the current study. Group 1 consisted of 28 subjects without or with a low level of Hb-Ch. Group 2 comprised 32 subjects with a considerably higher level of Hb-Ch. The activities of erythrocyte superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase, as well as the content of methaemoglobin (metHb) were measured in both groups. The results indicated that the amount of Hb-Ch neither influenced the activities of the erythrocyte anti-oxidant enzymes nor altered the level of metHb. However, a higher amount of Hb-Ch changed the correlations in the part of the anti-oxidant defence system relating to glutathione, suggesting increased peroxidative pressure from plasma lipids. Group 2 also had significantly increased concentrations of total plasma Ch and triglycerides. Together, these facts are strong indications that the anti-oxidant defence system in human erythrocytes finely retunes its composition according to plasma oxidative demands.U prethodnom radu pokazano je da lipidna frakcija koja se javlja u hemolizatu zdravih ljudi predstavlja holesterol (asosovan sa fosfolipidima) čvrsto vezan za hemoglobin (Hb-Ch). U ovom radu ispitivan je uticaj Hb-Ch na anti-oksidativni enzimski sistem u humanim eritrocitima. Određena je aktivnost superoksid-dizmutaze, katalaze, glutation-peroksidaze i glutation-reduktaze, kao i sadržaj met-hemoglobina (metHb) u eritrocitima 60 ljudi, podeljenih u dve grupe na osnovu količine Hb-Ch. Rezultati pokazuju da količina prisutnog Hb-Ch ne menja aktivnost merenih enzima, niti nivo metHb. Međutim, u grupi ispitanika sa povećanim sadržajem Hb-Ch zapažene su korelativne promene u delu anti-oksidativnog enzimskog sistema povezanog sa glutationom. U istoj grupi detektovane su i veće koncentracije ukupnog holesterola i triglicerida u plazmi, što zajedno ukazuje na povećani peroksidativni pritisak iz plazme. Ovi rezultati ukazuju da odbrambeni anti-oksidativni enzimski sistem u humanim eritrocitima prilagođava svoju organizaciju prema zahtevima iz svog okruženja.

    Common molecular mechanism of the hepatic lesion and the cardiac parasympathetic regulation in chronic hepatitis C infection: a critical role for the muscarinic receptor type 3

    Background: The pathophysiological overlapping between Sjorgens Syndrome (SS) and HCV, presence of anti-muscarinic receptor type 3 (M3R) antibodies in SS, the role that M3R plays in the regulation of the heart rate, has led to the assumption that cardiovagal dysfunction in HCV patients is caused by anti-M3R antibodies elicited by HCV proteins or by their direct interaction with M3R. Results: To identify HCV protein which possibly is crossreactive with M3R or which binds to this receptor, we performed the Informational Spectrum Method (ISM) analysis of the HCV proteome. This analysis revealed that NS5A protein represents the most probable interactor of M3R or that this viral protein could elicit antibodies which modulate function of this receptor. Further detailed structure/function analysis of NS5A and M3R performed by the ISM method extended with other Digital Signal processing (DSP) approaches revealed domains of these proteins which participate in their crossreactivity or in their direct interaction, representing promising diagnostic and therapeutic targets. Conclusions: Application of the ISM with other compatible bioinformatics methods offers new perspectives for identifying diagnostic and therapeutic targets for complicated forms of HCV and other viral infections. We show how the electron-ion interaction potential (EIIP) amino-acid scale used in the ISM combined with a robust, high performance hydrophobicity scale can provide new insights for understanding protein structure/function and protein-protein interactions
