    Random Networks Tossing Biased Coins

    In statistical mechanical investigations on complex networks, it is useful to employ random graphs ensembles as null models, to compare with experimental realizations. Motivated by transcription networks, we present here a simple way to generate an ensemble of random directed graphs with, asymptotically, scale-free outdegree and compact indegree. Entries in each row of the adjacency matrix are set to be zero or one according to the toss of a biased coin, with a chosen probability distribution for the biases. This defines a quick and simple algorithm, which yields good results already for graphs of size n ~ 100. Perhaps more importantly, many of the relevant observables are accessible analytically, improving upon previous estimates for similar graphs

    Folding and cytoplasm viscoelasticity contribute jointly to chromosome dynamics

    The chromosome is a key player of cell physiology, and its dynamics provides valuable information about its physical organization. In both prokaryotes and eukaryotes, the short-time motion of chromosomal loci has been described as a Rouse model in a simple or viscoelastic medium. However, little emphasis has been put on the role played by the folded organization of chromosomes on the local dynamics. Clearly, stress-propagation, and thus dynamics, must be affected by such organization, but a theory allowing to extract such information from data, e.g.\ of two-point correlations, is lacking. Here, we describe a theoretical framework able to answer this general polymer dynamics question, and we provide a general scaling analysis of the stress-propagation time between two loci at a given arclength distance along the chromosomal coordinate. The results suggest a precise way to detect folding information from the dynamical coupling of chromosome segments. Additionally, we realize this framework in a specific theoretical model of a polymer with variable-range interactions in a viscoelastic medium characterized by a tunable scaling exponent, where we derive analytical estimates of the correlation functions.Comment: 14 pages including supplementary material

    A new coupling solution for G3-PLC employment in MV smart grids

    This paper proposes a new coupling solution for transmitting narrowband multicarrier power line communication (PLC) signals over medium voltage (MV) power lines. The proposed system is based on an innovative PLC coupling principle, patented by the authors, which exploits the capacitive divider embedded in voltage detecting systems (VDS) already installed inside the MV switchboard. Thus, no dedicated couplers have to be installed and no switchboard modifications or energy interruptions are needed. This allows a significant cost reduction of MV PLC implementation. A first prototype of the proposed coupling system was presented in previous papers: it had a 15 kHz bandwidth useful to couple single carrier PSK modulated PLC signals with a center frequency from 50–200 kHz. In this paper, a new prototype is developed with a larger bandwidth, up to 164 kHz, thus allowing to couple multicarrier G3-PLC signals using orthogonal frequency division multiplexing (OFDM) digital modulation. This modulation ensures a more robust communication even in harsh power line channels. In the paper, the new coupling system design is described in detail. A new procedure is presented for tuning the coupling system parameters at first installation in a generic MV switchboard. Finally, laboratory and in-field experimental test results are reported and discussed. The coupling performances are evaluated measuring the throughput and success rate in the case of both 18 and 36 subcarriers, in one of the different tone masks standardized for the FCC-above CENELEC band (that is, from 154.6875–487.5 kHz). The experimental results show an efficient behavior of the proposed coupler allowing a two-way communication of G3-PLC OFDM signals on MV networks

    Curing hemoglobinopathies : challenges and advances of conventional and new gene therapy approaches

    Inherited hemoglobin disorders, including beta-thalassemia (BT) and sickle-cell disease (SCD), are the most common monogenic diseases worldwide, with a global carrier frequency of over 5%.1 With migration, they are becoming more common worldwide, making their management and care an increasing concern for health care systems. BT is characterized by an imbalance in the \u3b1/\u3b2-globin chain ratio, ineffective erythropoiesis, chronic hemolytic anemia, and compensatory hemopoietic expansion.1 Globally, there are over 25,000 births each year with transfusion-dependent thalassemia (TDT). The currently available treatment for TDT is lifelong transfusions and iron chelation therapy or allogenic bone marrow transplantation as a curative option. SCD affects 300 million people worldwide2 and severely impacts the quality of life of patients who experience unpredictable, recurrent acute and chronic severe pain, stroke, infections, pulmonary disease, kidney disease, retinopathy, and other complications. While survival has been dramatically extended, quality of life is markedly reduced by disease- and treatment-associated morbidity. The development of safe, tissue-specific and efficient vectors, and efficient gene-editing technologies has led to the development of several gene therapy trials for BT and SCD. However, the complexity of the approach presents its hurdles. Fundamental factors at play include the requirement for myeloablation on a patient with benign disease, the age of the patient, and the consequent bone marrow microenvironment. A successful path from proof-of-concept studies to commercialization must render gene therapy a sustainable and accessible approach for a large number of patients. Furthermore, the cost of these therapies is a considerable challenge for the health care system. While new promising therapeutic options are emerging,3,4 and many others are on the pipeline,5 gene therapy can potentially cure patients. We herein provide an overview of the most recent, likely potentially curative therapies for hemoglobinopathies and a summary of the challenges that these approaches entail

