    Status of the Combined Cycle Engine Rig

    Status for the past year is provided of the turbine-based Combined-Cycle Engine (CCE) Rig for the hypersonic project. As part of the first stage propulsion of a two-stage-to-orbit vehicle concept, this engine rig is designed with a common inlet that supplies flow to a turbine engine and a dual-mode ramjet / scramjet engine in an over/under configuration. At Mach 4 the inlet has variable geometry to switch the airflow from the turbine to the ramjet / scramjet engine. This process is known as inlet mode-transition. In addition to investigating inlet aspects of mode transition, the rig will allow testing of turbine and scramjet systems later in the test series. Fully closing the splitter cowl "cocoons" the turbine engine and increases airflow to the scramjet duct. The CCE Rig will be a testbed to investigate integrated propulsion system and controls technology objectives. Four phases of testing are planned to 1) characterize the dual inlet database, 2) collect inlet dynamics using system identification techniques, 3) implement an inlet control to demonstrate mode-transition scenarios and 4) demonstrate integrated inlet/turbine engine operation through mode-transition. Status of the test planning and preparation activities is summarized with background on the inlet design and small-scale testing, analytical CFD predictions and some details of the large-scale hardware. The final stages of fabrication are underway

    TBCC Inlet Experiments and Analysis

    A research plan is being implemented at NASA to investigate inlet mode transition for turbine-based combined-cycle (TBCC) propulsion for the hypersonic community. Unresolved issues have remained on how to design an inlet system to supply both a turbine engine and a ram/scramjet flowpath that operate with both high performance and stability. The current plan is aimed at characterizing the design, performance and operability of TBCC inlets through a series of experiments and analyses. A TBCC inlet has been designed that is capable of high performance (near MIL-E-5008B recovery) with smooth transitioning characteristics. Traditional design techniques were used in an innovative approach to balance the aerodynamic and mechanical constraints to create a new TBCC inlet concept. The inlet was designed for top-end Mach 7 scramjet speeds with an over/under turbine that becomes cocooned beyond its Mach 4 peak design point. Conceptually, this propulsion system was picked to meet the needs of the first stage of a two-stage to orbit vehicle. A series of increasing fidelity CFD-based tools are being used throughout this effort. A small-scale inlet experiment is on-going in the GRC 1'x1' Supersonic Wind Tunnel (SWT). Initial results from both the CFD analyses and test are discussed showing that high performance and smooth mode transitions are possible. The effort validates the design and is contributing to a large-scale inlet/propulsion test being planned for the GRC 10' x10' SWT. This large-scale effort provide the basis for a Combined Cycle Engine Testbed, (CCET), that will be able to address integrated propulsion system and controls objectives

    Superconductivity in AuNiGe Ohmic contacts to a GaAs-based high mobility two-dimensional electron gas

    To cool a high mobility two-dimensional electron gas (2DEG) at a GaAs-AlGaAs heterojunction to milliKelvin temperatures, we have fabricated low resistance Ohmic contacts based on alloys of Au, Ni, and Ge. The Ohmic contacts have a typical contact resistance of R C ≈ 0.8 ω at 4.2 K, which drops to 0.2 ω below 0.9 K. Scanning electron microscope images establish that the contacts have the same inhomogeneous microstructure that has been observed in previous studies. Measurements of the contact resistance R C, the four-terminal resistance along the top of a single contact, and the vertical resistance RV all show that there is a superconductor in the Ohmic contact, which can be turned completely normal with a magnetic field of 0.15 T. We briefly discuss how this superconductivity may be affecting the electrical transport measurements of 2DEGs, especially how it may hinder the cooling of electrons in a 2DEG below 0.1 K

    Utilisation of an operative difficulty grading scale for laparoscopic cholecystectomy

