    Regulatory activity revealed by dynamic correlations in gene expression noise

    Gene regulatory interactions are context dependent, active in some cellular states but not in others. Stochastic fluctuations, or 'noise', in gene expression propagate through active, but not inactive, regulatory links^(1,2). Thus, correlations in gene expression noise could provide a noninvasive means to probe the activity states of regulatory links. However, global, 'extrinsic', noise sources generate correlations even without direct regulatory links. Here we show that single-cell time-lapse microscopy, by revealing time lags due to regulation, can discriminate between active regulatory connections and extrinsic noise. We demonstrate this principle mathematically, using stochastic modeling, and experimentally, using simple synthetic gene circuits. We then use this approach to analyze dynamic noise correlations in the galactose metabolism genes of Escherichia coli. We find that the CRP-GalS-GalE feed-forward loop is inactive in standard conditions but can become active in a GalR mutant. These results show how noise can help analyze the context dependence of regulatory interactions in endogenous gene circuits

    A FLAMINGOS Deep Near Infrared Imaging Survey of the Rosette Complex I: Identification and Distribution of the Embedded Population

    We present the results of a deep near-infrared imaging survey of the Rosette Complex. We studied the distribution of young embedded sources using a variation of the Nearest Neighbor Method applied to a carefully selected sample of near-infrared excess (NIRX) stars which trace the latest episode of star formation in the complex. Our analysis confirmed the existence of seven clusters previously detected in the molecular cloud, and identified four more clusters across the complex. We determined that 60% of the young stars in the complex and 86% of the stars within the molecular cloud are contained in clusters, implying that the majority of stars in the Rosette formed in embedded clusters. We compare the sizes, infrared excess fractions and average extinction towards individual clusters to investigate their early evolution and expansion. We found that the average infrared excess fraction of clusters increases as a function of distance from NGC 2244, implying a temporal sequence of star formation across the complex. This sequence appears to be primordial, possibly resulting from the formation and evolution of the molecular cloud and not from the interaction with the HII region.Comment: Accepted by Astrophysical Journa

    Halocarbon ozone depletion and global warming potentials

    Concern over the global environmental consequences of fully halogenated chlorofluorocarbons (CFCs) has created a need to determine the potential impacts of other halogenated organic compounds on stratospheric ozone and climate. The CFCs, which do not contain an H atom, are not oxidized or photolyzed in the troposphere. These compounds are transported into the stratosphere where they decompose and can lead to chlorine catalyzed ozone depletion. The hydrochlorofluorocarbons (HCFCs or HFCs), in particular those proposed as substitutes for CFCs, contain at least one hydrogen atom in the molecule, which confers on these compounds a much greater sensitivity toward oxidation by hydroxyl radicals in the troposphere, resulting in much shorter atmospheric lifetimes than CFCs, and consequently lower potential for depleting ozone. The available information is reviewed which relates to the lifetime of these compounds (HCFCs and HFCs) in the troposphere, and up-to-date assessments are reported of the potential relative effects of CFCs, HCFCs, HFCs, and halons on stratospheric ozone and global climate (through 'greenhouse' global warming)

    Temperature dependent absorption cross-sections of HNO3 and N2O5

    Absorption cross-sections for HNO3 and N2O5 have been measured in the wavelength region 220-450 nm, using a dual beam diode array spectrometer with a spectral resolution of 0.3 nm. The results for both compounds are in good agreement with recommended values at room temperature. However, the cross-sections of both HNO3 and N2O5 show a marked reduction with decreasing temperature in the range 295-233 K. The calculated photolysis rate of HNO3 at the low temperatures and high solar zenith angles characteristic of the polar winter and spring is significantly lower than previously estimated

