    Determinación de autovalores usando el método optimizado de Rayleigh-Ritz y "pseudo" desarrollos de Fourier

    Hace algo más de un siglo Lord Rayleigh sugirió el uso de un parámetro de optimización en las funciones coordenadas polinómicas a ser utilizadas en la aplicación de su actualmente famoso método, teniendo como meta el minimizar el autovalor determinado. Tres décadas después el célebre analista y diseñador de sistemas mecánicos, Stodola, utilizó este enfoque al deteminar la frecuencia fundamental de un álabe. Linus Pauling y Bright-Wilson implementaron el enfoque utilizando funciones exponenciales en problemas de mecánica cuántica y Timoshenko y Goodier también lo hicieron, utilizando también funciones exponenciales, en un problema de torsión de barras elásticas. En este trabajo se presentan aplicaciones del criterio de Rayleigh, pero difiriendo del enfoque de los autores citados, se usan funciones trigonométricas que contienen parámetros de optimización en su argumento, tomando como sistemas mecánicos vibrantes casos de placas o losas rectangulares con un borde libre.Peer Reviewe

    Toxicity characterisation of Gambierdiscus species from the Canary Islands

    In the last decade, several outbreaks of ciguatera fish poisoning (CFP) have been reported in the Canary Islands (central northeast Atlantic Ocean), confirming ciguatera as an emerging alimentary risk in this region. Five Gambierdiscus species, G. australes, G. excentricus, G. silvae, G. carolinianus and G. caribaeus, have been detected in macrophytes from this area and are known to produce the ciguatoxins (CTXs) that cause CFP. A characterization of the toxicity of these species is the first step in identifying locations in the Canary Islands at risk of CFP. Therefore, in this study the toxicity of 63 strains of these five Gambierdiscus species were analysed using the erythrocyte lysis assay to evaluate their maitotoxin (MTX) content. In addition, 20 of the strains were also analysed in a neuroblastoma Neuro-2a (N2a) cytotoxicity assay to determine their CTX-like toxicity. The results allowed the different species to be grouped according to their ratios of CTX-like and MTX-like toxicity. MTX-like toxicity was especially high in G. excentricus and G. australes but much lower in the other species and lowest in G. silvae. CTX-like toxicity was highest in G. excentricus, which produced the toxin in amounts ranging between 128.2 ± 25.68 and 510.6 ± 134.2 fg CTX1B equivalents (eq) cell−1 (mean ± SD). In the other species, CTX concentrations were as follows: G. carolinianus (100.84 ± 18.05 fg CTX1B eq cell−1), G. australes (31.1 ± 0.56 to 107.16 ± 21.88 fg CTX1B eq cell−1), G. silvae (12.19 ± 0.62 to 76.79 ± 4.97 fg CTX1B eq cell−1) and G. caribaeus (<LOD to 90.37 ± 15.89 fg CTX1B eq cell−1). Unlike the similar CTX-like toxicity of G. australes and G. silvae strains from different locations, G. excentricus and G. caribaeus differed considerably according to the origin of the strain.     Fine scale physical-biological interactions during a shift from relaxation to upwelling with a focus on Dinophysis acuminata and its potential ciliate prey

    Wind reversals and quick transitions from relaxation to upwelling in coastal areas cause major changes in water column structure, phytoplankton distribution and dominance, and rates of physiological processes. The cruise "ASIMUTH-Rías" (17–21 June 2013) was carried out in the Galician Rías and adjacent shelf, at the time of a DSP outbreak, to study small-scale physical processes associated with late spring blooms of "D. acuminata" and accompanying microzooplanktonic ciliates with the overall objective of improving predictive models of their occurrence. The cruise coincided with the initiation of an upwelling pulse following relaxation and deepening of a previously formed thin layer of diatoms. A 36-h cell cycle study carried on 18–20 June showed the vertical excursions of the thin layer, mainly delimited by the 13.5–14 °C isotherms and turbulence levels (ε) of 10−8–10−6 m2 s −3, as well as marked changes in phytoplankton composition (increased density and dominance of diatoms). There was no evidence of daily vertical migration of D. acuminata, which remained in the top layer during the cycle study, but the opposite was observed in the ciliate populations. Dinophysis and its potential prey (Mesodinium species) cell maxima overlapped after midday, when the ciliate moved to the surface, suggesting an “ambush” strategy of Dinophysis to catch prey. A remarkable decline (from 0.65 to 0.33 d−1) in division rates (µ) of D. acuminata was associated with increased turbulence (ε 2°C in about 8 h). In contrast, high division rates (µmin ∼ 0.69 d−1) persisted at a mid-shelf station where environmental conditions below the mixed layer were more stable. The onset of upwelling pulses appears to have a double negative effect on the net growth of Dinophysis populations: a direct physical effect due to advective dispersion and an indirect effect, decreased division rates. The latter would be caused by the rapid cooling of the mixed layer, and the increased turbulence at the surface resulting in shear stress to the cells. The short-term impact of upwelling pulses (and the winds promoting it) on the physiology of "Dinophysis" and its ciliate prey, and the role of mid-shelf populations of "Dinophysis" as a relatively undisturbed reservoir for the inoculation of subsequent blooms are discussed.European Commission | Ref.     A Case Report of Acute Severe Myelitis and Meningitis Secondary to Varicella Zoster Virus Reactivation in a Patient with Acquired Immunodeficiency Syndrome

