    Study of the press forming mechanism of a thermoforming machine

    The paper presents kinematic and dynamic investigations of the main press forming mechanism of a thermoforming machine. A multibody analysis of this press forming mechanism, which lifts and rotates a press bed, was carried out. Press bed lifting, which is necessary to form the component, is performed by means of a first rod and toggle mechanism. Press bed rotation to eject the formed component is produced by means of a second rod and must be appropriately shifted. These rods are oscillating followers driven by cams, making it possible to precisely define trajectories as a function of the motor shaft rotation angle. Analysis is performed by numerically solving the equations of motion. Cam synthesis on the basis of the oscillating followers' trajectories makes it possible to obtain cam profiles in order to evaluate pressure angles and check that there is no undercutting. System dynamics is investigated in order to evaluate motor torque and analyse internal stresses on the hinges. In addition, some experimental results and those obtained with the dynamic model are compared. Performance improvement of the actual machine is carried out by modifying the trajectory of the press bed by means of a numerical code at the purpose developed. This approach is more convenient than the use of a commercial multibody code, which is not specifically built for parametric studies

    New insights into enterocin CRL35: mechanism of action and immunity revealed by heterologous expression in Escherichia coli

    The role of the class IIa bacteriocin membrane receptor protein remains unclear, and the following two different mechanisms have been proposed: the bacteriocin could interact with the receptor changing it to an open conformation or the receptor might act as an anchor allowing subsequent bacteriocin insertion and membrane disruption. Bacteriocin-producing cells synthesize an immunity protein that forms an inactive bacteriocin–receptor–immunity complex. To better understand the molecular mechanism of enterocin CRL35, the peptide was expressed as the suicidal probe EtpM-enterocin CRL35 in Escherichia coli, a naturally insensitive microorganism since it does not express the receptor. When the bacteriocin is anchored to the periplasmic face of the plasma membrane through the bitopic membrane protein, EtpM, E. coli cells depolarize and die. Moreover, co-expression of the immunity protein prevents the deleterious effect of EtpM-enterocin CRL35. The binding and anchoring of the bacteriocin to the membrane has demonstrated to be a sufficient condition for its membrane insertion. The final step of membrane disruption by EtpM-enterocin CRL35 is independent from the receptor, which means that the mannose PTS might not be involved in the pore structure.     Percepção dos associados perante as ações de responsabilidade social desenvolvidas em uma cooperativa de eletrificação

    Trabalho de Conclusão de Curso, apresentado para obtenção do grau de Bacharel, no Curso de Ciências Contábeis da Universidade do Extremo Sul Catarinense, UNESC.Por meio do cooperativismo é possível unir o desenvolvimento econômico e social, com melhores oportunidades para todos. O cooperativismo é regido por sete princípios que servem para conduzir os cooperativistas no mundo, dentre eles o 5º caracterizado como a educação, formação e informação e o 7º descrito como o interesse pela comunidade, e estão relacionados com a responsabilidade social. O trabalho visou principalmente identificar qual a percepção dos associados da COOPERA diante das ações de responsabilidade social que esta desenvolve. A metodologia aplicada caracteriza-se quanto aos objetivos como descritiva, quanto à abordagem do problema para a análise de dados como qualitativa e quantitativa em relação aos procedimentos, bibliográfica, estudo de caso e levantamento, com o emprego do questionário. Buscando alcançar mais um dos objetivos específicos deste trabalho, evidenciou-se os diversos projetos de caráter social que a cooperativa pesquisada desenvolve em prol dos associados e da comunidade, dentre eles destacam-se: a revistinha superxoke que leva para as crianças, informações sobre cooperação e preservação do meio ambiente; o projeto cooperjovem, onde realizam-se capacitações de professores, visando que estes desenvolvam atividades colaborativas com as crianças e também procurando difundir o cooperativismo na comunidade; e o projeto juventude cooperativista, que procura trazer o jovem para compreender e participar da cooperativa. Quanto à visão dos associados sobre as ações realizadas pela Cooperativa, verificou-se que a maioria tem conhecimento, ao menos de alguns dos projetos, valorizam e avaliam positivamente o que a COOPERA faz em relação aos projetos sociais. Mas, ainda existem alguns associados que não conhecem nenhum dos projetos e 44,45% não tem conhecimento sobre o balanço social publicado pela instituição pesquisada. Diante disso, conclui-se que a Cooperativa desenvolve várias ações de cunho social e que estas são bem avaliadas, mas nem sempre conhecidas. Por isso, percebe-se a necessidade de melhor divulgação das práticas de responsabilidade social, bem como do balanço social, pois as ações desenvolvidas são importantes e devem ser valorizadas

    New V. cholerae atypical El Tor variant emerged during the 2006 epidemic outbreak in Angola