    Background A reliable system for grading operative difficulty of laparoscopic cholecystectomy would standardise description of findings and reporting of outcomes. The aim of this study was to validate a difficulty grading system (Nassar scale), testing its applicability and consistency in two large prospective datasets. Methods Patient and disease-related variables and 30-day outcomes were identified in two prospective cholecystectomy databases: the multi-centre prospective cohort of 8820 patients from the recent CholeS Study and the single-surgeon series containing 4089 patients. Operative data and patient outcomes were correlated with Nassar operative difficultly scale, using Kendall’s tau for dichotomous variables, or Jonckheere–Terpstra tests for continuous variables. A ROC curve analysis was performed, to quantify the predictive accuracy of the scale for each outcome, with continuous outcomes dichotomised, prior to analysis. Results A higher operative difficulty grade was consistently associated with worse outcomes for the patients in both the reference and CholeS cohorts. The median length of stay increased from 0 to 4 days, and the 30-day complication rate from 7.6 to 24.4% as the difficulty grade increased from 1 to 4/5 (both p < 0.001). In the CholeS cohort, a higher difficulty grade was found to be most strongly associated with conversion to open and 30-day mortality (AUROC = 0.903, 0.822, respectively). On multivariable analysis, the Nassar operative difficultly scale was found to be a significant independent predictor of operative duration, conversion to open surgery, 30-day complications and 30-day reintervention (all p < 0.001). Conclusion We have shown that an operative difficulty scale can standardise the description of operative findings by multiple grades of surgeons to facilitate audit, training assessment and research. It provides a tool for reporting operative findings, disease severity and technical difficulty and can be utilised in future research to reliably compare outcomes according to case mix and intra-operative difficulty

    Consensus on exercise reporting template (Cert): Modified delphi study

Exercise interventions are often incompletely described in reports of clinical trials, hampering evaluation of results and replication and implementation into practice. Objective. The aim of this study was to develop a standardized method for reporting exercise programs in clinical trials: the Consensus on Exercise Reporting Template (CERT). Design and Methods. Using the EQUATOR Network’s methodological framework, 137 exercise experts were invited to participate in a Delphi consensus study. A list of 41 items was identified from a meta-epidemiologic study of 73 systematic reviews of exercise. For each item, participants indicated agreement on an 11-point rating scale. Consensus for item inclusion was defined a priori as greater than 70% agreement of respondents rating an item 7 or above. Three sequential rounds of anonymous online questionnaires and a Delphi workshop were used. Results. There were 57 (response rate=42%), 54 (response rate=95%), and 49 (response rate=91%) respondents to rounds 1 through 3, respectively, from 11 countries and a range of disciplines. In round 1, 2 items were excluded; 24 items reached consensus for inclusion (8 items accepted in original format), and 16 items were revised in response to participant suggestions. Of 14 items in round 2, 3 were excluded, 11 reached consensus for inclusion (4 items accepted in original format), and 7 were reworded. Sixteen items were included in round 3, and all items reached greater than 70% consensus for inclusion. Limitations. The views of included Delphi panelists may differ from those of experts who declined participation and may not fully represent the views of all exercise experts. Conclusions. The CERT, a 16-item checklist developed by an international panel of exercise experts, is designed to improve the reporting of exercise programs in all evaluative study designs and contains 7 categories: materials, provider, delivery, location, dosage, tailoring, and compliance. The CERT will encourage transparency, improve trial interpretation and replication, and facilitate implementation of effective exercise interventions into practice

    Calpain-mediated vimentin cleavage occurs upstream of MT1-MMP membrane translocation to facilitate endothelial sprout initiation

    Endothelial cells normally line the vasculature and remain quiescent. However, these cells can be rapidly stimulated to undergo morphogenesis and initiate new blood vessel formation given the proper cues. This study reports a new mechanism for initiating angiogenic sprout formation that involves vimentin, the major intermediate filament protein in endothelial cells. Initial studies confirmed vimentin was required for sphingosine 1-phosphate (S1P)- and growth factor (GF)-induced endothelial cell invasion, and vimentin was cleaved by calpains during invasion. Calpains were predominantly activated by GF and were required for sprout initiation. Because others have reported membrane type 1-matrix metalloproteinase (MT1-MMP) is required for endothelial sprouting responses, we tested whether vimentin and calpain acted upstream of MT1-MMP. Both calpain and vimentin were required for successful MT1-MMP membrane translocation, which was stimulated by S1P. In addition, vimentin complexed with MT1-MMP in a manner that required both the cytoplasmic domain of MT1-MMP and calpain activation, which increased the soluble pool of vimentin in endothelial cells. Altogether, these data indicate that pro-angiogenic signals converge to activate calpain-dependent vimentin cleavage and increase vimentin solubility, which act upstream to facilitate MT1-MMP membrane translocation, resulting in successful endothelial sprout formation in three-dimensional collagen matrices. These findings help explain why S1P and GF synergize to stimulate robust sprouting in 3D collagen matrices
