    A human factors approach to defining requirements for low-speed autonomous vehicles to enable intelligent platooning

    This paper presents results from a series of focus groups, aimed at enhancing technical engineering system requirements, for a public transport system, encompassing a fleet of platooning low-speed autonomous vehicles (LSAV; aka pods) in urban areas. A critical review of the pods was conducted, as part of a series of technical workshops, to examine the key areas of the system that could affect users and other stakeholders, such as businesses and the public. These initial findings were used to inform a series of focus groups, aimed at identifying the public's views of multiple autonomous vehicles being deployed in a pedestrianised area that can join and form platoons. Analysis of findings from the focus groups suggests that while people view platooning public transport vehicles favourably as a passenger, they have some concerns from a pedestrian perspective. Thematic analysis was applied to these findings and a systematic approach was used to identify where subjective outputs could be formalised to inform requirements. Finally, a step-by-step requirements elicitation process is presented that illustrates the method used to convert qualitative user data to objective engineering requirements

    Gap acceptance study of pedestrians crossing between platooning autonomous vehicles in a virtual environment

    Autonomous vehicles (AVs) operating in shared urban environments, often referred to as “pods”, will constantly have to interact with pedestrians. As a result, an effective strategy will be required for pods to continue operating, while in close proximity to people. This strategy could be in terms of active negotiation, where a pod identifies a person and gives way; or a more passive strategy, such as requiring pods to travel close together in platoons, in order to reduce the number of individual vehicle encounters. For this latter example, it is critical to understand how the spaces between pods and AVs in general are perceived by pedestrians, and what factors will persuade and dissuade crossing. Therefore, this paper seeks to understand this relationship, and presents results from a pedestrian gap acceptance study for platoons. To ensure the safety of participants, a virtual environment was used instead of real vehicles. The goal of the experiment described in this paper, is to understand the gap acceptance behaviour of participants, when presented with a platoon of pods in different environments. The experiment evaluated four vehicle speeds, from 1 km/h to 16 km/h, four temporal gaps, from 2 s to 5 s, and two environments. These environments were a typical road layout, with footpath and line markings, and a shared space, where all markings and separation between pod and pedestrian were removed. For each scenario, participants were asked if they would cross between the pods and how safe they felt about the situation, recorded as a Likert score. The results suggest that people are more likely to attempt to cross between a platoon of pods when they are travelling closer together in a shared space (no line markings or separation between vehicles and pedestrian), compared to a road environment (separated by raised pavement and road markings). However, it was also found that people’s subjective rating of safeness was higher in the road environment, when presented with a platoon of pods, compared to the shared space

    Strategic engagement: new models of relationship management for academic librarians

    How do we best bridge the gap between the Library and the diverse academic communities it serves? Librarians need new strategies for engagement. Traditional models of liaison, aligning solutions to disciplines, are yielding to functional specialisms, including a focus on building partnerships. This paper offers a snapshot of realignment across the Russell Group from subject support to relationship management. It then follows the journey of a newly-formed Faculty and School Engagement Team. Techniques are explored for building relationship capital, anchored to a model Strategic Engagement Cycle. Theory is contrasted with the challenges of securing real buy-in to new ways of working amid diverging agendas and assumptions, notably within the Library itself. Consideration is given to the retention of aspects of subject librarian roles. Investment in a relationship management function demands staunch and ongoing commitment to fulfil its promise, not only from its performers but from across the library community

    Is season of birth related to disordered eating and personality in women with eating disorders?

    We assessed the relation between season of birth and eating disorder symptoms and personality characteristics in a sample of 880 women with eating disorders and 580 controls from two Price Foundation Studies. Eating disorder symptoms were assessed using the Structured Interview of Anorexic and Bulimic Disorders and the Structured Clinical Interview for DSM-IV. Personality traits were assessed using the Temperament and Character Inventory and the Frost Multidimensional Perfectionism Scale. Date of birth was obtained from a sociodemographic questionnaire. No significant differences were observed 1) in season of birth across eating disorder subtypes and controls; nor 2) for any clinical or personality variables and season of birth. We found no evidence of season of birth variation in eating disorders symptoms or personality traits. Contributing to previous conflicting findings, the present results do not support a season of birth hypothesis for eating disorders

