    Effect of climate on the epidemiology of bovine hypodermosis in algeria

    In order to explore the effect of climate on cattle warble fly infestation, a total of 1.635 animals from 4 departments of Northern Algeria were examined visually and by manual palpation for the presence of warbles. Cattle were examined from March to June 2014, coinciding with the peak of emergence of warbles, and both the prevalence and intensity of infestation were recorded. The departments included in this study were located in the two different climatic areas in Northern Algeria: humid (Bejaia and Tizi Ouzou) and semi-arid (Tissemssilt and Ain Defla). The overall prevalence was 28.75%; the intensity of infestation ranged from 1 to 98 warbles per animal (mean 18.93±11.05). The prevalence and intensity of infestation within the departments with semi-arid climate (38.23%; 21.57±11.98) was significantly higher than in those with humid climate (20.74%; 14.84±7.86). The CHAID algorithm showed the climate as the most influencing factor for warble fly prevalence, followed by the husbandry system and breed. Logistic regression and multivariate ANOVA indicate that in addition to climate, other intrinsic (age, breed) and extrinsic factors (husbandry system, treatment) included in the study also were associated with both, prevalence and intensity of infestation. Our results indicate that in semiarid areas of Northern Algeria environmental conditions are more favorable for the development of free stages (pupae and adult flies) of Hypoderma spp life-cycle than in humid areas

    Intrinsic Factors Influencing the Infection by Helminth Parasites in Horses under an Oceanic Climate Area (NW Spain)

    A coprological survey to determine the influence of some intrinsic factors (breed, age, and sex) on the infection by helminth parasites in equine livestock (n = 418) under an oceanic climate area (NW Spain) was conducted. Faecal samples were individually collected and analyzed by the coprological techniques. The main strongylid genera identified were Trichonema and Cyalocephalus spp (small strongyles) and Strongylus and Triodontophorus (large strongyles). The prevalence of gastrointestinal nematode was 89% (95% CI 86, 92) and 1% cestoda (0, 2). The percentage of horses with strongyloid parasites was 89% (86, 92), 11% (8, 14) for Parascaris, and 3% (1, 5) for Oxyuris. The highest prevalence for ascariosis was observed in the youngest horses (<3 years), for oxyurosis in the >10 years animals, and for strongylosis in the 3–10 years ones. Females were significantly more parasitized than males. A negative correlation between the age and the egg-excretion of ascarids and strongyles was recorded. The autochthonous and the English Pure Blood horses were the most parasitized. We concluded that the infections by helminths, especially the strongyloids, are significantly common in the region, so that greater importance should be given to this situation

It is unknown, however, whether this technology will be suitable for the treatment of DNA repair deficiency syndromes such as Fanconi anemia (FA), with defects in homology-directed DNA repair. In this study, we used zinc finger nucleases and integrase-defective lentiviral vectors to demonstrate for the first time that FANCA can be efficiently and specifically targeted into the AAVS1 safe harbor locus in fibroblasts from FA-A patients. Strikingly, up to 40% of FA fibroblasts showed gene targeting 42 days after gene editing. Given the low number of hematopoietic precursors in the bone marrow of FA patients, gene-edited FA fibroblasts were then reprogrammed and re-differentiated toward the hematopoietic lineage. Analyses of gene-edited FA-iPSCs confirmed the specific integration of FANCA in the AAVS1 locus in all tested clones. Moreover, the hematopoietic differentiation of these iPSCs efficiently generated disease-free hematopoietic progenitors. Taken together, our results demonstrate for the first time the feasibility of correcting the phenotype of a DNA repair deficiency syndrome using gene-targeting and cell reprogramming strategies

    ResistĂȘncia anti-helmĂ­ntica em nematoides gastrintestinais de pequenos ruminantes: avanços e limitaçÔes para seu diagnĂłstico

    A seleção e a crescente disseminação de nematoides resistentes aos anti-helmĂ­nticos mais comumente utilizados, benzimidazĂłis (BZs), imidazotiazĂłis e lactonas macrocĂ­clicas (LMs), constituem um sĂ©rio entrave na produção de pequenos ruminantes em todo o mundo. O uso de mĂ©todos eficientes e sensĂ­veis para a detecção e o monitoramento da resistĂȘncia anti-helmĂ­ntica no campo torna-se urgente, especialmente para os grupos de BZs e LMs, devido aos constantes relatos de resistĂȘncia. A obtenção de um diagnĂłstico preciso e precoce da resistĂȘncia Ă© extremamente importante para auxiliar a tomada de decisĂŁo em programas de controle parasitĂĄrio, com o objetivo de preservar a vida Ăștil dos produtos e limitar o desenvolvimento da resistĂȘncia nas populaçÔes de nematoides. Os testes in vivo e, mais recentemente, os testes in vitro tĂȘm sido desenvolvidos para a detecção de nematoides resistentes aos principais grupos de anti-helmĂ­nticos. No entanto, a disponibilidade de testes in vitro validados e o seu uso prĂĄtico ainda sĂŁo muito limitados. Embora o teste de redução na contagem de ovos nas fezes (TRCOF, in vivo - indireto) seja o principal mĂ©todo de escolha para a detecção de resistĂȘncia no campo, vem recebendo crĂ­ticas quanto Ă  validade dos resultados, e passa por significativas modificaçÔes. AlĂ©m disso, o desenvolvimento de tĂ©cnicas moleculares a partir de alteraçÔes genĂŽmicas gerou avanços considerĂĄveis nessa ĂĄrea de investigação, com o uso de mutaçÔes nos cĂłdons 167, 198 e 200 do gene da ÎČ-tubulina como principais SNPs (polimorfismos de nucleotĂ­deo Ășnico; do inglĂȘs Single Nucleotide Polymorphisms) associados Ă  resistĂȘncia aos BZs.     The field efficacy of a single dose treatment against natural M. capillaris infection using different anthelmintic drugs, extensively employed in ovine parasite control programs in Galicia (N.W. Spain), and the effect of protostrongylid infection on ovine respiratory functions, were evaluated. Five groups (n = 5) of ewes were used in this study; animals were treated with injectable ivermectin (0.2 mg/kg), levamisole (7.5 mg/kg) and albendazole (5 mg/kg) and monitored at 0, 7, 21, 42 and 63 days posttreatment (d.p.t.) by enumeration of the lungworm larvae per gram of faeces (l.p.g.) and determination of gas tension and pH in venous blood using an i-Stat portable clinical analyzer. No statistical difference was found either in larval elimination between untreated and treated groups or in the reduction in larval counts in all of the treated groups. A significant increase in partial oxygen tension (pO2) and saturation (sO2) between day 0 and 7 p.t. was observed in all treated animals. These values decreased significantly at day 21 to previous levels. There were no statistical differences in blood gases between uninfected and treated groups. We can conclude that under Galician field conditions, parasitic control programs are not totally effective against M. capillaris infection
