    SpxA1 and SpxA2 act coordinately to fine-tune stress responses and virulence in Streptococcus pyogenes

    SpxA is a unique transcriptional regulator highly conserved among members of the phylum Firmicutes that binds RNA polymerase and can act as an antiactivator. Why some Firmicutes members have two highly similar SpxA paralogs is not understood. Here, we show that the SpxA paralogs of the pathogen Streptococcus pyogenes, SpxA1 and SpxA2, act coordinately to regulate virulence by fine-tuning toxin expression and stress resistance. Construction and analysis of mutants revealed that SpxA1− mutants were defective for growth under aerobic conditions, while SpxA2− mutants had severely attenuated responses to multiple stresses, including thermal and oxidative stresses. SpxA1− mutants had enhanced resistance to the cationic antimicrobial molecule polymyxin B, while SpxA2− mutants were more sensitive. In a murine model of soft tissue infection, a SpxA1− mutant was highly attenuated. In contrast, the highly stress-sensitive SpxA2− mutant was hypervirulent, exhibiting more extensive tissue damage and a greater bacterial burden than the wild-type strain. SpxA1− attenuation was associated with reduced expression of several toxins, including the SpeB cysteine protease. In contrast, SpxA2− hypervirulence correlated with toxin overexpression and could be suppressed to wild-type levels by deletion of speB. These data show that SpxA1 and SpxA2 have opposing roles in virulence and stress resistance, suggesting that they act coordinately to fine-tune toxin expression in response to stress. SpxA2− hypervirulence also shows that stress resistance is not always essential for S. pyogenes pathogenesis in soft tissue

    Heavily obscured AGN in the local Universe

    We present here a new powerful diagnostic plot to select heavily obscured AGN in the local universe by combining infrared (Spitzer, IRAS) and X-ray (XMM) information.     The Palermo Swift-BAT hard X-ray catalogue III. Results after 54 months of sky survey

    We present the Second Palermo Swift-BAT hard X-ray catalogue obtained by analysing data acquired in the first 54 months of the Swift mission. Using our software dedicated to the analysis of data from coded mask telescopes, we analysed the BAT survey data in three energy bands (15-30 keV, 15-70 keV, 15-150 keV), obtaining a list of 1256 detections above a significance threshold of 4.8 standard deviations. The identification of the source counterparts is pursued using two strategies: the analysis of field observations of soft X-ray instruments and cross-correlation of our catalogue with source databases.The survey covers 50% of the sky to a 15--150 keV flux limit of 1.0 x 10^-11 erg s^-1 cm^-2 and 9.2 x 10^-12 erg s^-1 cm^-2 for |b| 10 degrees, respectively. The Second Palermo Swift-BAT hard X-ray catalogue includes 1079 (86%) hard X-ray sources with an associated counterpart (26 with a double association and 2 with a triple association) and 177 BAT excesses (14%) that still lack a counterpart. The distribution of the BAT sources among the different object classes consists of 19% Galactic sources, 57% extragalactic sources, and 10% sources with a counterpart at softer energies whose nature has not yet been determined. About half of the BAT associated sources lack a counterpart in the ROSAT catalogues. This suggests that either moderate or strong absorption may be preventing their detection in the ROSAT energy band. The comparison of our BAT catalogue with the Fermi Large Area Telescope First Source Catalogue identifies 59 BAT/Fermi correspondences: 48 blazars, 3 Seyfert galaxies, 1 interacting galaxy, 3 high mass X-ray binaries, and 4 pulsars/supernova remnants. This small number of correspondences indicates that different populations make the sky shine in these two different energy bands

    A semi-automatic system for labelling seafood products and obtaining fishery management data: A case study of the bottom trawl fishery in the central Mediterranean Sea

    This study is on the implementation of a semi-automatic labelling system (LS) of the Mediterranean Sea seafood harvest to address the increased need for seafood authentication and inherent difficulties of commonly used indirect techniques for estimating fisheries yield and fishing effort. Sensitive datarequired for anti-counterfeit policies, such as date and catch area, can be acquired and recorded on the label by user-friendly automated software that excludes any possible manipulation by the crew. Based on results obtained from the installation of the LS on bottom commercial trawlers, the system certified the origin of the seafood products and simultaneously provided, indirectly, geospatial fisheries yield and fishing effort data of the main exploited species

    Testing the gamma-ray burst variability/peak luminosity correlation on a Swift homogeneous sample

    We test the gamma-ray burst correlation between temporal variability and peak luminosity of the γ\gamma-ray profile on a homogeneous sample of 36 Swift/BAT GRBs with firm redshift determination. This is the first time that this correlation can be tested on a homogeneous data sample. The correlation is confirmed, as long as the 6 GRBs with low luminosity (<5x10^{50} erg s^{-1} in the rest-frame 100-1000 keV energy band) are ignored. We confirm that the considerable scatter of the correlation already known is not due to the combination of data from different instruments with different energy bands, but it is intrinsic to the correlation itself. Thanks to the unprecedented sensitivity of Swift/BAT, the variability/peak luminosity correlation is tested on low-luminosity GRBs.     As the possibility to decouple temporal and spatial variations of the electromagnetic field, leading to a wavelength stretching, has been recognized to be of paramount importance for practical applications, we generalize the idea of stretchability from the framework of electromagnetic waves to massive particles. A necessary and sufficient condition which allows one to identify energetically stable configuration of a 1D quantum particle characterized by arbitrary large spatial regions where the associated wave-function exhibit a flat, non-zero profile is presented, together with examples on well-known and widely used potential profiles and an application to 2D models

    The Crab pulsar light curve in the soft gamma ray range: FIGARO II results

    The FIGARO II experiment (a large area, balloon borne, crystal scintillator detector working from 0.15 to 4.3 MeV) observed the Crab pulsar on 1990 Jul. 9 for about seven hours. The study of the pulse profile confirms some structures detected with a low significance during the shorter observation of 1986, and adds new important elements to the picture. In particular, between the two main peaks, two secondary peaks appear centered at phase values 0.1 and 0.3, in the energy range 0.38 to 0.49 MeV; in the same energy range, a spectral feature at 0.44 MeV, interpreted as a redshifted positron annihilation line, was observed during the same balloon flight in the phase interval including the second main peak and the neighboring secondary peak. If the phase interval considered is extended to include also the other secondary peak, the significance of the spectral line appears to increase
