    Regional spatial analysis of an ethnic region in terms of resources, potential, and capital marketing in the example of the Kashubia region (Poland)

    PURPOSE: To determine the essence of the differences between resource marketing, potential marketing and capital marketing, and to verify them on the example of an ethnic region as recognized as Kashuby in Poland. The detailed considerations were put into the context of an ethnic region (ethnoregion), the area of which often escapes from the rigid framework of administrative divisions, and the organization and management of its space faces many difficulties.DESIGN/METHODOLOGY/APPROACH: The regional spatial analysis used an inductive approach, applying a strategic method of assessing resources, potential and capital - SWOT-TOWS analysis. A strategic position was calculated for each component of the assessment and a separate inference procedure was carried out.FINDINGS: The study shows that there is a significant difference in the evaluation of resources, potential and capital of the ethnic region under study, which translates into the adopted marketing strategies, which also show significant variation.PRACTICAL IMPLICATIONS: The results of the study highlight the more effective alignment of marketing activities to be carried out by resource, potential and capital marketing.ORIGINALITY/VALUE: Previously used approaches have not distinguished separate evaluation of resources, potential and capital. This resulted in the fact that marketing activities, often refer to the promotion of capital, i.e. an already functioning business against the background of selected local resources.     Acid selective pro-dye for cellular compartments

    A novel pro-dye approach for the acid-selective staining of the subcellular compartments for better permeability and selectivity was applied. The designed sensor has suitable physicochemical properties such as a large Stokes shift and a long-lived intracellular fluorescence. The Schiff base fragment was used for the acid-sensitive release of a fluorophore without affecting the overall stability of the biological systems. Due to the presence of an imine bond in its structure and its unique fluorescent properties, it can be presented as a “pro-dye” for acidic structures such as lysosomes. As a result of an imine bond cleavage, a new fluorescent compound is released, whose substantially shifted excitation and emission wavelengths enable a more selective and effective imaging of lysosomes and endosomes. The presented report provides the chemical, physicochemical and optical profiles as well as biological assays and theoretical calculations

    Identifying the cause of a disease and the mechanism responsible for dysfunction in our organism is crucial for determination of an effective treatment pathway. The most primary changes are these at the cellular level. Fluorescent sensors enable us to get insight into a world of biological structures and to study cellular processes essential for proper functioning of our organism. Therefore, this group of compounds has gained considerable attention over the past years as safe tools useful for diagnostics and searching for new therapeutic targets. The main goal of the PhD thesis was synthesis of new fluorophores based on a styrylquinoline scaffold and analysis of their properties. A big part of the research was biological tests which enabled to determine the application potential of the newly synthesized compounds as cellular dyes. The scope of work covered, among others, determination of characteristics of the compounds based on analysis of spectroscopic measurements. Additionally, to gain deeper understanding of spectroscopic nature of the dyes and to explain newly discovered relationships, theoretical calculations was carried out with use of Gaussian09 software and density functional theory (DFT). Moreover, biological experiments were performed to determine cytotoxicity of the compounds towards human colon cancer cell line HCT 116 and normal human dermal fibroblasts (NHDF). The place of dyes accumulation in a cell was determine as well. In the case of the lysosomal dye, the reason of significant change of emitted signal in acidic structures was also explained. Furthermore, an attempt was made to establish the mechanism of zinc ions binding to one of the compounds and to determine processes responsible for activation of the fluorescent response after chelation (CHEF, chelation - enhanced fluorescence). It is worth mentioning that good spectroscopic properties of obtained compounds and successfully performed cellular imaging experiments indicate their commercial potential

    Urinary carboxylic acids (UCAs) in subjects with autism spectrum disorder and their association with bacterial overgrowth

    Abstract In this study, the levels of concentration of carboxylic acids (benzoic acid, p-hydroxybenzoic acid, p-hydroxyphenylacetic acid, and hippuric acid) in the urine of autistic children were investigated and compared. The increased excretion of carboxylic acids is related to excessive bacterial activity in the gut, called bacterial overgrowth, which has been related to autism spectrum disorder (ASD) as an impairment in the gut-brain axis. The investigation was based on the analysis of urine samples obtained from 120 ASD children. To identify and quantify urinary carboxylic acids (UCAs), we applied gas chromatography coupled with mass spectrometry (GC-MS). Additionally, we checked the influence of probiotic supplementation, gender, body mass index (BMI) value and age of children on the level of different selected compounds. Most of the obtained results were found within reference ranges. In some cases, the levels of benzoic acid, p-hydroxybenzoic acid, and p-hydroxyphenylacetic acid were particularly elevated. Statistically significant differences were observed in supplementation with probiotics and the level of p-hydroxyphenylacetic acid (p=0.036). The obtained results may indicate disturbances in the intestinal flora in some autistic patients and suggest that supplements may have an influence on the levels of carboxylic acids in urine. Due to the small population of children taking the supplement, further study are needed
