    Road and bridge construction across gypsum karst in England

    Gypsum karst problems in the Permian and Triassic sequences of England have caused difficult conditions for bridge and road construction. In Northern England, the Ripon Bypass crosses Permian strata affected by active gypsum karst and severe subsidence problems. Here, the initial borehole site investigation for the road was supplemented by resistivity tomography studies. The roadway was reinforced with two layers of tensile membrane material within the earth embankment. This will prevent dangerous catastrophic collapse, but will allow sagging to show where problems exist. The River Ure Bridge was constructed across an area of subsidence pipes filled with alluvial deposits. It was built with extra strength, larger than normal foundations. If one pier fails, the bridge is designed for adjacent arches to span the gap without collapse. The bridge piers are also fitted with electronic load monitoring to warn of failure. In the Midlands area of England, road construction over Triassic gypsum has required a phase of ground improvement on the Derby Southern Bypass. Here, the gypsum caps a hill where it was formerly mined; it dips through a karstic dissolution zone into an area of complete dissolution and collapse. The road and an associated flyover were built across these ground conditions. A major grouting program before the earthworks began treated the cavities in the mine workings and the cavernous margin of the gypsum mass. Within the karstic dissolution zone, gypsum blocks and cavities along the route were identified by conductivity and resistivity geophysical surveys, excavated and backfilled. In the areas of complete dissolution and collapse, the road foundation was strengthened with vibrated stone columns and a reinforced concrete road deck was used

    Vacuum Structure of Two-Dimensional ϕ4\phi^4 Theory on the Orbifold S1/Z2S^{1}/Z_{2}

    We consider the vacuum structure of two-dimensional ϕ4\phi^4 theory on S1/Z2S^{1}/Z_{2} both in the bosonic and the supersymmetric cases. When the size of the orbifold is varied, a phase transition occurs at Lc=2π/mL_{c}=2\pi/m, where mm is the mass of ϕ\phi. For L<LcL<L_{c}, there is a unique vacuum, while for L>LcL>L_{c}, there are two degenerate vacua. We also obtain the 1-loop quantum corrections around these vacuum solutions, exactly in the case of L<LcL<L_{c} and perturbatively for LL greater than but close to LcL_{c}. Including the fermions we find that the "chiral" zero modes around the fixed points are different for LLcLL_{c}. As for the quantum corrections, the fermionic contributions cancel the singular part of the bosonic contributions at L=0.     Database Search Strategies for Proteomic Data Sets Generated by Electron Capture Dissociation Mass Spectrometry

    Large data sets of electron capture dissociation (ECD) mass spectra from proteomic experiments are rich in information; however, extracting that information in an optimal manner is not straightforward. Protein database search engines currently available are designed for low resolution CID data, from which Fourier transform ion cyclotron resonance (FT-ICR) ECD data differs significantly. ECD mass spectra contain both z-prime and z-dot fragment ions (and c-prime and c-dot); ECD mass spectra contain abundant peaks derived from neutral losses from charge-reduced precursor ions; FT-ICR ECD spectra are acquired with a larger precursor m/z isolation window than their low-resolution CID counterparts. Here, we consider three distinct stages of postacquisition analysis: (1) processing of ECD mass spectra prior to the database search; (2) the database search step itself and (3) postsearch processing of results. We demonstrate that each of these steps has an effect on the number of peptides identified, with the postsearch processing of results having the largest effect. We compare two commonly used search engines: Mascot and OMSSA. Using an ECD data set of modest size (3341 mass spectra) from a complex sample (mouse whole cell lysate), we demonstrate that search results can be improved from 630 identifications (19% identification success rate) to 1643 identifications (49% identification success rate). We focus in particular on improving identification rates for doubly charged precursors, which are typically low for ECD fragmentation.     'I-I' and 'I-me' : Transposing Buber's interpersonal attitudes to the intrapersonal plane

    Hermans' polyphonic model of the self proposes that dialogical relationships can be established between multiple I-positions1 (e.g., Hermans, 2001a). There have been few attempts, however, to explicitly characterize the forms that these intrapersonal relationships may take. Drawing on Buber's (1958) distinction between the 'I-Thou' and 'I-It' attitude, it is proposed that intrapersonal relationships can take one of two forms: an 'I-I' form, in which one I-position encounters and confirms another I-position in its uniqueness and wholeness; and an 'I-Me' form, in which one I-position experiences another I-position in a detached and objectifying way. This article argues that this I-Me form of intrapersonal relating is associated with psychological distress, and that this is so for a number of reasons: Most notably, because an individual who objectifies and subjugates certain I-position cannot reconnect with more central I-positions when dominance reversal (Hermans, 2001a) takes place. On this basis, it is suggested that a key role of the therapeutic process is to help clients become more able to experience moments of I-I intrapersonal encounter, and it is argued that this requires the therapist to confirm the client both as a whole and in terms of each of his or her different voices

    Evolutionary rates and gene dispensability associate with replication timing in the Archaeon Sulfolobus islandicus

    In bacterial chromosomes, the position of a gene relative to the single origin of replication generally reflects its replication timing, how often it is expressed, and consequently, its rate of evolution. However, because some archaeal genomes contain multiple origins of replication, bias in gene dosage caused by delayed replication should be minimized and hence the substitution rate of genes should associate less with chromosome position. To test this hypothesis, six archaeal genomes from the genus Sulfolobus containing three origins of replication were selected, conserved orthologs were identified, and the evolutionary rates (dN and dS) of these orthologs were quantified. Ortholog families were grouped by their consensus position and designated by their proximity to one of the three origins (O1, O2, O3). Conserved orthologs were concentrated near the origins and most variation in genome content occurred distant from the origins. Linear regressions of both synonymous and nonsynonymous substitution rates on distance from replication origins were significantly positive, the rates being greatest in the region furthest from any of the origins and slowest among genes near the origins. Genes near O1 also evolved faster than those near O2 and O3, which suggest that this origin may fire later in the cell cycle. Increased evolutionary rates and gene dispensability are strongly associated with reduced gene expression caused in part by reduced gene dosage during the cell cycle. Therefore, in this genus of Archaea as well as in many Bacteria, evolutionary rates and variation in genome content associate with replication timing