    <p>Abstract</p> <p>Background</p> <p><it>V. cholerae </it>is the etiological agent of cholera, a major public health concern in most developing countries. Virulence of <it>V. cholerae </it>relies on the powerful cholera toxin, encoded by the CTX prophage. The emergence of new pathogenic variants in the recent years has been mostly associated with new CTX prophage rearrangements.</p> <p>Results</p> <p>In this retrospective study, we show that the epidemic <it>V. cholerae </it>O1 El Tor strain responsible for the 2006 outbreak in Angola is clonally and genetically different from El Tor strains circulating in the 1990s in the same area. Strains from 2006 carry ICE<it>Vch</it>Ang3 of the SXT/R391 family. This ICE is associated with a narrower multidrug resistance profile compared to the one conferred by plasmid p3iANG to strains of the 1990s. The CTX prophage carried by 2006 El Tor strains is characterized by <it>rstR<sup>ET </sup></it>and <it>ctxB<sup>Cla </sup></it>alleles organized in a RS1-RS2-Core array on chromosome I. Interestingly, the newly emerging atypical strain belongs to a clade previously known to comprise only clinical isolates from the Indian subcontinent that also contain the same ICE of the SXT/R391 family.</p> <p>Conclusions</p> <p>Our findings remark the appearance of a novel <it>V. cholerae </it>epidemic variant in Africa with a new CTXΦ arrangement previously described only in the Indian Subcontinent.</p

    Activation State of the Ras2 Protein and Glucose-induced Signaling in Saccharomyces cerevisiae

    The activity of adenylate cyclase in the yeast Saccharomyces cerevisiae is controlled by two G-protein systems, the Ras proteins and the Gα protein Gpa2. Glucose activation of cAMP synthesis is thought to be mediated by Gpa2 and its G-protein-coupled receptor Gpr1. Using a sensitive GTP-loading assay for Ras2 we demonstrate that glucose addition also triggers a fast increase in the GTP loading state of Ras2 concomitant with the glucose-induced increase in cAMP. This increase is severely delayed in a strain lacking Cdc25, the guanine nucleotide exchange factor for Ras proteins. Deletion of the Ras-GAPs IRA2 (alone or with IRA1) or the presence of RAS2Val19 allele causes constitutively high Ras GTP loading that no longer increases upon glucose addition. The glucose-induced increase in Ras2 GTP-loading is not dependent on Gpr1 or Gpa2. Deletion of these proteins causes higher GTP loading indicating that the two G-protein systems might directly or indirectly interact. Because deletion of GPR1 or GPA2 reduces the glucose-induced cAMP increase the observed enhancement of Ras2 GTP loading is not sufficient for full stimulation of cAMP synthesis. Glucose phosphorylation by glucokinase or the hexokinases is required for glucose-induced Ras2 GTP loading. These results indicate that glucose phosphorylation might sustain activation of cAWDP synthesis by enhancing Ras2 GTP loading likely through inhibition of the Ira proteins. Strains with reduced feedback inhibition on cAMP synthesis also display elevated basal and induced Ras2 GTP loading consistent with the Ras2 protein acting as a target of the feedback-inhibition mechanism

    LPS Challenge Regulates Gene Expression and Tissue Localization of a Ciona intestinalis Gene through an Alternative Polyadenylation Mechanism

    subtractive hybridization strategy for the identification of differentially expressed genes was performed between LPSchallenged and naive Ciona intestinalis. This strategy allowed the characterization of two transcripts (Ci8short and Ci8long) generated by the use of two Alternative Polyadenylation sites. The Ci8long transcript contains a protein domain with relevant homology to several components of the Receptor Transporting Protein (RTP) family not present in the Ci8short mRNA. By means of Real Time PCR and Northern Blot, the Ci8short and Ci8long transcripts showed a different pattern of gene expression with the Ci8short mRNA being strongly activated after LPS injection in the pharynx. In situ hybridization analysis demonstrated that the activation of the APA site also influenced the tissue localization of the Ci8short transcript. This analysis showed that the Ci8long mRNA was expressed in hemocytes meanwhile the Ci8short mRNA was highly transcribed also in vessel endothelial cells and in the epithelium of pharynx. These findings demonstrated that regulation of gene expression based on different polyadenylation sites is an ancestral powerful strategy influencing both the level of expression and tissue distribution of alternative transcripts

    On the spatial spread of the Rice Water Weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera: Erirhinidae), in Italy

    A five year study has been made to establish the spread of the rice water weevil Lissorhoptrus oryzophilus (Coleoptera: Erirhinidae) in Northern Italy. Data obtained with GPS from 2005 throughout 2009 were first georeferenced with SW ArcGis&reg; 9.2, then overlapped and compared to the map of the European environmental landscape based on the interpretation of satellite images (CORINE Land Cover map) and to the hydrographic chart CT10 (Technical Regional map 10000). The analysis of the radial rate of spread per year indicates a deceleration in the expansion from 10.864 &plusmn; 6.801 km/year in 2005 to 5.318 &plusmn; 1.401 km/year in 2009. In five years the weevil has expanded its distribution in nearly all rice paddies in Lombardy and Piedmont, over an area of about 200,000 ha, which correspond to 86% of the total Italian rice area. Its expansion is thought to follow a type of stratified dispersal, due both to insect adult active dispersal and to accidental movements caused by human transportation
