    Aircraft dynamic loads and vibrations resulting from landing impact and from runway and taxiway unevenness are recognized as significant factors in causing fatigue damage, dynamic stress on the airframe, crew and passenger discomfort, and reduction of the pilot's ability to control the aircraft during ground operations. One potential method for improving operational characteristics of aircraft on the ground is the application of active control technology to the landing gears to reduce ground loads applied to the airframe. An experimental investigation was conducted on series-hydraulic active control nose gear. The experiments involved testing the gear in both passive and active control modes. Results of this investigation show that a series-hydraulic active control gear is feasible and that such a gear is effective in reducing the loads transmitted by the gear to the airframe during ground operations

    Magnetospheric eclipses in the double pulsar system J0737-3039

    We argue that eclipses of radio emission from the millisecond pulsar A in the double pulsar system J0737-3039 are due to synchrotron absorption by plasma in the closed field line region of the magnetosphere of its normal pulsar companion B. A's radio beam only illuminates B's magnetosphere for about 10 minutes surrounding the time of eclipse. During this time it heats particles at r\gtrsim 10^9 cm to relativistic energies and enables extra plasma to be trapped by magnetic mirroring. An enhancement of the plasma density by a factor \sim 10^2 is required to match the duration and optical depth of the observed eclipses. The extra plasma might be supplied by a source near B through B\gamma pair creation by energetic photons produced in B's outer gap. Excitation of pairs' gyrational motions by cyclotron absorption of A's radio beam can result in their becoming trapped between conjugate mirror points in B's magnetosphere. Because the trapping efficiency decreases with increasing optical depth, the plasma density enhancement saturates even under steady state illumination. The result is an eclipse with finite, frequency dependent, optical depth. After illumination by A's radio beam ceases, the trapped particles cool and are lost. The entire cycle repeats every orbital period. We speculate that the asymmetries between eclipse ingress and egress result in part from the magnetosphere's evolution toward a steady state when illuminated by A's radio beam.     Resonant Cyclotron Radiation Transfer Model Fits to Spectra from Gamma-Ray Burst GRB870303

    We demonstrate that models of resonant cyclotron radiation transfer in a strong field (i.e. cyclotron scattering) can account for spectral lines seen at two epochs, denoted S1 and S2, in the Ginga data for GRB870303. Using a generalized version of the Monte Carlo code of Wang et al. (1988,1989b), we model line formation by injecting continuum photons into a static plane-parallel slab of electrons threaded by a strong neutron star magnetic field (~ 10^12 G) which may be oriented at an arbitrary angle relative to the slab normal. We examine two source geometries, which we denote "1-0" and "1-1," with the numbers representing the relative electron column densities above and below the continuum photon source plane. We compare azimuthally symmetric models, i.e. models in which the magnetic field is parallel to the slab normal, with models having more general magnetic field orientations. If the bursting source has a simple dipole field, these two model classes represent line formation at the magnetic pole, or elsewhere on the stellar surface. We find that the data of S1 and S2, considered individually, are consistent with both geometries, and with all magnetic field orientations, with the exception that the S1 data clearly favor line formation away from a polar cap in the 1-1 geometry, with the best-fit model placing the line-forming region at the magnetic equator. Within both geometries, fits to the combined (S1+S2) data marginally favor models which feature equatorial line formation, and in which the observer's orientation with respect to the slab changes between the two epochs.     Very-high-energy gamma radiation associated with the unshocked wind of the Crab pulsar

    We show that the relativistic wind in the Crab pulsar, which is commonly thought to be invisible in the region upstream of the termination shock at R < 0.1 pc, in fact could be directly observed through its inverse Compton gamm-ray emission. The search for such specific component of radiation in the gamma-ray spectrum of the Crab can provide unique information about the unshocked pulsar wind that is not accessible at other wavelengths.Comment: 11 pages, 11 figures, to appear in one of the April issues of MNRA

    TATA-Binding Protein Recognition and Bending of a Consensus Promoter Are Protein Species Dependent

