    Population expansion in the North African Late Pleistocene signalled by mitochondrial DNA haplogroup U6

    Background <br/> The archaeology of North Africa remains enigmatic, with questions of population continuity versus discontinuity taking centre-stage. Debates have focused on population transitions between the bearers of the Middle Palaeolithic Aterian industry and the later Upper Palaeolithic populations of the Maghreb, as well as between the late Pleistocene and Holocene. <br/> Results Improved resolution of the mitochondrial DNA (mtDNA) haplogroup U6 phylogeny, by the screening of 39 new complete sequences, has enabled us to infer a signal of moderate population expansion using Bayesian coalescent methods. To ascertain the time for this expansion, we applied both a mutation rate accounting for purifying selection and one with an internal calibration based on four approximate archaeological dates: the settlement of the Canary Islands, the settlement of Sardinia and its internal population re-expansion, and the split between haplogroups U5 and U6 around the time of the first modern human settlement of the Near East. <br/> Conclusions <br/> A Bayesian skyline plot placed the main expansion in the time frame of the Late Pleistocene, around 20 ka, and spatial smoothing techniques suggested that the most probable geographic region for this demographic event was to the west of North Africa. A comparison with U6's European sister clade, U5, revealed a stronger population expansion at around this time in Europe. Also in contrast with U5, a weak signal of a recent population expansion in the last 5,000 years was observed in North Africa, pointing to a moderate impact of the late Neolithic on the local population size of the southern Mediterranean coast

    Flocculation of Arthrospira maxima for improved harvesting

    The 6th International Conference on Energy and Environment ResearchThe environmental impacts associated with the burning of fossil fuels coupled with growing concerns about security of energy supply, motivated the search for more sustainable forms of energy production, among which came microalgae for biofuels production. However, the commercial production of microalgae biofuels is still not competitive compared to fossil fuels, as it is necessary to solve some process bottlenecks, among which biomass harvesting, that is the focus of this work. Hence, this work intends to study the harvesting of microalga Arthrospira maxima through flocculation by pH variation and/or addition of CaCl2 as flocculant. Thus, it is described the effect of pH variation (in the range 6 to 12), followed by the addition of flocculant, on the harvesting efficiency.     Combining M-FISH and Quantum Dot technology for fast chromosomal assignment of transgenic insertions

    <p>Abstract</p> <p>Background</p> <p>Physical mapping of transgenic insertions by Fluorescence in situ Hybridization (FISH) is a reliable and cost-effective technique. Chromosomal assignment is commonly achieved either by concurrent G-banding or by a multi-color FISH approach consisting of iteratively co-hybridizing the transgenic sequence of interest with one or more chromosome-specific probes at a time, until the location of the transgenic insertion is identified.</p> <p>Results</p> <p>Here we report a technical development for fast chromosomal assignment of transgenic insertions at the single cell level in mouse and rat models. This comprises a simplified 'single denaturation mixed hybridization' procedure that combines multi-color karyotyping by Multiplex FISH (M-FISH), for simultaneous and unambiguous identification of all chromosomes at once, and the use of a Quantum Dot (QD) conjugate for the transgene detection.</p> <p>Conclusions</p> <p>Although the exploitation of the unique optical properties of QD nanocrystals, such as photo-stability and brightness, to improve FISH performance generally has been previously investigated, to our knowledge this is the first report of a purpose-designed molecular cytogenetic protocol in which the combined use of QDs and standard organic fluorophores is specifically tailored to assist gene transfer technology.</p

    L1 sequence of a new human papillomavirus type-58 variant associated with cervical intraepithelial neoplasia

    The present study on molecular characterization of a human papillomavirus (HPV) isolated in Central Brazil describes the L1 gene sequence from a new variant of HPV-58, the isolate Bsb-02. The sample was from a smear obtained from a woman with cervical intraepithelial neoplasia grade II. The whole L1 gene from isolate Bsb-02 was sequenced automatically, showing 99.1% nucleotide identity with the gene from the HPV-58 reference. The clustering between Bsb-02 and HPV-58 reference sequence was also supported by phylogenetic analysis. Fourteen nucleotide substitutions were observed: eight were synonymous and six were associated with amino acid substitutions. A10V and V144I have not been previously described. At GenBank, the only complete L1 sequence from HPV-58 in addition to the HPV-58 reference one is that of Bsb-02. These data provide information that may be relevant to HPV diagnosis and to rational vaccine strategies. HPV variants may also be associated with host immune responses and with the risk of cervical neoplasia

    Suppression of static stripe formation by next-neighbor hopping

    We show from real-space Hartree-Fock calculations within the extended Hubbard model that next-nearest neighbor (t') hopping processes act to suppress the formation of static charge stripes. This result is confirmed by investigating the evolution of charge-inhomogeneous corral and stripe phases with increasing t' of both signs. We propose that large t' values in YBCO prevent static stripe formation, while anomalously small t' in LSCO provides an additional reason for the appearance of static stripes only in these systems.Comment: 4 pages, 5 figure

