    Impact of wildfire on interdune ecology and sediments: an example from the Simpson Desert, Australia

    The stability of many sand dunes and their interdunes is dependent on vegetation and surface crust cover. When this cover is removed, the sand can be activated and fine sediments deflated making the dunefields into sources of dust. This paper reports the impact of devegetation by wildfire on an interdune in the Simpson Desert, Australia. The fire occurred in 2001 and six years after the event pronounced differences between a pair of burnt and unburnt sites was clearly discernible. The variables examined included vegetation assemblage, cyanobacteria abundance and sediment aggregation, particle-size distribution and colour; but whether they apply to all such situations is uncertain. Rate of recovery has been slow and the differences are likely to have been sustained by a combination of negative feedback processes and climate

    Epigenomic and transcriptomic signatures of a Klinefelter syndrome (47,XXY) karyotype in the brain

    Klinefelter syndrome (KS) is the most common sex-chromosome aneuploidy in humans. Most affected individuals carry one extra X-chromosome (47,XXY karyotype) and the condition presents with a heterogeneous mix of reproductive, physical and psychiatric phenotypes. Although the mechanism(s) by which the supernumerary X-chromosome determines these features of KS are poorly understood, skewed X-chromosome inactivation (XCI), gene-dosage dysregulation, and the parental origin of the extra X-chromosome have all been implicated, suggesting an important role for epigenetic processes. We assessed genomic, methylomic and transcriptomic variation in matched prefrontal cortex and cerebellum samples identifying an individual with a 47,XXY karyotype who was comorbid for schizophrenia and had a notably reduced cerebellum mass compared with other individuals in the study (n = 49). We examined methylomic and transcriptomic differences in this individual relative to female and male samples with 46,XX or 46,XY karyotypes, respectively, and identified numerous locus-specific differences in DNA methylation and gene expression, with many differences being autosomal and tissue-specific. Furthermore, global DNA methylation, assessed via the interrogation of LINE-1 and Alu repetitive elements, was significantly altered in the 47,XXY patient in a tissue-specific manner with extreme hypomethylation detected in the prefrontal cortex and extreme hypermethylation in the cerebellum. This study provides the first detailed molecular characterization of the prefrontal cortex and cerebellum from an individual with a 47,XXY karyotype, identifying widespread tissue-specific epigenomic and transcriptomic alterations in the brain. © 2014 Landes Bioscience

    Somatosensory dysfunction is masked by variable cognitive deficits across patients on the Alzheimer’s disease spectrum

    Background: Alzheimer’s disease (AD) is generally thought to spare primary sensory function; however, such interpretations have drawn from a literature that has rarely taken into account the variable cognitive declines seen in patients with AD. As these cognitive domains are now known to modulate cortical somato-sensory processing, it remains possible that abnormalities in somatosensory function in patients with AD have been suppressed by neuropsychological variability in previous research. Methods: In this study, we combine magnetoencephalographic (MEG) brain imaging during a paired-pulse somatosensory gating task with an extensive battery of neuropsychological tests to investigate the influence of cognitive variability on estimated differences in somatosensory function between biomarker-confirmed patients on the AD spectrum and cognitively-normal older adults. Findings: We show that patients on the AD spectrum exhibit largely non-significant differences in somato-sensory function when cognitive variability is not considered (p-value range: .020-.842). However, once attention and processing speed abilities are considered, robust differences in gamma-frequency somatosensory response amplitude (p \u3c .001) and gating (p = .004) emerge, accompanied by significant statistical suppression effects. Interpretation: These findings suggest that patients with AD exhibit insults to functional somatosensory processing in primary sensory cortices, but these effects are masked by variability in cognitive decline across individuals

    Bacteriophages are viruses that infect bacteria. There are an estimated 1031 phage on the planet, making them the most abundant form of life. We are rapidly approaching the centenary of their identification, and yet still have only a limited understanding of their role in the ecology and evolution of bacterial populations. Temperate prophage carriage is often associated with increased bacterial virulence. The rise in use of technologies, such as genome sequencing and transcriptomics, has highlighted more subtle ways in which prophages contribute to pathogenicity. This review discusses the current knowledge of the multifaceted effects that phage can exert on their hosts and how this may contribute to bacterial adaptation during infection

    The Sundowner Winds Experiment (SWEX) pilot study: Understanding downslope windstorms in the Santa Ynez Mountains, Santa Barbara, California

    Sundowner winds are downslope gusty winds often observed on the southern slopes of the Santa Ynez Mountains (SYM) in coastal Santa Barbara (SB), California. They typically peak near sunset and exhibit characteristics of downslope windstorms through the evening. They are SB\u27s most critical fire weather in all seasons and represent a major hazard for aviation. The Sundowner Winds Experiment Pilot Study was designed to evaluate vertical profiles of winds, temperature, humidity, and stability leeward of the SYM during a Sundowner event. This was accomplished by launching 3-hourly radiosondes during a significant Sundowner event on 28-29 April 2018. This study showed that winds in the lee of the SYM exhibit complex spatial and temporal patterns. Vertical profiles showed a transition from humid onshore winds from morning to mid-afternoon to very pronounced offshore winds during the evening after sunset. These winds accompanied mountain waves and a northerly nocturnal lee jet with variable temporal behavior. Around sunset, the jet was characterized by strong wind speeds enhanced by mountain-wave breaking. Winds weakened considerably at 2300 PDT 29 April but enhanced dramatically at 0200 PDT 29 April at much lower elevations. These transitions were accompanied by changes in stability profiles and in the Richardson number. A simulation with the Weather Research and Forecasting (WRF) Model at 1-km grid spacing was examined to evaluate the skill of the model in capturing the observed winds and stability profiles and to assess mesoscale processes associated with this event.     'Pedometers cost buttons': the feasibility of implementing a pedometer based walking programme within the community

    Background: Recent studies have suggested that walking interventions may be effective (at least in the short term) at increasing physical activity amongst those people who are the most inactive. This is a leading objective of contemporary public health policy in the UK and worldwide. However, before committing money from limited budgets to implement walking interventions more widely in the community, policymakers will want to know whether similar impacts can be expected and whether any changes will be required to the process to ensure uptake and success. This paper utilises the findings from a recent community-based pedometer study (Walking for Wellbeing in the West - WWW) undertaken in Glasgow, Scotland to address issues of feasibility. Methods: An economic analysis of the WWW study assessed the costs of the interventions (minimal and maximal) and combined these with the effects to present incremental cost-effectiveness ratios (cost/person achieving the target of an additional 15,000 steps/week). A qualitative evaluation, involving focus group discussions with WWW participants and short interviews with members of the WWW research team, explored perceived benefits and barriers associated with walking, as well as the successful aspects and challenges associated with the interventions. Results: The incremental cost effectiveness associated with the interventions was estimated as (sic)92 and (sic)591 per person achieving the target for the minimal and maximal interventions respectively. The qualitative evaluation gave insight into the process by which the results were achieved, and identified several barriers and facilitators that would need to be addressed before implementing the interventions in the wider community, in order to ensure their effective transfer. These included assessing the impact of the relationship between researchers and participants on the results, and the motivational importance of monitoring and assessing performance. Conclusions: The results suggest that pedometer based walking interventions may be considered cost-effective and suitable for implementation within the wider community. However, several research gaps remain, including the importance and impact of the researcher/participant relationship, the impact of assessment on motivation and effectiveness, and the longer term impact on physical and mental health, resource utilisation and quality of life
