Literarisches Sachsen. Studie zur Verbindung von Literaturmuseen und literarischen GedenkstĂ€tten in einer SĂ€chsischen LiteraturstraĂe.
âWĂ€hrend ĂŒber das Geschichtsbewusstsein einer Nation die chronologisch geordneten GeschichtsbĂŒcher AufschluĂ geben, findet das GedĂ€chtnis einer Nation seinen Niederschlag in der GedĂ€chtnislandschaft seiner Erinnerungsorte.â schreibt Aleida Assmann in ErinnerungsrĂ€ume. Formen und Wandlungen des kulturellen GedĂ€chtnisses (MĂŒnchen: Verlag C. H. Beck oHG, 1999. S. 337.). In der vorliegenden Masterarbeit wird auf Basis dieser Definition eine Anwendung auf den Bereich der Literatur- und Kulturwissenschaften, im Sinne einer Fokussierung auf literarische Erinnerungsorte, vollzogen, die als Grundlage des kulturellen GedĂ€chtnisses angesehen werden und demzufolge zur IdentitĂ€tsstiftung einer Region beitragen. Hierbei steht die Vernetzung ausgewĂ€hlter literarischer Erinnerungsorte, bestehend aus Literaturmuseen und literarischen GedenkstĂ€tten der Region Sachsen, im Vordergrund. Das Ziel dieser Masterarbeit ist einerseits die literaturwissenschaftliche Auseinandersetzung mit Schriftstellern, die in Sachsen gelebt und/ oder gewirkt haben und andererseits die Darstellung des kulturellen Einflusses der bestehenden literarischen GedenkstĂ€tten und Literaturmuseen in der âRegionâ Sachsen.
Im Rahmen dieser Masterarbeit wurde eine Bestandsaufnahme von 231 Schriftstellern angefertigt, die im heutigen Gebiet des Freistaates Sachsen gelebt und/ oder gewirkt haben, aber auch alle Museen, DenkmĂ€ler, GrabmĂ€ler, Gedenktafeln und BĂŒsten, die an den jeweiligen Schriftsteller erinnern. Hieraus wurden fĂŒr die vorliegende Masterarbeit 15 Schriftsteller nach spezifischen Kriterien ausgewĂ€hlt und diese, ebenso wie die zugehörigen Literaturmuseen und literarischen GedenkstĂ€tten, vorgestellt
Heterogener Studierendenschaft begegnen. MaĂnahmen zur Flexibilisierung des Studiums an der TU Dresden
Vor dem Hintergrund der zunehmenden HeterogenitĂ€t Studierender werden im Rahmen des Projektes âStudiengĂ€nge flexibel gestaltenâ an der TU Dresden MaĂnahmen erprobt, die eine Flexibilisierung des Studiums ermöglichen sollen. Neben der Entwicklung eines E-Scout-Programms, das Lehrende bei der Umsetzung von digital gestĂŒtzten Lernangeboten unterstĂŒtzt, wird im Projektkontext ein evidenzbasiertes Entscheidungsmodell fĂŒr Lehrende entwickelt, dass Empfehlungen fĂŒr passende Lehrformate vorschlĂ€gt.
