    Information organized in any fashion needs to be understood, needs to be communicated in order to be useful. Baudrillard’s views on consumers in and of a postmodern society are employed to describe the users (researchers) vis-à-vis interactions with the metadata described. Hyperreality leads naturally into Introna’s call for a personal relationship as the sine qua non of obligation. This, along with Wittgenstein’s notion of ambiguities in language (meaning) grounded in activities and practices, leads to an ethical level of practice. A number of models are suggested to aid in communication and achieve obligation in information organization

    Increasing Microcystis cell density enhances microcystin synthesis: a mesocosm study

    An experimental protocol using mesocosms was established to study the effect of Microcystis sp. cell abundance on microcystin production. The mesocosms (55 L) were set up in a shallow eutrophic lake and received either no (control), low (to simulate a moderate surface accumulation), or high (to simulate a dense surface scum) concentrations of Microcystis sp. cells collected from the lake water adjacent to the mesocosms. In the low- and high-cell addition mesocosms (2 replicates of each), the initial addition of Microcystis sp. cells doubled the starting cell abundance from 500 000 to 1 000 000 cells mL⁻Âč, but there was no detectable effect on microcystin quotas. Two further cell additions were made to the high-cell addition mesocosms after 60 and 120 min, increasing densities to 2 900 000 and 7 000 000 cells mL-1, respectively. Both additions resulted in marked increases in microcystin quotas from 0.1 pg cell-1 to 0.60 and 1.38 pg cell⁻Âč, respectively, over the 240 min period. Extracellular microcystins accounted for <12% of the total microcystin load throughout the whole experiment. The results of this study indicate a relationship between Microcystis cell abundance and/or mutually correlated environmental parameters and microcystin synthesis

    Increased Coprophagic Activity of the Beetle, Tenebrio molitor, on Feces Containing Eggs of the Tapeworm, Hymenolepis diminuta

    When provided with fecal pellets from uninfected (control) rats and rats infected with the tapeworm Hymenolepis diminuta, more fed and starved (72 h) female and starved male Tenebrio molitor fed on fecal pellets from infected- than from control rats; compared to fecal pellets from controls rats, fed males avoided the infective fecal pellets. Uninfective and infective fecal pellets had similar moisture contents, so increased coprophagic activity was not due to differences in moisture content. Fed and starved males and females were fed on fecal pellets containing tapeworm eggs and examined for cysticercoids. Significantly greater numbers of starved beetles than fed beetles were infected with cysticercoids, but the numbers of infected males and females within each treatment were not significantly different. On the other hand, males contained significantly greater numbers of cysticercoids than did females, and there was no significant difference between the numbers of cysticercoids recovered from fed and starved beetles. The data support the hypothesis that the feeding behavior of T. molitor on rat feces is altered by the presence of tapeworm eggs. The data demonstrated further that transmission dynamics are affected by a complex interaction of the beetle's sex and nutritional status

    The Role of Environmental Exposures in the Etiology of Eosinophilic Esophagitis A Systematic Review

    Eosinophilic esophagitis (EoE) is an emerging clinicopathologic entity defined by abnormal esophageal eosinophilic infiltration. Management of this disease is hampered by limited understanding of etiologic and controllable risk factors. The aim of this systematic review was to determine the environmental risk factors for EoE. We searched PubMed, Web of Science, and EMBASE databases from 1950 through June 30, 2015. To identify additional relevant studies, we hand searched bibliographies of included articles. We limited the review to articles using human subjects, and consisting of case reports, case series, cross-sectional and cohort studies, or clinical trials. 19 articles discuss the risk of environmental exposures on EoE and indicate that environment plays a large role in the etiology of EoE. Seasonal, geographic, and climate-based differences in disease prevalence have been shown but the exact mediators of this process, possibly aeroallergens that vary over time and from place to place, remains elusive

    Comparison of the protein-coding genomes of three deep-sea, sulfur-oxidising bacteria: “Candidatus Ruthia magnifica”, “Candidatus Vesicomyosocius okutanii” and Thiomicrospira crunogena

    Abstract Objective “ Candidatus Ruthia magnifica”, “Candidatus Vesicomyosocius okutanii” and Thiomicrospira crunogena are all sulfur-oxidising bacteria found in deep-sea vent environments. Recent research suggests that the two symbiotic organisms, “Candidatus R. magnifica” and “Candidatus V. okutanii”, may share common ancestry with the autonomously living species T. crunogena. We used comparative genomics to examine the genome-wide protein-coding content of all three species to explore their similarities. In particular, we used the OrthoMCL algorithm to sort proteins into groups of putative orthologs on the basis of sequence similarity. Results The OrthoMCL inflation parameter was tuned using biological criteria. Using the tuned value, OrthoMCL delimited 1070 protein groups. 63.5% of these groups contained one protein from each species. Two groups contained duplicate protein copies from all three species. 123 groups were unique to T. crunogena and ten groups included multiple copies of T. crunogena proteins but only single copies from the other species. “Candidatus R. magnifica” had one unique group, and had multiple copies in one group where the other species had a single copy. There were no groups unique to “Candidatus V. okutanii”, and no groups in which there were multiple “Candidatus V. okutanii” proteins but only single proteins from the other species. Results align with previous suggestions that all three species share a common ancestor. However this is not definitive evidence to make taxonomic conclusions and the possibility of horizontal gene transfer was not investigated. Methodologically, the tuning of the OrthoMCL inflation parameter using biological criteria provides further methods to refine the OrthoMCL procedure