    The structure and behavior of full-length human TBP binding the adenovirus major late promoter (AdMLP) have been characterized using biophysical methods. The human protein induces a 97° bend in DNAAdMLP. The high-resolution functional data provide a quantitative energetic and kinetic description of the partial reaction sequence as native human TBP binds rapidly to a consensus promoter with high affinity. The reaction proceeds with successive formation of three bound species, all having strongly bent DNA, with the concurrence of binding and bending demonstrated by both fluorescence and anisotropy stopped flow. These results establish the protein species dependence of the TBP−DNAAdMLP structure and recognition mechanism. Additionally, the strong correlation between the DNA bend angle and transcription efficiency demonstrated previously for yeast TBP is shown to extend to human TBP. The heterologous NH2-terminal domains are the apparent source of the species-specific differences. Together with previous studies the present work establishes that TBPwt−DNATATA function and structure depend both on the TATA box sequence and on the TBP species

    Exogenous 17-β Estradiol Administration Blunts Progression of Established Angiotensin II-Induced Abdominal Aortic Aneurysms in Female Ovariectomized Mice

    BACKGROUND: Abdominal aortic aneurysms (AAAs) occur predominately in males. However, AAAs in females have rapid growth rates and rupture at smaller sizes. Mechanisms contributing to AAA progression in females are undefined. We defined effects of ovariectomy, with and without 17-β estradiol (E2), on progression of established angiotensin II (AngII)-induced AAAs in female mice. METHODS: We used neonatal testosterone exposures at 1 day of age to promote susceptibility to AngII-induced AAAs in adult female Ldlr-/- mice. Females were infused with AngII for 28 days to induce AAAs, and then stratified into groups that were sham, ovariectomized (Ovx, vehicle), or Ovx with E2 administration for 2 months of continued AngII infusions. Aortic lumen diameters were quantified by ultrasound and analyzed by linear mixed model, and maximal AAA diameters were analyzed by one-way ANOVA. Atherosclerosis was quantified en face in the aortic arch. AAA tissue sections were analyzed for cellular composition. We quantified effects of E2 on abdominal aortic smooth muscle cell (SMC) growth, α-actin and transforming growth factor-beta (TGF-β) production, and wound healing. RESULTS: Serum E2 concentrations were increased significantly by E2. Aortic lumen diameters increased over time in sham-operated and Ovx (vehicle) females, but not in Ovx females administered E2. At day 70, E2 administration decreased significantly aortic lumen diameters compared to Ovx vehicle and sham-operated females. Compared to Ovx females (vehicle), maximal AAA diameters were reduced significantly by E2. AAA tissue sections from Ovx females administered E2 exhibited significant increases in α-actin and decreases in neutrophils compared to Ovx females administered vehicle. In abdominal aortic SMCs, E2 resulted in a concentration-dependent increase in α-actin, elevated TGF-β, and more rapid wound healing. E2 administration to Ovx females also significantly reduced atherosclerotic lesions compared to sham-operated females. This effect was accompanied by significant reductions in serum cholesterol concentrations. CONCLUSIONS: E2 administration to Ovx females abolished progressive growth and decreased severity of AngII-induced AAAs. These effects were accompanied by increased SMC α-actin, elevated TGF-β, and reduced neutrophils. Similarly, E2 administration reduced AngII-induced atherosclerosis. These results suggest that loss of E2 in post-menopausal females may contribute to progressive growth of AAAs

    Chemical characterization of fine particulate matter emitted by peat fires in Central Kalimantan, Indonesia, during the 2015 El Niño