    The effect of Nannochloropsis oceanica feed inclusion on rabbit muscle proteome

    Nannochloropsis oceanica is a microalga, highly concentrated in protein and EPA (eicosapentaenoic acid). It has a recalcitrant cell wall that decreases nutrient digestibility. The objective of this study is to evaluate the effect of Nannochloropsis oceanica feed inclusion on the muscle proteome of fattening rabbits. Twenty rabbits were housed in individual cages for 5 weeks and were fed a control diet (n = 10) and one with 4.45% inclusion of microalga, replacing whole soybean meal (n = 10). After slaughter, samples of gastrocnemius muscle were taken for label-free proteomic analysis. A total of 1497 proteins were identified, 46 with differential abundance. Control rabbits had high abundance of proteins related to protein metabolism, suggesting higher muscle protein turnover. They also had higher abundance of structural proteins, suggesting a less tender meat by comparison with algae-fed rabbits. These had high abundance of proteins related to amino acid catabolism (Phe, Tyr) and synthesis (Gln).     Mineral profiling of muscle and hepatic tissues of Australian Merino, Damara and Dorper lambs: Effect of weight loss

    Seasonal weight loss (SWL) is a major constraint to extensive animal production systems. The Australian sheep production is based on merino sheep, a European breed not tolerant to SWL. Tolerant alternative breeds such as the fat-tailed Damara and the Dorper have been increasingly used in Australia and elsewhere, due to their robustness. The aim of this study was to understand the mineral profile of muscle and liver tissues of Australian Merino, Damara and Dorper, when subjected to SWL in order to understand SWL-tolerance physiology. Twenty-four lambs were divided randomly between growing (control) and nutritionally restricted groups for each breed. The trial lasted 42 days. Animals were weighed bi-weekly and at the end of the trial, lambs were slaughtered. Liver and muscle samples were taken immediately after slaughter. Mineral assessment was carried out using inductively coupled plasma–optical emission spectrometry. Analysis of variance showed mineral concentrations were generally increased in the muscle of restricted animals, mainly because of fat tissue mobilization. An increase in Zn and Fe concentrations indicates an increase of enzymatic activity in the liver of restricted sheep as well as differential abundance of Fe-containing proteins.     Multiparasitism in Crab-eating fox (Cerdocyon thous) (Carnivora: Canidae), including new records, in the Brazilian Pampa - case report

    ABSTRACT Cerdocyon thous is considered the most common wild canid in Rio Grande do Sul, and can serve as a host for several parasites, playing an important role in maintaining the biological cycle. Despite being a widespread and abundant species, they have a relatively little-known fauna of endo and ectoparasites, mainly focusing on the different regions of their occurrence. With this work, the objective was to report the multiparasitism in a specimen of C. thous in the Brazilian Pampa. Using modified Centrifuge-Flotation and Spontaneous Sedimentation techniques, a high taxonomic diversity of parasites was observed, and the finding of eggs compatible with Lagochilascaris spp. in C. thous in Brazil, in addition to Uncinaria spp. eggs and Cystoisospora spp. oocysts in the same host in the south of the country. The presence of eggs of different helminths in the feces of C. thous is extremely important, mainly for public health, but also for the knowledge of the biodiversity of parasites present in wild populations of canids, until then little studied, thus allowing the implementation of control programs

    Influence of dietary Chlorella vulgaris and carbohydrate-active enzymes on growth performance, meat quality and lipid composition of broiler chickens

    Article in pressHerein, we investigated the effect of Chlorella vulgaris as ingredient (10% of incorporation) in broiler diets, supplemented or not with 2 formulations of Carbohydrate-Active enZymes (CAZymes; Rovabio Excel AP and a mixture of recombinant CAZymes, composed by an exo-b-glucosaminidase, an alginate lyase, a peptidoglycan N-acetylmuramic acid deacetylase and a lysozyme), on growth performance, meat quality, fatty acid composition, oxidative stability, and sensory traits. One hundred twenty 1-day-old Ross 308 male birds were randomly assigned to one of the 4 experimental diets (n 5 30): corn-soybean meal–basal diet (control), basal diet with 10% C. vulgaris (CV), CV supplemented with 0.005% of a commercialCAZymecocktail (Rovabio Excel AP), (CV1R), and CV supplemented with 0.01% of a 4- CAZyme mixture previously selected (CV 1 M) during the experimental period lasted from day 21 to day 35. Body weight gain and feed conversion rate of broilers were not affected by C. vulgaris but digesta viscosity increased more than 2-fold (P , 0.001) relative to the control. In addition, neither cooking loss, shear force, juiciness, flavor nor off-flavor was impaired by dietary treatments (P.0.05). By contrast, the dietary C. vulgaris increased tenderness, yellowness (b*) and total carotenoids in breast and thigh meats. However, no additional protective effect against lipid oxidation was observed in meat with the inclusion of microalga. Chlorella vulgaris, independently of CAZymes, had a minor impact on meat fatty acid composition but improved the proportion of some beneficial fatty acids. In summary, our data indicate a slight improvement of broiler meat quality and lipid nutritional value, without impairment of broilers’ growth performance, thus supporting the usefulness of this microalga in poultry diets, up to this high level of incorporation.