Der Beitrag stellt aktuelle Rahmenbedingungen der UniversitĂ€t vor und zeigt auf, auf welchen Ebenen der Hochschulstruktur die AnsĂ€tze zu verorten sind. Die ProjektmaĂnahmen machen deutlich, dass die Gestaltung flexiblen Lernens an Hochschulen unterschiedliche Facetten hat und vielfĂ€ltig ausgestaltet werden kann. Dies wird in einem entwickelten Vorgehensmodell abgebildet. Den theoretischen Rahmen bildet die Herleitung einer Systematik von FlexibilisierungsmaĂnahmen, in die die erprobten Konzepte eingeordnet und hinsichtlich ihrer Potentiale zur Ăbertragbarkeit auf andere Hochschulen diskutiert werden
LOS gehtâs! Learning Experience durch Gamification
Mit der Digitalisierung im Bildungsbereich sind Hochschulen aufgefordert, neue und mit digitalen Technologien angereicherte Lehr- und Lernformate bereitzustellen. Die sĂ€chsischen Hochschulen können dafĂŒr auf eine flĂ€chendeckende, technologische Infrastruktur [u. a. das Lernmanagementsystem (LMS) OPAL] zurĂŒckgreifen. Im Lehralltag zeigen sich jedoch fach- und hochschulĂŒbergreifend Ă€hnliche PhĂ€nomene: Die digitalen Infrastrukturen werden vorwiegend fĂŒr administrative Zwecke (bspw. fĂŒr Kurseinschreibung oder Materialbereitstellung) und nur in geringem MaĂe zur innovativen Gestaltung von Lehr-Lern-Arrangements genutzt. Dabei bietet die vorhandene Infrastruktur reichlich Potenzial fĂŒr motivational anspruchsvolle, digitale Lehr-Lern-Arrangements in allen Fachgebieten. Mit der Steigerung der Learning Experience, also der Lernerfahrung, sollen positive Emotionen geweckt und fĂŒr den Lernprozess genutzt werden. Lernen soll Freude machen. Mit dem Projekt LOS (Learning Experience in OPAL mit Spielelementen) wird ein frei zugĂ€ngliches Transfer- und Weiterbildungsangebot aufgebaut, mit dem Lehrende fĂŒr die Steigerung der Learning Experience ihrer Lernangebote durch den Einsatz von Spielelementen sensibilisiert und qualifiziert werden. Damit soll langfristig die AttraktivitĂ€t und der Nutzungsgrad der sĂ€chsischen Lernplattform aus Sicht aller akademischen Stakeholder gesteigert werden. Chronological aging leads to apoptosis in yeast
During the past years, yeast has been successfully established as a model to study mechanisms of apoptotic regulation. However, the beneficial effects of such a cell suicide program for a unicellular organism remained obscure. Here, we demonstrate that chronologically aged yeast cultures die exhibiting typical markers of apoptosis, accumulate oxygen radicals, and show caspase activation. Age-induced cell death is strongly delayed by overexpressing YAP1, a key transcriptional regulator in oxygen stress response. Disruption of apoptosis through deletion of yeast caspase YCA1 initially results in better survival of aged cultures. However, surviving cells lose the ability of regrowth, indicating that predamaged cells accumulate in the absence of apoptotic cell removal. Moreover, wild-type cells outlast yca1 disruptants in direct competition assays during long-term aging. We suggest that apoptosis in yeast confers a selective advantage for this unicellular organism, and demonstrate that old yeast cells release substances into the medium that stimulate survival of the clone
Microenvironmentâinduced restoration of cohesive growth associated with focal activation of P âcadherin expression in lobular breast carcinoma metastatic to the colon
Invasive lobular carcinoma (ILC) is a special breast cancer type characterized by noncohesive growth and Eâcadherin loss. Focal activation of Pâcadherin expression in tumor cells that are deficient for Eâcadherin occurs in a subset of ILCs. Switching from an Eâcadherin deficient to Pâcadherin proficient status (EPS) partially restores cellâcell adhesion leading to the formation of cohesive tubular elements. It is unknown what conditions control EPS. Here, we report on EPS in ILC metastases in the large bowel. We reviewed endoscopic colon biopsies and colectomy specimens from a 52âyearâold female (index patient) and of 18 additional patients (reference series) diagnosed with metastatic ILC in the colon. EPS was assessed by immunohistochemistry for Eâcadherin and Pâcadherin. CDH1 /Eâcadherin mutations were determined by nextâgeneration sequencing. The index patient's colectomy showed transmural metastatic ILC harboring a CDH1 /Eâcadherin p.Q610* mutation. ILC cells displayed different growth patterns in different anatomic layers of the colon wall. In the tunica muscularis