    Fine particulate matter (PM2:5) was collected in situ from peat smoke during the 2015 El Niño peat fire episode in Central Kalimantan, Indonesia. Twenty-one PM samples were collected from 18 peat fire plumes that were primarily smoldering with modified combustion efficiency (MCE) values of 0.725-0.833. PM emissions were determined and chemically characterized for elemental carbon (EC), organic carbon (OC), water-soluble OC, water-soluble ions, metals, and organic species. Fuel-based PM2:5 mass emission factors (EFs) ranged from 6.0 to 29.6 g kg1 with an average of 17:36:0 g kg1. EC was detected only in 15 plumes and comprised 1% of PM mass. Together, OC (72 %), EC (1 %), water-soluble ions (1 %), and metal oxides (0.1 %) comprised 7411% of gravimetrically measured PM mass. Assuming that the remaining mass is due to elements that form organic matter (OM; i.e., elements O, H, N) an OM-to-OC conversion factor of 1.26 was estimated by linear regression. Overall, chemical speciation revealed the following characteristics of peat-burning emissions: high OC mass fractions (72%), primarily water-insoluble OC (8411 %C), low EC mass fractions (1 %), vanillic to syringic acid ratios of 1.9, and relatively high n-alkane contributions to OC (6.2 %C) with a carbon preference index of 1.2-1.6. Comparison to laboratory studies of peat combustion revealed similarities in the relative composition of PM but greater differences in the absolute EF values. The EFs developed herein, combined with estimates of the mass of peat burned, are used to estimate that 3.2-11 Tg of PM2:5 was emitted to atmosphere during the 2015 El Niño peatland fire event in Indonesia. Combined with gas-phase measurements of CO2, CO, CH4, and volatile organic carbon from Stockwell et al. (2016), it is determined that OC and EC accounted for 2.1 and 0.04% of total carbon emissions, respectively. These in situ EFs can be used to improve the accuracy of the representation of Indonesian peat burning in emission inventories and receptor-based models

    Life in Hot Carbon Monoxide: The Complete Genome Sequence of Carboxydothermus hydrogenoformans Z-2901

    We report here the sequencing and analysis of the genome of the thermophilic bacterium Carboxydothermus hydrogenoformans Z-2901. This species is a model for studies of hydrogenogens, which are diverse bacteria and archaea that grow anaerobically utilizing carbon monoxide (CO) as their sole carbon source and water as an electron acceptor, producing carbon dioxide and hydrogen as waste products. Organisms that make use of CO do so through carbon monoxide dehydrogenase complexes. Remarkably, analysis of the genome of C. hydrogenoformans reveals the presence of at least five highly differentiated anaerobic carbon monoxide dehydrogenase complexes, which may in part explain how this species is able to grow so much more rapidly on CO than many other species. Analysis of the genome also has provided many general insights into the metabolism of this organism which should make it easier to use it as a source of biologically produced hydrogen gas. One surprising finding is the presence of many genes previously found only in sporulating species in the Firmicutes Phylum. Although this species is also a Firmicutes, it was not known to sporulate previously. Here we show that it does sporulate and because it is missing many of the genes involved in sporulation in other species, this organism may serve as a “minimal” model for sporulation studies. In addition, using phylogenetic profile analysis, we have identified many uncharacterized gene families found in all known sporulating Firmicutes, but not in any non-sporulating bacteria, including a sigma factor not known to be involved in sporulation previously

Delivering an effective drug load to the posterior section of the ocular tissues, while using a non-invasive technique, has always been a challenge. In this regard, the goal of the present study was to develop sustained release triamcinolone acetonide (TA) loaded polymeric matrix films for ocular delivery. The TA-films were prepared in two different polymer matrices, with drug loadings of 10% and 20% w/w, and they were evaluated for ocular distribution in vivo in a conscious rabbit model. A 4% w/v TA suspension (TA-C) was used as a control for in vitro and in vivo studies. The TA-films, prepared with melt-cast technology, used polyethylene oxide (PEO) and Soluplus® as the polymer matrix. The films were evaluated with respect to assay, content uniformity, excipient interaction, and permeability across isolated rabbit sclera. The distribution of TA in the ocular tissues, post topical administration, was determined in New Zealand male albino rabbits as a function of dose, and was compared against TA-C. The assay of the 10% and 20% w/w film was in the range from 70–79% and 92–94% for the Soluplus® and PEO films, respectively, and content uniformity was in the range of 95–103% for both the films. The assay of the TA from Soluplus® films was less compared with the PEO films and showed an interaction with TA, as revealed by Differential Scanning Calorimetry (DSC). Hence, Soluplus® films were not selected for further studies. No interaction was observed between the drug and PEO polymer matrix. The enhancement of trans-scleral flux and permeability of TA was about 1.16 and 1.33-folds, respectively, from the 10% w/w PEO and 3.5 and 2.12-folds, respectively, from the 20% w/w PEO films, as compared with TA-C formulations. The in vivo studies demonstrate that significantly higher TA levels were observed in the anterior and posterior segments of the eye at the end of 6h with the PEO films. Therefore, the PEO based polymeric films were able to deliver TA into the back of the eye efficiently and for prolonged periods. View Full-Tex
