    Fracture initiation in multi-phase materials: a systematic three-dimensional approach using a FFT-based solver

    This paper studies a two-phase material with a microstructure composed of a hard brittle reinforcement phase embedded in a soft ductile matrix. It addresses the full three-dimensional nature of the microstructure and macroscopic deformation. A large ensemble of periodic microstructures is used, whereby the individual grains of the two phases are modeled using equi-sized cubes. A particular solution strategy relying on the Fast Fourier Transform is adopted, which has a high computational efficiency both in terms of speed and memory footprint, thus enabling a statistically meaningful analysis. This solution method naturally accompanies the regular microstructural model, as the Fast Fourier Transform relies on a regular grid. Using the many considered microstructures as an ensemble, the average arrangement of phases around fracture initiation sites is objectively identified by the correlation between microstructure and fracture initiation -- in three dimensions. The results show that fracture initiates where regions of the hard phase are interrupted by bands of the soft phase that are aligned with the direction of maximum shear. In such regions, the hard phase is arranged such that the area of the phase boundary perpendicular to the principal strain direction is maximum, leading to high hydrostatic tensile stresses, while not interrupting the shear bands that form in the soft phase. The local incompatibility that is present around the shear bands is responsible for a high plastic strain. By comparing the response to a two-dimensional microstructure it is observed that the response is qualitatively similar (both macroscopically and microscopically). One important difference is that the local strain partitioning between the two phases is over-predicted by the two-dimensional microstructure, leading to an overestimation of damage

    A first-in-human phase I, dose-escalation, multicentre study of HSP990 administered orally in adult patients with advanced solid malignancies

    Heat-shock protein 990 (HSP990) is a potent and selective synthetic small-molecule HSP90 inhibitor. The primary objectives of this phase I first-in-human study were to determine dose-limiting toxicities (DLTs), maximum-tolerated dose (MTD) and recommended phase II dose (RP2D). Secondary objectives included characterisation of the safety profile, pharmacokinetics (PKs) and pharmacodynamics (PDs). Heat-shock protein 990 was administered orally once or two times weekly on a 28-day cycle schedule in patients with advanced solid tumours. Dose escalation was guided by a Bayesian logistic regression model with overdose control. A total of 64 patients were enrolled. Fifty-three patients received HSP990 once weekly at 2.5, 5, 10, 20, 30, 50 or 60 mg, whereas 11 patients received HSP990 two times weekly at 25 mg. Median duration of exposure was 8 weeks (range 1-116 weeks) and 12 patients remained on treatment for >16 weeks. Dose-limiting toxicities occurred in seven patients and included diarrhoea, QTc prolongation, ALT/AST elevations and central neurological toxicities. The most common drug-related adverse events were diarrhoea, fatigue and decreased appetite. Further dose escalation beyond 60 mg once weekly was not possible owing to neurological toxicity. Rapid absorption, no drug accumulation and large interpatient variability in PK exposures were observed. No objective responses were seen; 25 patients had a best overall response of stable disease. Heat-shock protein 990 is relatively well tolerated, with neurological toxicity being the most relevant DLT. The single agent MTD/RP2D of HSP990 was declared at 50 mg once weekly

    A phase field model incorporating strain gradient viscoplasticity : application to rafting in Ni-base superalloys

    International audienceThe first formulation of a phase field model accounting for size-dependent viscoplasticity is developed to study materials in which microstructure evolution and viscoplastic behavior are strongly coupled. Plasticity is introduced using a continuum strain gradient formalism which captures the size effect of the viscoplastic behavior. First, the influence of this size effect on the mechanical behavior of the material is discussed in static microstructures. Then, the dynamic coupling between microstructure evolution and viscoplastic activity is addressed and illustrated by the rafting of the microstructure observed in Ni-base superalloys under creep conditions. It is found that the plastic size effect has only a moderate impact on the shape of the rafts but is crucial to reproduce the macroscopic mechanical behavior of that particular material

    Coupled glide-climb diffusion-enhanced crystal plasticity

    International audienceThis paper presents a fully coupled glide-climb crystal plasticity model, whereby climb is controlled by the diffusion of vacancies. An extended strain gradient crystal plasticity model is therefore proposed, which incorporates the climbing of dislocations in the governing transport equations. A global-local approach is adopted to separate the scales and assess the influence of local diffusion on the global plasticity problem. The kinematics of the crystal plasticity model is enriched by incorporating the climb kinematics in the crystallographic split of the plastic strain rate tensor. The potential of the fully coupled theory is illustrated by means of two single slip examples that illustrate the interaction between glide and climb in either bypassing a precipitate or destroying a dislocation pile-up

    Diagnóstico del manejo de los efluentes porcinos generados en la provincia de Córdoba

    La intensificación de la producción porcina genera grandes volúmenes de efluentes que deben ser manejados adecuadamente. Su acumulación y aplicaciones no controladas al suelo pueden provocar impactos negativos en el ambiente. Este trabajo se planteó como objetivo estimar la generación de efluente porcino en la provincia de Córdoba y diagnosticar actuales prácticas de manejo en el departamento de Marcos Juárez. Las estimaciones realizadas evidencian que el potencial de generación de efluente porcino en la provincia es de 811911 m. año-1, los cuales aportan 2517 toneladas N año-1. Esta generación se concentra en los departamentos sur-sureste, ligados a la mayor existencia animal. A su vez, el departamento Marcos Juárez genera 119378 m. año-1 de efluente porcino. De este volumen generado un 61% fue almacenado en sistemas impermeabilizados, 51% fue sometido a sistemas de tratamiento y un 68% fue aplicado al suelo. Sin embargo, al evaluar el porcentaje de productores que llevaron a cabo estas prácticas, se evidenció que estos fueron minoría, pero con mayor número de animales

    Cariprazine add-on in inadequate clozapine response: A report on two cases

    Cariprazine is a novel antipsychotic drug that exerts partial agonism of dopamine D2/D3receptors with preferential binding to the D3receptor, antagonism of 5HT2B receptors, and partial agonism of 5HT1A. Currently, cariprazine has shown clinical efficacy in patients with schizophrenia and with bipolar disorder, as well as adjunctive treatment in patients with Major Depressive Disorder (MDD) and drug-resistant MDD. In the present case series, we report on two patients with treatment-resistant schizophrenia and partial response to clozapine who benefit from combination with cariprazine. The effects of cariprazine combination were remarkable also concerning the adverse metabolic effects of clozapine
