    First use of Timepix3 hybrid pixel detectors in ultra-high vacuum for beam profile measurements

    A transverse beam gas ionization profile monitor is currently under development for the CERN Proton Synchrotron (PS) to provide non-destructive continuous measurements during a beam cycle. The implementation is exploring a novel use of the Timepix3 hybrid pixel detector mounted inside the ultra-high vacuum of the accelerator beam pipe to provide direct detection of ionization electrons. In early 2017, a prototype monitor was installed and has been used successfully to measure the transverse beam profile. The evolution of the transverse beam profile throughout the beam cycle has been measured and specific time windows within a beam cycle have been studied, for example the transition crossing. A radiation tolerant readout system for the Timepix3 detectors has been implemented which enables the connection of up to four detectors located in a highly radioactive environment. The first version of the readout was installed together with the prototype monitor in 2017 and a new version of the readout is currently under development which will enable the full speed data rate of the pixel detectors. Use of the radiation tolerant readout system can be envisioned for other beam instrumentation applications, which could provide new insight to beam diagnostics

    A Dynamical Model of Oocyte Maturation Unveils Precisely Orchestrated Meiotic Decisions

    Maturation of vertebrate oocytes into haploid gametes relies on two consecutive meioses without intervening DNA replication. The temporal sequence of cellular transitions driving eggs from G2 arrest to meiosis I (MI) and then to meiosis II (MII) is controlled by the interplay between cyclin-dependent and mitogen-activated protein kinases. In this paper, we propose a dynamical model of the molecular network that orchestrates maturation of Xenopus laevis oocytes. Our model reproduces the core features of maturation progression, including the characteristic non-monotonous time course of cyclin-Cdks, and unveils the network design principles underlying a precise sequence of meiotic decisions, as captured by bifurcation and sensitivity analyses. Firstly, a coherent and sharp meiotic resumption is triggered by the concerted action of positive feedback loops post-translationally activating cyclin-Cdks. Secondly, meiotic transition is driven by the dynamic antagonism between positive and negative feedback loops controlling cyclin turnover. Our findings reveal a highly modular network in which the coordination of distinct regulatory schemes ensures both reliable and flexible cell-cycle decisions

    What’s in a Relationship: An Ontological Analysis

    Abstract. In a series of publications, we have proposed a foundational system of ontological categories which has been successfully used to evaluate and im-prove the quality of conceptual modeling grammars and models. In this article, we continue this work by using this foundational ontology to provide real-world semantics and sound modeling guidelines for one of the most fundamental (and yet one of the most problematic) constructs in conceptual modeling, namely, the relationship type. In addition, we systematically compare our approach with a classical ontological treatment of this construct in the literature, provided by the BWW framework.

    Installation and Hardware commissioning of the Multi-Turn extraction at the CERN proton synchrotron

    The implementation of the new Multi-Turn Extraction (MTE) at the CERN Proton Synchrotron required major hardware changes for the nearly 50-year old accelerator. The installation of new Pulse Forming Networks (PFN) and refurbished kicker magnets for the extraction, new sextupole and octupole magnets, new power converters, together with an in-depth review of the machine aperture leading to the design of new vacuum chambers was required. As a result, a heavy programme of interventions had to be scheduled during the winter shut-down 2007-8. The newly installed hardware and its commissioning is presented and discussed in details

    Soluble CD36 Ectodomain Binds Negatively Charged Diacylglycerol Ligands and Acts as a Co-Receptor for TLR2

    BACKGROUND:Cluster of differentiation 36 (CD36) is a transmembrane glycoprotein involved in many biological processes, such as platelet biology, angiogenesis and in the aetiopathology of atherosclerosis and cardiovascular diseases. Toll-like receptors (TLRs) are one of the most important receptors of the innate immune system. Their main function is the recognition of conserved structure of microorganisms. This recognition triggers signaling pathways that activate transcription of cytokines and co-stimulatory molecules which participate in the generation of an immune response against microbes. In particular, TLR2 has been shown to recognize a broad range of ligands. Recently, we showed that CD36 serves as a co-receptor for TLR2 and enhances recognition of specific diacylglycerides derived from bacteria. METHODOLOGY/ PRINCIPAL FINDINGS:Here, we investigate the mechanism by which CD36 contributes to ligand recognition and activation of TLR2 signaling pathway. We show that the ectodomain of murine CD36 (mCD36ED) directly interacts with negatively charged diacylglycerol ligands, which explains the specificity and selectivity of CD36 as a TLR2 co-receptor. We also show that mCD36ED amplifies the pro-inflammatory response to lipoteichoic acid in macrophages of wild-type mice and restores the pro-inflammatory response of macrophages from mice deficient in CD36 (oblivious), but not from mice deficient in cluster of differentiation 14 (CD14) (heedless). CONCLUSION/ SIGNIFICANCE: These data indicate that the CD36 ectodomain is the only relevant domain for activation of TLR2 signaling pathway and that CD36 and CD14 have a non-redundant role for loading ligands onto TLR2 in the plasma-membrane. The pro-inflammatory role of soluble CD36 can be relevant in the activation of the immune response against pathogens, as well as in the progression of chronic diseases. Therefore, an increased level of soluble forms of CD36, which has been reported to be increased in type II diabetic patients, could accelerate atherosclerosis by increasing the pro-inflammatory response to diacylglycerol ligands