    Background: Myelitis post Herpes-Zoster is a rare condition that is typically associated with immunocompromised states. It usually starts as an acute loss of sensory and motor functions below the affected spinal cord level. The condition can range in severity from a mild to a fatal presentation. Other neurological complications include meningitis, atypical presentations should encourage the search for undiagnosed immunosuppression states. The Case: We describe the case of a 42-year-old man, with previously undiagnosed HIV, who developed acute myelitis and meningitis after the appearance of the classic zoster lesions. On lumbar puncture and subsequent CSF analysis, the patient was found to have Froin's Syndrome. The patient was initiated with ceftriaxone, vancomycin, and acyclovir regimen and prophylactic antiphymic treatment was also added. After 14 days in the hospital, the fever, headache, and neck stiffness subsided while the sphincter function and lower limb paraplegia did not improve. Conclusion: Varicella zoster virus reactivation suggests underlying immunosuppression. This case demonstrates the importance of being cognizant to the wide range of clinical manifestations that may suggest spinal cord involvement after clinical reactivation. Furthermore, physicians also need to be mindful that Acquired Immunodeficiency Syndrome (AIDS) and other immunodeficiency states could present with atypical clinical manifestations

    Caracterización molecular y curvas de temperatura de aislados de Botrytis Caroliniana yb. Cinerea obtenidos desde manzanas con pudrición calicinal en la región del Maule

    44 p.Los manzanos (Malus domestica Borkh.) es un frutal de hoja caduca que presenta una superficie importante con 32.371 ha, siendo la región del Maule la superficie más importante (60%) a nivel nacional. La pudrición calicinal de la manzana es causado predominantemente por Botrytis cinerea, siendo esta enfermedad una de las importantes durante precosecha y poscosecha. Recientemente se han identificado a otras especies de Botrytis causando pudrición gris en diferentes hospederos (pera, ciruelo, kiwis, vid entre otros). Por ello, es crucial realizar diversos tipos de estudios que puedan identificar las posibles especies de Botrytis para determinar de forma más exacta la etiología de la pudrición calicinal de manzanas en la región del Maule. Por lo tanto, el objetivo del presente estudio fue caracterizar molecularmente y estudiar el comportamiento bajo distintas temperaturas de aislados de Botrytis cinerea de alta esporulación y aislados de Botrytis sp. de baja esporulación obtenidos recientemente causando pudrición calicinal en manzanos en la región del Maule. La identificación molecular determino que los aislados con fenotipos de Botrytis sp. de colonias de baja esporulación (Bca-PC-A3; Bca-PC-A13; Bca-TF-A27; Bca-TF-A28; Bca-TF-A30) obtuvieran similitudes entre 99 al 100% con la especie tipo de Botrytis caroliniana con los genes G3PDH, HSP60 y RPB2. De la misma forma los aislados de colonias grises de alta esporulación (Bci-PC-A1, Bci-PC-A6, Bci-TF-A22, Bci-TF-A23 y Bci-TFA29), mostraron una similitud del 100% para los genes G3PDH, HSP60 y RPB2 con espécimen tipo de B. cinerea aislado MUCL-87. El análisis filogenetico identifico los aislados en agrupaciones junto con el ex-type de B. caroliniana y B. cinerea. La temperatura de crecimiento óptimo para los aislados de B. caroliniana fue de 20 °C y para los aislados de B. cinerea fue entre los 15 a 20°C. Finalmente, se identificó a la especie Botrytis caroliniana que mostro diferencias genéticas y fisiológicas diferentes de B. cinere

    Latitudinal Variation in the Toxicity and Sexual Compatibility of Alexandrium catenella Strains from Southern Chile