    Evidence for Significant Overlap between Common Risk Variants for Crohn's Disease and Ankylosing Spondylitis

    BACKGROUND: A multicenter genome-wide association scan for Crohn's Disease (CD) has recently reported 40 CD susceptibility loci, including 29 novel ones (19 significant and 10 putative). To gain insight into the genetic overlap between CD and ankylosing spondylitis (AS), these markers were tested for association in AS patients. PRINCIPAL FINDINGS: Two previously established associations, namely with the MHC and IL23R loci, were confirmed. In addition, rs2872507, which maps to a locus associated with asthma and influences the expression of the ORMDL3 gene in lymphoblastoid cells, showed a significant association with AS (p = 0.03). In gut biopsies of AS and CD patients, ORMDL3 expression was not significantly different from controls and no correlation was found with the rs2872507 genotype (Spearman's rho: -0.067). The distribution of p-values for the remaining 36 SNPs was significantly skewed towards low p-values unless the top 5 ranked SNPs (ORMDL3, NKX2-3, PTPN2, ICOSLG and MST1) were excluded from the analysis. CONCLUSIONS: Association analysis using risk variants for CD led to the identification of a new risk variant associated with AS (ORMDL3), underscoring a role for ER stress in AS. In addition, two known and five potentially relevant associations were detected, contributing to common susceptibility of CD and AS

    Interaction Pattern of Arg 62 in the A-Pocket of Differentially Disease-Associated HLA-B27 Subtypes Suggests Distinct TCR Binding Modes

    The single amino acid replacement Asp116His distinguishes the two subtypes HLA-B*2705 and HLA-B*2709 which are, respectively, associated and non-associated with Ankylosing Spondylitis, an autoimmune chronic inflammatory disease. The reason for this differential association is so far poorly understood and might be related to subtype-specific HLA:peptide conformations as well as to subtype/peptide-dependent dynamical properties on the nanoscale. Here, we combine functional experiments with extensive molecular dynamics simulations to investigate the molecular dynamics and function of the conserved Arg62 of the α1-helix for both B27 subtypes in complex with the self-peptides pVIPR (RRKWRRWHL) and TIS (RRLPIFSRL), and the viral peptides pLMP2 (RRRWRRLTV) and NPflu (SRYWAIRTR). Simulations of HLA:peptide systems suggest that peptide-stabilizing interactions of the Arg62 residue observed in crystal structures are metastable for both B27 subtypes under physiological conditions, rendering this arginine solvent-exposed and, probably, a key residue for TCR interaction more than peptide-binding. This view is supported by functional experiments with conservative (R62K) and non-conservative (R62A) B*2705 and B*2709 mutants that showed an overall reduction in their capability to present peptides to CD8+ T cells. Moreover, major subtype-dependent differences in the peptide recognition suggest distinct TCR binding modes for the B*2705 versus the B*2709 subtype

    Analyzing multitarget activity landscapes using protein-ligand interaction fingerprints: interaction cliffs.

The final peer-reviewed version is available from ACS at http://pubs.acs.org/doi/abs/10.1021/ci500721x.Activity landscape modeling is mostly a descriptive technique that allows rationalizing continuous and discontinuous SARs. Nevertheless, the interpretation of some landscape features, especially of activity cliffs, is not straightforward. As the nature of activity cliffs depends on the ligand and the target, information regarding both should be included in the analysis. A specific way to include this information is using protein-ligand interaction fingerprints (IFPs). In this paper we report the activity landscape modeling of 507 ligand-kinase complexes (from the KLIFS database) including IFP, which facilitates the analysis and interpretation of activity cliffs. Here we introduce the structure-activity-interaction similarity (SAIS) maps that incorporate information on ligand-target contact similarity. We also introduce the concept of interaction cliffs defined as ligand-target complexes with high structural and interaction similarity but have a large potency difference of the ligands. Moreover, the information retrieved regarding the specific interaction allowed the identification of activity cliff hot spots, which help to rationalize activity cliffs from the target point of view. In general, the information provided by IFPs provides a structure-based understanding of some activity landscape features.