    Can gold be used as a hedge against the risks of Sharia-compliant securities? Application for Islamic portfolio management

    In this paper, we investigate whether gold hedges Sharia-compliant stocks and Sukuk during the period from September 2005 to October 2017. The inference is taken by using both the DCC-GARCH model and the wavelet coherence analysis. On the whole, our finding suggests that gold is not effective in hedging the fluctuations of Sharia-compliant securities. However, we find that combining gold with stocks (and Sukuk) is useful in diversification and portfolio optimization. These results imply that, while gold is an excellent hedge for plain vanilla securities, it is not for Islamic exposures. This is important in light of the increasing amount of assets that are managed according to Islamic screening

    Antarctic Bedmap data: Findable, Accessible, Interoperable, and Reusable (FAIR) sharing of 60 years of ice bed, surface, and thickness data

    One of the key components of this research has been the mapping of Antarctic bed topography and ice thickness parameters that are crucial for modelling ice flow and hence for predicting future ice loss and the ensuing sea level rise. Supported by the Scientific Committee on Antarctic Research (SCAR), the Bedmap3 Action Group aims not only to produce new gridded maps of ice thickness and bed topography for the international scientific community, but also to standardize and make available all the geophysical survey data points used in producing the Bedmap gridded products. Here, we document the survey data used in the latest iteration, Bedmap3, incorporating and adding to all of the datasets previously used for Bedmap1 and Bedmap2, including ice bed, surface and thickness point data from all Antarctic geophysical campaigns since the 1950s. More specifically, we describe the processes used to standardize and make these and future surveys and gridded datasets accessible under the Findable, Accessible, Interoperable, and Reusable (FAIR) data principles. With the goals of making the gridding process reproducible and allowing scientists to re-use the data freely for their own analysis, we introduce the new SCAR Bedmap Data Portal (https://bedmap.scar.org, last access: 1 March 2023) created to provide unprecedented open access to these important datasets through a web-map interface. We believe that this data release will be a valuable asset to Antarctic research and will greatly extend the life cycle of the data held within it.     Evaluating quality of obstetric care in low-resource settings: Building on the literature to design tailor-made evaluation instruments - an illustration in Burkina Faso

    <p>Abstract</p> <p>Background</p> <p>There are many instruments available freely for evaluating obstetric care quality in low-resource settings. However, this profusion can be confusing; moreover, evaluation instruments need to be adapted to local issues. In this article, we present tools we developed to guide the choice of instruments and describe how we used them in Burkina Faso to facilitate the participative development of a locally adapted instrument.</p> <p>Methods</p> <p>Based on a literature review, we developed two tools: a conceptual framework and an analysis grid of existing evaluation instruments. Subsequently, we facilitated several sessions with evaluation stakeholders in Burkina Faso. They used the tools to develop a locally adapted evaluation instrument that was subsequently tested in six healthcare facilities.</p> <p>Results</p> <p>Three outputs emerged from this process:</p> <p>1) A comprehensive conceptual framework for the quality of obstetric care, each component of which is a potential criterion for evaluation.</p> <p>2) A grid analyzing 37 instruments for evaluating the quality of obstetric care in low-resource settings. We highlight their key characteristics and describe how the grid can be used to prepare a new evaluation.</p> <p>3) An evaluation instrument adapted to Burkina Faso. We describe the experience of the Burkinabé stakeholders in developing this instrument using the conceptual framework and the analysis grid, while taking into account local realities.</p> <p>Conclusions</p> <p>This experience demonstrates how drawing upon existing instruments can inspire and rationalize the process of developing a new, tailor-made instrument. Two tools that came out of this experience can be useful to other teams: a conceptual framework for the quality of obstetric care and an analysis grid of existing evaluation instruments. These provide an easily accessible synthesis of the literature and are useful in integrating it with the context-specific knowledge of local actors, resulting in evaluation instruments that have both scientific and local legitimacy.</p