    Mean-field methods in evolutionary duplication-innovation-loss models for the genome-level repertoire of protein domains

    We present a combined mean-field and simulation approach to different models describing the dynamics of classes formed by elements that can appear, disappear or copy themselves. These models, related to a paradigm duplication-innovation model known as Chinese Restaurant Process, are devised to reproduce the scaling behavior observed in the genome-wide repertoire of protein domains of all known species. In view of these data, we discuss the qualitative and quantitative differences of the alternative model formulations, focusing in particular on the roles of element loss and of the specificity of empirical domain classes.Comment: 10 Figures, 2 Table

    1814-1819The correct use of closed field chambers to determine N2O emissions requires defining the time of day that best represents the daily mean N2O flux. A short-term field experiment was carried out on a Mollisol soil, on which annual crops were grown under no-till management in the Pampa Ondulada of Argentina. The N2O emission rates were measured every 3 h for three consecutive days. Fluxes ranged from 62.58 to 145.99 ug N-N2O m-2 h-1 (average of five field chambers) and were negatively related (R2 equal 0.34, p less than 0.01) to topsoil temperature (14 - 20 oC). N2O emission rates measured between 9:00 and 12:00 am presented a high relationship to daily mean N2O flux (R2 equal 0.87, p less than 0.01), showing that, in the study region, sampling in the mornings is preferable for GHG

    Physical descriptions of the bacterial nucleoid at large scales, and their biological implications.

    Recent experimental and theoretical approaches have attempted to quantify the physical organization (compaction and geometry) of the bacterial chromosome with its complement of proteins (the nucleoid). The genomic DNA exists in a complex and dynamic protein-rich state, which is highly organized at various length scales. This has implications for modulating (when not directly enabling) the core biological processes of replication, transcription and segregation. We overview the progress in this area, driven in the last few years by new scientific ideas and new interdisciplinary experimental techniques, ranging from high space- and time-resolution microscopy to high-throughput genomics employing sequencing to map different aspects of the nucleoid-related interactome. The aim of this review is to present the wide spectrum of experimental and theoretical findings coherently, from a physics viewpoint. In particular, we highlight the role that statistical and soft condensed matter physics play in describing this system of fundamental biological importance, specifically reviewing classic and more modern tools from the theory of polymers. We also discuss some attempts toward unifying interpretations of the current results, pointing to possible directions for future investigation

    Effects of Dietary Extruded Linseed (Linum usitatissimum) and Oregano (Origanum vulgare) on Growth Traits, Carcass Composition and Meat Quality of Grigia di Potenza Suckling Kids

    The aim of this trial was to compare the influence of supplementing diets with extruded linseed and oregano on growth parameters and meat qualitative traits in Grigia di Potenza breed suckling kids. Twenty-four male kids, exclusively fed milk from their dams, were assigned to the following diets: C) group control fed without any supplement; L) group fed control feed containing 3% extruded linseed (Linum usitatissimum L.); and LO) group fed control diet with 0.6% dried oregano (Origanum vulgare) and 3% extruded linseed. Growth performance as well as slaughtering traits and meat cuts of kids were not significantly influenced (P > 0.05) by dietary treatments. Conversely, kids in linseed group reported the lower (P < 0.05) percentage of dissectible fat in leg and loin. The meat from Longissimus lumborum and Semimembranosus muscles of kids in linseed diet had the lowest (P < 0.05) cooking loss percentage, whereas the proximate chemical composition of both meat muscles did not vary among treatments (P > 0.05). The experimental diets partially modulated the kid meat fatty acid composition in both muscles, where feeding linseed and oregano improved (P < 0.05) the content of DPA and reduced MUFA. Based on the current findings, it can be concluded that linseed and oregano supplementation can be used in goat diet as no significant detrimental effects on productive performance and meat quality of suckling kids were observed
