    Fatigue in Women with Fibromyalgia: A Gene-Physical Activity Interaction Study

    Fatigue is a cardinal symptom in fibromyalgia. Fatigue is assumed to be the result of genetic susceptibility and environmental factors. We aimed at examining the role of genetic susceptibility for fatigue in southern Spanish women with fibromyalgia, by looking at single nucleotide polymorphisms in 34 fibromyalgia candidate-genes, at the interactions between genes, and at the gene-physical activity interactions. We extracted DNA from saliva of 276 fibromyalgia women to analyze gene-polymorphisms. Accelerometers registered physical activity and sedentary behavior. Fatigue was assessed with the Multidimensional Fatigue Inventory. Based on the Bonferroni's and False Discovery Rate values, we found that the genotype of the rs4453709 polymorphism (sodium channel protein type 9 subunit alpha, SCN9A, gene) was related to reduced motivation (AT carriers showed the highest reduced motivation) and reduced activity (AA carriers showed the lowest reduced activity). Carriers of the heterozygous genotype of the rs1801133 (methylene tetrahydrofolate reductase, MTHFR, gene) or rs4597545 (SCN9A gene) polymorphisms who were physically active reported lower scores on fatigue compared to their inactive counterparts. Highly sedentary carriers of the homozygous genotype of the rs7607967 polymorphism (AA/GG genotype; SCN9A gene) presented more reduced activity (a dimension of fatigue) than those with lower levels of sedentary behavior. Collectively, findings from the present study suggest that the contribution of genetics and gene-physical activity interaction to fatigue in fibromyalgia is modest

    Land- and water-based exercise intervention in women with fibromyalgia: the al-andalus physical activity randomised controlled trial

    Background The al-Andalus physical activity intervention study is a randomised control trial to investigate the effectiveness of a land- and water-based exercise intervention for reducing the overall impact of fibromyalgia (primary outcome), and for improving tenderness and pain-related measures, body composition, functional capacity, physical activity and sedentary behaviour, fatigue, sleep quality, health-related quality of life, and cognitive function (secondary outcomes) in women with fibromyalgia. Methods/Design One hundred eighty women with fibromyalgia (age range: 35-65 years) will be recruited from local associations of fibromyalgia patients in Andalucía (Southern Spain). Patients will be randomly assigned to a usual care (control) group (n = 60), a water-based exercise intervention group (n = 60) or a land-based exercise intervention group (n = 60). Participants in the usual care group will receive general physical activity guidelines and participants allocated in the intervention groups will attend three non-consecutive training sessions (60 min each) per week during 24 weeks. Both exercise interventions will consist of aerobic, muscular strength and flexibility exercises. We will also study the effect of a detraining period (i.e., 12 weeks with no exercise intervention) on the studied variables. Discussion Our study attempts to reduce the impact of fibromyalgia and improve patients' health status by implementing two types of exercise interventions. Results from this study will help to assess the efficacy of exercise interventions for the treatment of fibromyalgia. If the interventions would be effective, this study will provide low-cost and feasible alternatives for health professionals in the management of fibromyalgia.     QUERCETIN EFFECTS ON WEIGHT GAIN AND CALORIC INTAKE IN EXERCISED RATS

    Quercetin is a flavonoid which activates oxidative metabolism. Quercetin may reduce weight gain by decreasing feed efficiency. The present study aims to evaluate weight gain, caloric intake and feed efficiency in exercised and sedentary rats supplemented with quercetin. Wistar rats were divided into four groups: quercetin-exercise training (QT), quercetin-sedentary (QS), placebo-exercise training (PT) and placebo-sedentary (PS). Rats were exercised and/or orally supplemented with quercetin (25 mg · kg-1 on alternate days) during six weeks. Weight gain of the QT group decreased when compared with the PT and PS groups. Exercised groups increased cumulative caloric intake during the experimental period. The QT group rats also reduced their feed efficiency when compared with the QS and PS groups. These results suggest that quercetin is not able to decrease weight gain because no differences were found between placebo and quercetin condition either in the sedentary or in the training condition

    Efectos del ejercicio aeróbico interválico, combinado con entrenamiento de fuerza y de la restricción calórica, sobre la composición corporal de ratas obesas