    The bloom-forming toxic dinoflagellate Alexandrium catenella was first detected in southern Chile (39.5–55° S) 50 years ago and is responsible for most of the area’s cases of paralytic shellfish poisoning (PSP). Given the complex life history of A. catenella, which includes benthic sexual cysts, in this study, we examined the potential link between latitude, toxicity, and sexual compatibility. Nine clones isolated from Chilean Patagonia were used in self- and out-crosses in all possible combinations (n = 45). The effect of latitude on toxicity, reproductive success indexes, and cyst production was also determined. Using the toxin profiles for all strains, consisting of C1, C2, GTX4, GTX1, GTX3, and NeoSTX, a latitudinal gradient was determined for their proportions (%) and content per cell (pg cell−1), with the more toxic strains occurring in the north (−40.6° S). Reproductive success also showed a latitudinal tendency and was lower in the north. None of the self-crosses yielded resting cysts. Rather, the production of resting cysts was highest in pairings of clones separated by distances of 1000–1650 km. Our results contribute to a better understanding of PSP outbreaks in the region and demonstrate the importance of resting cysts in fueling new toxic events. They also provide additional evidence that the introduction of strains from neighboring regions is a cause for concern.En prens

    Modelling the Spatial and Temporal Dynamics of Paralytic Shellfish Toxins (PST) at Different Scales: Implications for Research and Management

    Harmful algal blooms, in particular recurrent blooms of the dinoflagellate Alexandrium catenella, associated with paralytic shellfish poisoning (PSP), frequently limit commercial shellfish harvests, resulting in serious socio-economic consequences. Although the PSP-inducing species that threaten the most vulnerable commercial species of shellfish are very patchy and spatially heterogeneous in their distribution, the spatial and temporal scales of their effects have largely been ignored in monitoring programs and by researchers. In this study, we examined the spatial and temporal dynamics of PSP toxicity in the clam (Ameghinomya antiqua) in two fishing grounds in southern Chile (Ovalada Island and Low Bay). During the summer of 2009, both were affected by an intense toxic bloom of A. catenella (up to 1.1 × 106 cells L-1). Generalized linear models were used to assess the potential influence of different environmental variables on the field detoxification rates of PSP toxins over a period of 12 months. This was achieved using a four parameter exponential decay model to fit and compare field detoxification rates per sampling site. The results show differences in the spatial variability and temporal dynamics of PSP toxicity, given that greater toxicities (+10-fold) and faster detoxification (20% faster) are observed at the Ovalada Island site, the less oceanic zone, and where higher amounts of clam are annually produced.     Insulin glargine affects the expression of Igf-1r, Insr, and Igf-1 genes in colon and liver of diabetic rats

    Objective(s): The mitogenic effect of the analogous insulin glargine is currently under debate since several clinical studies have raised the possibility that insulin glargine treatment has a carcinogenic potential in different tissues. This study aimed to evaluate the Igf-1r, Insr, and Igf-1 gene expression in colon and liver of streptozotocin-induced diabetic rats in response to insulin glargine, neutral protamine Hagedorn (NPH) insulin, and metformin treatments. Materials and Methods: Male Wistar rats were induced during one week with streptozotocin to develop Type 2 Diabetes (T2D) and then randomly distributed into four groups. T2D rats included in the first group received insulin glargine, the second group received NPH insulin, the third group received metformin; finally, untreated T2D rats were included as the control group. All groups were treated for seven days; after the treatment, tissue samples of liver and colon were obtained. Quantitative PCR (qPCR) was performed to analyze the Igf-1r, Insr and Igf-1 gene expression in each tissue sample. Results: The liver tissue showed overexpression of the Insr and Igf-1r genes (P>0.001) in rats treated with insulin glargine in comparison with the control group. Similar results were observed for the Insr gene (P>0.011) in colonic tissue of rats treated with insulin glargine. Conclusion: These observations demonstrate that insulin glargine promote an excess of insulin and IGF-1 receptors in STZ-induced diabetic rats, which could overstimulate the mitogenic signaling pathways

    Snow and ice in the desert: reflections from a decade of connecting cryospheric science with communities in the semiarid Chilean Andes

    Citizen science and related engagement programmes have proliferated in recent years throughout the sciences but have been reasonably limited in the cryospheric sciences. In the semiarid Andes we at the Centro de Estudios Avanzados en Zonas Áridas have developed a range of initiatives together with the wider community and stakeholder institutions to improve our understanding of the role snow and ice play in headwater catchments. In this paper we reflect on ongoing engagement with communities living and working in and near study sites of cryospheric science in northern Chile as a strategy that can both strengthen the research being done and empower local communities