    Stratospheric processes: Observations and interpretation

    Explaining the observed ozone trends discussed in an earlier update and predicting future trends requires an understanding of the stratospheric processes that affect ozone. Stratospheric processes occur on both large and small spatial scales and over both long and short periods of time. Because these diverse processes interact with each other, only in rare cases can individual processes be studied by direct observation. Generally the cause and effect relationships for ozone changes were established by comparisons between observations and model simulations. Increasingly, these comparisons rely on the developing, observed relationships among trace gases and dynamical quantities to initialize and constrain the simulations. The goal of this discussion of stratospheric processes is to describe the causes for the observed ozone trends as they are currently understood. At present, we understand with considerable confidence the stratospheric processes responsible for the Antarctic ozone hole but are only beginning to understand the causes of the ozone trends at middle latitudes. Even though the causes of the ozone trends at middle latitudes were not clearly determined, it is likely that they, just as those over Antarctica, involved chlorine and bromine chemistry that was enhanced by heterogeneous processes. This discussion generally presents only an update of the observations that have occurred for stratospheric processes since the last assessment (World Meteorological Organization (WMO), 1990), and is not a complete review of all the new information about stratospheric processes. It begins with an update of the previous assessment of polar stratospheres (WMO, 1990), followed by a discussion on the possible causes for the ozone trends at middle latitudes and on the effects of bromine and of volcanoes

A distinguished group of editors and experts who were already engaged in one way or another with the issues inherent in making research data public came together with statisticians to initiate a dialogue about policies and practicalities of requiring published research to be accompanied by publication of the research data.     ALMA CN Zeeman Observations of AS 209: Limits on Magnetic Field Strength and Magnetically Driven Accretion Rate

    While magnetic fields likely play an important role in driving the evolution of protoplanetary disks through angular momentum transport, observational evidence of magnetic fields has only been found in a small number of disks. Although dust continuum linear polarization has been detected in an increasing number of disks, its pattern is more consistent with that from dust scattering than from magnetically aligned grains in the vast majority of cases. Continuum linear polarization from dust grains aligned to a magnetic field can reveal information about the magnetic field´s direction, but not its strength. On the other hand, observations of circular polarization in molecular lines produced by Zeeman splitting offer a direct measure of the line-of-sight magnetic field strength in disks. We present upper limits on the net toroidal and vertical magnetic field strengths in the protoplanetary disk AS 209 derived from Zeeman splitting observations of the CN 2-1 line. The 3σ upper limit on the net line-of-sight magnetic field strength in AS 209 is 5.0 mG on the redshifted side of the disk and 4.2 mG on the blueshifted side of the disk. Given the disk´s inclination angle, we set a 3σ upper limit on the net toroidal magnetic field strength of 8.7 and 7.3 mG for the red and blue sides of the disk, respectively, and 6.2 and 5.2 mG on the net vertical magnetic field on the red and blue sides of the disk. If magnetic disk winds are a significant mechanism of angular momentum transport in the disk, magnetic fields of a strength close to the upper limits would be sufficient to drive accretion at the rate previously inferred for regions near the protostar.Fil: Harrison, Rachel.     NON-PHARMACEUTICAL TREATMENT OF DEPRESSION USING A MULTIMODAL APPROACH

    One hundred forty-one individuals suffering from chronic depression, unresponsive to previous drug therapy, were treated with a 44-hour program of education, Cranial Electrical Stimulation (CES), Brain Wave Synchronization (BWS), musical conditioning, and a mentally programmed quartz or glass "crystal" randomly assigned with therapists and patients blinded to the crystal's composition. Eighty· four percent of the depressed patients were improved at the end of two weeks of therapy, apparently as a result of the multimodal therapy and group interaction. The results at three months follow-up suggest a positive subtle energy effect of quartz: 70% of the depressed patients who received quartz remained improved, while only 31.5% of the depressed patients receiving glass remained improved. These differences are highly statistically significant. It appears that mentally "programmed" quartz may offer a significant reinforcement to allow patients better long-term recovery than would occur with placebo (glass). The cost effectiveness of such a therapeutic approach is significant. Other therapists are encouraged to replicate these studies
