    Translating COGNISTAT and the Use of the Cognitive Interview Approach: Observations and Challenges

    Cognitive functioning is a salient issue among people in the late adulthood stage where mental health declines with age. A common cognitive disability among elders is Dementia including Alzheimer’s disease. Cognitive screening tools such as the Mini- Mental Status Examination and the Montreal Cognitive Assessment are most commonly used to measure the cognitive ability areas leading to a diagnostic evaluation. COGNISTAT as a neuropsychological instrument is a recent screener being introduced to a few outpatient clinics. In this study; COGNISTAT is translated to Filipino for the first time using a rigorous procedure and pilot tested on elderly volunteers in local settings. The translation was done by an interdisciplinary team of a Geriatric Physician; a Speech Pathologist; and a Clinical Psychologist. The Filipino-translated COGNISTAT was administered to 22 elders in two batches using the cognitive interview method. Challenges were identified in the initial testing phase which has implications for future adaptations of foreign instruments

    Diagnóstico energético del sistema de circulación de agua fría en buques tipo nodriza fluvial

    The air conditioning system of a riverine patrol and supply type ship constructed by Cotecmar is constituted by a cold water circulation circuit through which centrifugal pump supplies 21 fan coils, by means of which the parameters of human comfort are guaranteed. In this article, the Power Diagnosis of the system is presented with the aim of establishing the possible opportunities for saving on the ships, calculating the losses in the system and determining the equation and the equivalent curve with which the “pump-system” set’s point of operation was found. The result of the diagnosis shows that most of the losses that take place are due to pipes, accessories and fan coils, reason why a group of measures oriented towards the reduction of power consumption are proposed and that are reflected in a better operation and in the advantage taken of the resources with which one counts upon.El sistema de aire acondicionado de un buque tipo Nodriza Fluvial construido por Cotecmar está constituido por un circuito de circulación de agua fría a través del cual una bomba centrífuga abastece a 21 fan coils, mediante los cuales se garantizan los parámetros de confort humano. En este artículo se presenta el diagnóstico energético del sistema con el objetivo de establecer las posibles oportunidades de ahorro en el buque, calculando las pérdidas en el sistema y determinándose la ecuación y la curva equivalente con las cuales se encontró el punto de operación del conjunto “bomba-sistema”. El resultado del diagnóstico arroja que las mayores pérdidas son debidas a tuberías, accesorios y fan coils, por lo que se propone un grupo de medidas encaminadas a la reducción del consumo energético que se vean reflejadas en una mejor operación y aprovechamiento de los recursos con que se cuenta

    A novel form of deleted in breast cancer 1 (DBC1) lacking the N-terminal domain does not bind SIRT1 and is dynamically regulated in vivo

    The protein Deleted in Breast Cancer-1 is a regulator of several transcription factors and epigenetic regulators, including HDAC3, Rev-erb-alpha, PARP1 and SIRT1. It is well known that DBC1 regulates its targets, including SIRT1, by protein-protein interaction. However, little is known about how DBC1 biological activity is regulated. In this work, we show that in quiescent cells DBC1 is proteolytically cleaved, producing a protein (DN-DBC1) that misses the S1-like domain and no longer binds to SIRT1. DN-DBC1 is also found in vivo in mouse and human tissues. Interestingly, DN-DBC1 is cleared once quiescent cells re-enter to the cell cycle. Using a model of liver regeneration after partial hepatectomy, we found that DN-DBC1 is down-regulated in vivo during regeneration. In fact, WT mice show a decrease in SIRT1 activity during liver regeneration, coincidentally with DN-DBC1 downregulation and the appearance of full length DBC1. This effect on SIRT1 activity was not observed in DBC1 KO mice. Finally, we found that DBC1 KO mice have altered cell cycle progression and liver regeneration after partial hepatectomy, suggesting that DBC1/DN-DBC1 transitions play a role in normal cell cycle progression in vivo after cells leave quiescence. We propose that quiescent cells express DN-DBC1, which either replaces or coexist with the full-length protein, and that restoring of DBC1 is required for normal cell cycle progression in vitro and in vivo. Our results describe for the first time in vivo a naturally occurring form of DBC1, which does not bind SIRT1 and is dynamically regulated, thus contributing to redefine the knowledge about its function

    Multivalent Tryptophan‐ and Tyrosine‐Containing [60]Fullerene Hexa‐Adducts as Dual HIV and Enterovirus A71 Entry Inhibitors

    Unprecedented 3D hexa-adducts of [60]fullerene peripherally decorated with twelve tryptophan (Trp) or tyrosine (Tyr) residues have been synthesized. Studies on the antiviral activity of these novel compounds against HIV and EV71 reveal that they are much more potent against HIV and equally active against EV71 than the previously described dendrimer prototypes AL-385 and AL-463, which possess the same number of Trp/Tyr residues on the periphery but attached to a smaller and more flexible pentaerythritol core. These results demonstrate the relevance of the globular 3D presentation of the peripheral groups (Trp/Tyr) as well as the length of the spacer connecting them to the central core to interact with the viral envelopes, particularly in the case of HIV, and support the hypothesis that [60]fullerene can be an alternative and attractive biocompatible carbon-based scaffold for this type of highly symmetrical dendrimers. In addition, the functionalized fullerenes here described, which display twelve peripheral negatively charged indole moieties on their globular surface, define a new and versatile class of compounds with a promising potential in biomedical applications

    High lipid content of oocytes and embryos in domestic animals is one of the well-known factors associated with poor cryosurvival. Herein, we wanted to determine whether the use of delipidated estrous sheep serum during in vitro maturation (IVM) of ovine oocytes reduces the cytoplasmic lipid droplets content and improves embryo development and cryotolerance after vitrification. Cumulus oocytes complexes (COCs) were matured in vitro for 24 h in medium supplemented with whole or delipidated estrous sheep serum prior to vitrification. Neutral lipid present in lipid droplets of COCs, cleavage rate, embryo development rate on Day 6 and Day 8, and hatching rate on Day 8, were compared among experimental groups. Endoplasmic reticulum stress genes were evaluated in in vitro matured COCs under different lipid conditions prior to vitrification. The lipid droplets' content (mean fluorescence intensity) of oocytes cultured with IVM media supplemented with delipidated serum was lower than COCs matured with whole serum (7.6 ± 1.7 vs. 22.8 ± 5.0 arbitrary units, respectively; P< 0.05). Despite IVM treatment, oocytes subjected to vitrification showed impaired competence compared with the non-vitrified groups (P<0.05). No significant differences in embryo production were observed in non-vitrified COCs after maturation in delipidated or whole serum (33.4±4.9 vs 31.9 ±4.2). COCs matured in delipidated serum and subjected to vitrification showed increased expression of ATF4, ATF6, GRP78, and CHOP10 genes (ER stress markers). Collectively, our results demonstrate that although supplementation of IVM medium with delipidated estrous sheep serum reduces the presence of cytoplasmic lipid droplets in oocytes after maturation, oocyte cryotolerance is not improved. Notably, the expression of genes associated with the unfolded protein response (UPR) was increased in COCs, with fewer lipid droplets subjected to vitrification, suggesting that oocyte cryopreservation is associated with ER stress and activation of adaptive responses