    Objetivo: Investigar los efectos de un programa de ejercicio aeróbico interválico, combinado con entrenamiento de fuerza, y de la restricción calórica, sobre el peso y la composición corporal de ratas genéticamente obesas. Método: Un total de 32 ratas genéticamente obesas fueron divididas aleatoriamente en dos grupos (n = 16) con o sin ejercicio aeróbico interválico combinado con entrenamiento de fuerza. A su vez, cada grupo, con o sin ejercicio aeróbico interválico combinado con entrenamiento de fuerza, fue dividido en dos grupos, con o sin restricción calórica, resultando cuatro grupos de intervención (n = 8). El programa de ejercicio aeróbico interválico combinado con entrenamiento de fuerza se llevó a cabo en una misma sesión de 60 min, cinco días por semana, durante dos meses. Los grupos con restricción calórica consumieron un 30% menos de alimento que los ad libitum. Se determinó el peso corporal y la masa de la carcasa mediante una balanza de precisión y la masa grasa, muscular y el agua corporal, mediante un analizador de composición corporal, basado en resonancia magnética nuclear, de diseño especial para ratas. Resultados: El peso corporal final fue menor en aquellos animales que realizaron restricción calórica frente a aquellos cuya alimentación fue ad libitum (p < 0.001). La masa grasa fue menor y la masa muscular mayor en aquellos animales que desarrollaron el ejercicio aeróbico interválico combinado con entrenamiento de fuerza, frente a los sedentarios (p < 0.001), sin diferencias atendiendo a la restricción calórica (p > 0.05). Hemos observado interacciones entrenamiento versus restricción calórica, en masa muscular y agua total. Los animales que incrementaron más su masa muscular y agua corporal, fueron los que combinaron el entrenamiento con la restricción calórica (ambos p < 0.01). Conclusiones: Tanto el programa de ejercicio aeróbico interválico, combinado con entrenamiento de fuerza desarrollado, como la restricción calórica, fueron eficaces, mejorando la composición corporal de ratas obesas, pero el ejercicio lo hizo en mayor magnitud

    Identification of candidate genes associated with fibromyalgia susceptibility in southern Spanish women : The al-Ándalus project

    Background: Candidate-gene studies on fibromyalgia susceptibility often include a small number of single nucleotide polymorphisms (SNPs), which is a limitation. Moreover, there is a paucity of evidence in Europe. Therefore, we compared genotype frequencies of candidate SNPs in a well-characterised sample of Spanish women with fibromyalgia and healthy non-fibromyalgia women. Methods: A total of 314 women with a diagnosis of fibromyalgia (cases) and 112 non-fibromyalgia healthy (controls) women participated in this candidate-gene study. Buccal swabs were collected for DNA extraction. Using TaqMan™ OpenArray™, we analysed 61 SNPs of 33 genes related to fibromyalgia susceptibility, symptoms, or potential mechanisms. Results: We observed that the rs841 and rs1799971 GG genotype was more frequently observed in fibromyalgia than in controls (p = 0.04 and p = 0.02, respectively). The rs2097903 AT/TT genotypes were also more often present in the fibromyalgia participants than in their control peers (p = 0.04). There were no differences for the remaining SNPs. Conclusions: We identified, for the first time, associations of the rs841 (guanosine triphosphate cyclohydrolase 1 gene) and rs2097903 (catechol-O-methyltransferase gene) SNPs with higher risk of fibromyalgia susceptibility. We also confirmed that the rs1799971 SNP (opioid receptor μ1 gene) might confer genetic risk of fibromyalgia. We did not adjust for multiple comparisons, which would be too stringent and yield to non-significant differences in the genotype frequencies between cases and controls. Our findings may be biologically meaningful and informative, and should be further investigated in other populations. Of particular interest is to replicate the present study in a larger independent sample to confirm or refute our findings. On the other hand, by including 61 SNPs of 33 candidate-genes with a strong rationale (they were previously investigated in relation to fibromyalgia susceptibility, symptoms or potential mechanisms), the present research is the most comprehensive candidate-gene study on fibromyalgia susceptibility to date
