    Enhancement of antimicrobial activity of antibiotics and antifungals by the use of natural products from Pityrogramma calomelanos (L.) link

    The ethanol extract and methanol fraction of Pityrogramma calomelanos (L.) link were evaluated for antibacterial, antifungal and modulatory activities against strains of Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Candida albicans, C. krusei and C. tropicalis. The antimicrobial activity of the natural products was evaluated by the microdilution method associated or not with aminoglycosides and antifungals. The ethanol extract and methanol fraction of P. calomelanos showed good activity against S. aureus when associated with aminoglycosides and with benzoilmetronidazol against species of the genus Candida. These results indicate that P. calomelanos should be studied as a possible source of natural products to combat bacteria and fungi either directly or by modulating the mechanisms of resistance of these microorganisms, enhancing the antimicrobial activity of these drugs and combating microbial resistance

    Escherichia coli is known to produce enterotoxins whose properties and its role in diarrheal disease has been extensively investigated. Some species of Staphylococcus are often recognized as etiological agents of many animal and human opportunistic infections. This study is the first test of change in resistance of antibiotic activity by Cordia verbenacea DC. against multiresistant strains of Escherichia coli and Staphylococcus aureus. In this study, the hexane and methanol extract of Cordia verbenacea DC. were tested for antibacterial activity alone and in combination with aminoglycosides against bacterial strains. The synergy of the methanolic and hexane were verified by microdilution method. A synergistic effect of both extracts combined with the aminoglycosides was demonstrated. It is therefore suggested that the extracts from Cordia verbenacea DC. could be used as a source of natural products derived from this plant with resistance-modifying antibacterial activity, providing a new weapon against the problem of bacterial resistance to antibiotics.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    Fungos dermatofíticos são patógenos oportunistas, cujos agentes etiológicos pertencem aos gêneros Trichophyton, Microsporum e Epidermophyton. O gênero Trichophyton é composto por diversas espécies, dentre as quais Trichophyton rubrum se destaca pela frequência com que é isolado, podendo produzir praticamente todos os quadros clínicos de dermatofitose. Suas características principais são a tendência à cronicidade e a resistência aos tratamentos convencionais. Este estudo foi realizado com o objetivo de avaliar o efeito antifúngico através da técnica de diluição em HIA de extratos de Croton campestris A., Ocimum gratissimum L. e produtos naturais de Rhinella jimi. Os extratos de C. campestris e O. gratissimum demonstraram atividade antifúngica, assim como os extratos, frações e solução de R. jimi, com exceção da fração hexânica do extrato metanólico. Estes resultados indicam que os organismos testados devem ser estudados como uma possível fonte de produtos naturais com atividade antifúngica, principalmente contra dermatófitos

    Analysis of bioactivities and chemical composition of Ziziphus joazeiro Mart. using HPLC–DAD

    AbstractThe aim of this study was to evaluate the chemical profile and antioxidant, antimicrobial and antiparasitic activities of the hydroalcoholic extract of the leaves of Ziziphus joazeiro Mart. (HELZJ). The antioxidant DPPH and FRAP assays and chemical profile were determined by colorimetric methods and HPLC/DAD. The antiparasitic, antibiotic and antibiotic-modifying activity were evaluated by microdilution assays. The HPLC–DAD assay showed the presence of mostly tannins and flavonoids, such as caffeic acid and quercetin. The levels of polyphenols and flavonoids were 183.136mg/g extract and 7.37mg/g extract, respectively. DPPH and FRAP showed low antioxidant activity for the extract. The antibacterial and antifungal activities were not of clinical relevance, showing MIC>1024μg/mL. However, synergism was observed between HELZJ and the antibiotics amikacin and gentamicin, which resulted in decreased bacterial drug resistance. EHFZJ showed low toxicity in fibroblasts in vitro, while antiparasitic results against Trypnosoma cruzi, Leishmania braziliensis and Leishmania infantum were not clinically relevant. Thus, our results indicate that Z. joazeiro Mart. (HELZJ) could be a source of plant-derived natural products that could lead to the development of promising new antibiotic compounds for infectious diseases

    Chemical composition and toxicity of essential oils of Piper spp. against larvae of Aedes aegypti L. (Diptera: Culicidae)

    Aedes aegypti L. é um dos vetores do dengue. No Brasil, tem ganhado muita atenção no setor da saúde pública, uma vez que esta doença tem se tornando mais agressiva na forma hemorrágica na popula- ção. Esse estudo teve como objetivo investigar o efeito de óleos essenciais de Piper aduncum, Piper marginatum e Piper nigrum contra larvas de Aedes aegypti. Em um esforço para encontrar uma maneira natural, eficaz e acessível para controlar esta doença endêmica, as atividades dos óleos essenciais, a partir das plantas, foram analisadas por comparação através da medida da CL50 . Os óleos essenciais obtidos por hidrodestilação foram analisados por CG/EM. Os principais componentes identificados foram: β-pineno (32,7%) e E-cariofileno (17,1%) em P. aduncum; isoelemecina (21,7%) e apiol (20,1%) em P. marginatum e E-cariofileno (24,2%) e Óxido-cariofileno (20,1%) em P. nigrum. Os resultados mostram que Piper marginatum apresentou CL50 de 8,29 μg/mL e este trabalho é o primeiro relato de atividade larvicida de P. aduncum. Estes resultados sugerem que os óleos essenciais de espécies do gênero Piper são promissores como larvicidas contra larvas de A. aegypti.Aedes aegypti L. is one of the vectors of dengue fever. In Brazil it has gain much attention in the Public Health sector since this disease has becoming more aggressive in the hemorrhagic form in the population. This study aimed to investigate the effect of Piper aduncum, Piper marginatum and Piper nigrum essential oils against Aedes aegypti larvae. In an effort to find a natural effective and affordable way to control this endemic disease, the larvicidal activities of essential oils from the plants were analyzed for activity comparison by measurement of their LC50 . The essential oils isolated by hydrodistillation were analyzed by GC/MS. The main components identified were: β-pinene (32.7%) and E-caryophyllene (17.1%) in P. aduncum; isoelemecin (21.7%) and apiole (20.1%) in P. marginatum and E-caryophyllene (24.2%) and caryophyllene oxide (20.1%) in P. nigrum. The results show that Piper marginatum presented the LC50 of 8.29 μg/mL and these are the first report about the larvicidal activity of P. aduncum.