propria and the tela submucosa, ILC cells featured noncohesive growth and were Eâcadherinânegative and Pâcadherinânegative. However, ILC cells invading the mucosa formed cohesive tubular elements in the intercryptal stroma of the lamina propria mucosae. Interâcryptal ILC cells switched to a Pâcadherinâpositive phenotype in this microenvironmental niche. In the reference series, colon mucosa infiltration was evident in 13 of 18 patients, one of which showed intercryptal EPS and conversion to cohesive growth as described in the index patient. The large bowel is a common metastatic site in ILC. In endoscopic colon biopsies, the typical noncohesive growth of ILC may be concealed by microenvironmentâinduced EPS and conversion to cohesive growth
Resistance to mesenchymal reprogramming sustains clonal propagation in metastatic breast cancer
The acquisition of mesenchymal traits is considered a hallmark of breast cancer progression. However, the functional relevance of epithelial-to-mesenchymal transition (EMT) remains controversial and context dependent. Here, we isolate epithelial and mesenchymal populations from human breast cancer metastatic biopsies and assess their functional potential in vivo. Strikingly, progressively decreasing epithelial cell adhesion molecule (EPCAM) levels correlate with declining disease propagation. Mechanistically, we find that persistent EPCAM expression marks epithelial clones that resist EMT induction and propagate competitively. In contrast, loss of EPCAM defines clones arrested in a mesenchymal state, with concomitant suppression of tumorigenicity and metastatic potential. This dichotomy results from distinct clonal trajectories impacting global epigenetic programs that are determined by the interplay between human ZEB1 and its target GRHL2. Collectively, our results indicate that susceptibility to irreversible EMT restrains clonal propagation, whereas resistance to mesenchymal reprogramming sustains disease spread in multiple models of human metastatic breast cancer, including patient-derived cells in vivo
High-Level Expression of Wild-Type p53 in Melanoma Cells is Frequently Associated with Inactivity in p53 Reporter Gene Assays
Background: Inactivation of the p53 pathway that controls cell cycle progression, apoptosis and senescence, has been proposed to occur in virtually all human tumors and p53 is the protein most frequently mutated in human cancer. However, the mutational status of p53 in melanoma is still controversial; to clarify this notion we analysed the largest series of melanoma samples reported to date. Methodology/Principal Findings: Immunohistochemical analysis of more than 180 melanoma specimens demonstrated that high levels of p53 are expressed in the vast majority of cases. Subsequent sequencing of the p53 exons 5â8, however, revealed only in one case the presence of a mutation. Nevertheless, by means of two different p53 reporter constructs we demonstrate transcriptional inactivity of wild type p53 in 6 out of 10 melanoma cell lines; the 4 other p53 wild type melanoma cell lines exhibit p53 reporter gene activity, which can be blocked by shRNA knock down of p53. Conclusions/Significance: In melanomas expressing high levels of wild type p53 this tumor suppressor is frequently inactivated at transcriptional level
DNA methylation-based classification of sinonasal tumors
The diagnosis of sinonasal tumors is challenging due to a heterogeneous spectrum of various differential diagnoses as well as poorly defined, disputed entities such as sinonasal undifferentiated carcinomas (SNUCs). In this study, we apply a machine learning algorithm based on DNA methylation patterns to classify sinonasal tumors with clinical-grade reliability. We further show that sinonasal tumors with SNUC morphology are not as undifferentiated as their current terminology suggests but rather reassigned to four distinct molecular classes defined by epigenetic, mutational and proteomic profiles. This includes two classes with neuroendocrine differentiation, characterized by IDH2 or SMARCA4/ARID1A mutations with an overall favorable clinical course, one class composed of highly aggressive SMARCB1-deficient carcinomas and another class with tumors that represent potentially previously misclassified adenoid cystic carcinomas. Our findings can aid in improving the diagnostic classification of sinonasal tumors and could help to change the current perception of SNUCs