    Effect of natalizumab on disease progression in secondary progressive multiple sclerosis (ASCEND). a phase 3, randomised, double-blind, placebo-controlled trial with an open-label extension

    Background: Although several disease-modifying treatments are available for relapsing multiple sclerosis, treatment effects have been more modest in progressive multiple sclerosis and have been observed particularly in actively relapsing subgroups or those with lesion activity on imaging. We sought to assess whether natalizumab slows disease progression in secondary progressive multiple sclerosis, independent of relapses. Methods: ASCEND was a phase 3, randomised, double-blind, placebo-controlled trial (part 1) with an optional 2 year open-label extension (part 2). Enrolled patients aged 18–58 years were natalizumab-naive and had secondary progressive multiple sclerosis for 2 years or more, disability progression unrelated to relapses in the previous year, and Expanded Disability Status Scale (EDSS) scores of 3·0–6·5. In part 1, patients from 163 sites in 17 countries were randomly assigned (1:1) to receive 300 mg intravenous natalizumab or placebo every 4 weeks for 2 years. Patients were stratified by site and by EDSS score (3·0–5·5 vs 6·0–6·5). Patients completing part 1 could enrol in part 2, in which all patients received natalizumab every 4 weeks until the end of the study. Throughout both parts, patients and staff were masked to the treatment received in part 1. The primary outcome in part 1 was the proportion of patients with sustained disability progression, assessed by one or more of three measures: the EDSS, Timed 25-Foot Walk (T25FW), and 9-Hole Peg Test (9HPT). The primary outcome in part 2 was the incidence of adverse events and serious adverse events. Efficacy and safety analyses were done in the intention-to-treat population. This trial is registered with ClinicalTrials.gov, number NCT01416181. Findings: Between Sept 13, 2011, and July 16, 2015, 889 patients were randomly assigned (n=440 to the natalizumab group, n=449 to the placebo group). In part 1, 195 (44%) of 439 natalizumab-treated patients and 214 (48%) of 448 placebo-treated patients had confirmed disability progression (odds ratio [OR] 0·86; 95% CI 0·66–1·13; p=0·287). No treatment effect was observed on the EDSS (OR 1·06, 95% CI 0·74–1·53; nominal p=0·753) or the T25FW (0·98, 0·74–1·30; nominal p=0·914) components of the primary outcome. However, natalizumab treatment reduced 9HPT progression (OR 0·56, 95% CI 0·40–0·80; nominal p=0·001). In part 1, 100 (22%) placebo-treated and 90 (20%) natalizumab-treated patients had serious adverse events. In part 2, 291 natalizumab-continuing patients and 274 natalizumab-naive patients received natalizumab (median follow-up 160 weeks [range 108–221]). Serious adverse events occurred in 39 (13%) patients continuing natalizumab and in 24 (9%) patients initiating natalizumab. Two deaths occurred in part 1, neither of which was considered related to study treatment. No progressive multifocal leukoencephalopathy occurred. Interpretation: Natalizumab treatment for secondary progressive multiple sclerosis did not reduce progression on the primary multicomponent disability endpoint in part 1, but it did reduce progression on its upper-limb component. Longer-term trials are needed to assess whether treatment of secondary progressive multiple sclerosis might produce benefits on additional disability components. Funding: Biogen

    Molecular biomarkers in the context of focal therapy for prostate cancer: Recommendations of a delphi consensus from the focal therapy society

    BACKGROUND: Focal therapy (FT) for prostate cancer (PCa) is promising. However, long-term oncological results are awaited and there is no consensus on follow-up strategies. Molecular biomarkers (MB) may be useful in selecting, treating and following up men undergoing FT, though there is limited evidence in this field to guide practice. We aimed to conduct a consensus meeting, endorsed by the Focal Therapy Society, amongst a large group of experts, to understand the potential utility of MB in FT for localized PCa. METHODS: A 38-item questionnaire was built following a literature search. The authors then performed three rounds of a Delphi Consensus using DelphiManager, using the GRADE grid scoring system, followed by a face-to-face expert meeting. Three areas of interest were identified and covered concerning MB for FT, 1) the current/present role; 2) the potential/future role; 3) the recommended features for future studies. Consensus was defined using a 70% agreement threshold. RESULTS: Of 95 invited experts, 42 (44.2%) completed the three Delphi rounds. Twenty-four items reached a consensus and they were then approved at the meeting involving (N.=15) experts. Fourteen items reached a consensus on uncertainty, or they did not reach a consensus. They were re-discussed, resulting in a consensus (N.=3), a consensus on a partial agreement (N.=1), and a consensus on uncertainty (N.=10). A final list of statements were derived from the approved and discussed items, with the addition of three generated statements, to provide guidance regarding MB in the context of FT for localized PCa. Research efforts in this field should be considered a priority. CONCLUSIONS: The present study detailed an initial consensus on the use of MB in FT for PCa. This is until evidence becomes available on the subject
