    A Hybrid Multi-Distance Phase and Broadband Spatially Resolved Spectrometer and Algorithm for Resolving Absolute Concentrations of Chromophores in the Near-Infrared Light Spectrum

    For resolving absolute concentration of tissue chromophores in the human adult brain with near-infrared spectroscopy it is necessary to calculate the light scattering and absorption, at multiple wavelengths with some depth resolution. To achieve this we propose an instrumentation configuration that combines multi-distance frequency and broadband spectrometers to quantify chromophores in turbid media by using a hybrid spatially resolved algorithm. Preliminary results in solid phantoms as well as liquid dynamic homogeneous and inhomogeneous phantoms and in-vivo muscle measurements showed encouraging results

    Underwater near infrared spectroscopy (uNIRS) can measure training adaptations in adolescent swimmers

    The development of an underwater near-infrared spectroscopy (uNIRS) device has enabled previously unattainable measurements of peripheral muscle hemodynamics and oxygenation to be taken within the natural aquatic environment. The purposes of this study were (i) to trial the use of uNIRS, in a real world training study, and (ii) to monitor the effects of a swim training program upon muscle oxygenation status in short distance swimming. 14 junior club level swimmers completed a repeated swim sprint test before and after an eight week endurance training program. A waterproof, portable NIRS device was attached to the vastus lateralis. uNIRS successfully measured changes in muscle oxygenation and blood volume in all individuals; rapid sub-second time resolution of the device was able to demonstrate muscle oxygenation changes during the characteristic swim movements. Post training heart rate recovery and swim performance time were significantly improved. uNIRS data also showed significant changes. A larger rise in deoxyhemoglobin during individual sprints suggested training induced an increase in muscle oxygen extraction; a faster recovery time for muscle oxygenation suggested positive training induced changes and significant changes in muscle blood volume also occur. As a strong correlation was seen between an increased reoxygenation rate and an improved swim performance time, these findings support the use of uNIRS as a new performance analysis tool in swimming

    Reduction of cytochrome C oxidase during vasovagal hypoxia-ischemia in human adult brain: a case study

    Near-infrared spectroscopy (NIRS)-derived measurement of oxidized cytochrome c oxidase concentration ([oxCCO]) has been used as an assessment of the adequacy of cerebral oxygen delivery. We report a case in which a reduction in conscious level was associated with a reduction in [oxCCO]. Hypoxaemia was induced in a 31-year-old, healthy male subject as part of an ongoing clinical study. Midway through the hypoxaemic challenge, the subject experienced an unexpected vasovagal event with bradycardia, hypotension and reduced cerebral blood flow (middle cerebral artery blood flow velocity decrease from 70 to 30 cm s(-1)) that induced a brief reduction in conscious level. An associated decrease in [oxCCO] was observed at 35 mm (-1.6 μM) but only minimal change (-0.1 μM) at 20-mm source-detector separation. A change in optical scattering was observed, but path length remained unchanged. This unexpected physiological event provides an unusual example of a severe reduction in cerebral oxygen delivery and is the first report correlating change in clinical status with changes in [oxCCO]

    Cytochrome c oxidase response to changes in cerebral oxygen delivery in the adult brain shows higher brain-specificity than haemoglobin

    The redox state of cerebral mitochondrial cytochrome c oxidase monitored with near-infrared spectroscopy (Δ[oxCCO]) is a signal with strong potential as a non-invasive, bedside biomarker of cerebral metabolic status. We hypothesised that the higher mitochondrial density of brain compared to skin and skull would lead to evidence of brain-specificity of the Δ[oxCCO] signal when measured with a multi-distance near-infrared spectroscopy (NIRS) system. Measurements of Δ[oxCCO] as well as of concentration changes in oxygenated (Δ[HbO2]) and deoxygenated haemoglobin (Δ[HHb]) were taken at multiple source-detector distances during systemic hypoxia and hypocapnia (decrease in cerebral oxygen delivery), and hyperoxia and hypercapnia (increase in cerebral oxygen delivery) from 15 adult healthy volunteers. Increasing source-detector spacing is associated with increasing light penetration depth and thus higher sensitivity to cerebral changes. An increase in Δ[oxCCO] was observed during the challenges that increased cerebral oxygen delivery and the opposite was observed when cerebral oxygen delivery decreased. A consistent pattern of statistically significant increasing amplitude of the Δ[oxCCO] response with increasing light penetration depth was observed in all four challenges, a behaviour that was distinctly different from that of the haemoglobin chromophores, which did not show this statistically significant depth gradient. This depth-dependence of the Δ[oxCCO] signal corroborates the notion of higher concentrations of CCO being present in cerebral tissue compared to extracranial components and highlights the value of NIRS-derived Δ[oxCCO] as a brain-specific signal of cerebral metabolism, superior in this aspect to haemoglobin

    Metabolic, hygric and ventilatory physiology of a hypermetabolic marsupial, the honey possum (Tarsipes rostratus)

    The honey possum is the only non-volant mammal to feed exclusively on a diet of nectar and pollen. Like other mammalian and avian nectarivores, previous studies indicated that the honey possum's basal metabolic rate was higher than predicted for a marsupial of equivalent body mass. However, these early measurements have been questioned. We re-examined the basal metabolic rate (2.52 +/- A 0.222 ml O(2) g(-1) h(-1)) of the honey possum and confirm that it is indeed higher (162%) than predicted for other marsupials both before and after accounting for phylogenetic history. This, together with its small body mass (5.4 +/- A 0.14 g; 1.3% of that predicted by phylogeny) may be attributed to its nectarivorous diet and mesic distribution. Its high-basal metabolic rate is associated with a high-standard body temperature (36.6 +/- A 0.48A degrees C) and oxygen extraction (19.4%), but interestingly the honey possum has a high point of relative water economy (17.0A degrees C) and its standard evaporative water loss (4.33 +/- A 0.394 mg H(2)O g(-1) h(-1)) is not elevated above that of other marsupials, despite its mesic habitat and high dietary water intake.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP

    Comparative physiology of Australian quolls (Dasyurus; Marsupialia)

    Quolls (Dasyurus) are medium-sized carnivorous dasyurid marsupials. Tiger (3,840 g) and eastern quolls (780 g) are mesic zone species, northern quolls (516 g) are tropical zone, and chuditch (1,385 g) were once widespread through the Australian arid zone. We found that standard physiological variables of these quolls are consistent with allometric expectations for marsupials. Nevertheless, inter-specific patterns amongst the quolls are consistent with their different environments. The lower T ^sub b^ of northern quolls (34°C) may provide scope for adaptive hyperthermia in the tropics, and they use torpor for energy/water conservation, whereas the larger mesic species (eastern and tiger quolls) do not appear to. Thermolability varied from little in eastern (0.035°C °C^sup -1^) and tiger quolls (0.051°C ºC^sup -1^) to substantial in northern quolls (0.100°C ºC^sup -1^) and chuditch (0.146°C ºC^sup -1^), reflecting body mass and environment. Basal metabolic rate was higher for eastern quolls (0.662 ± 0.033 ml O^sub 2^ g^sup -1^ h^sup -1^), presumably reflecting their naturally cool environment. Respiratory ventilation closely matched metabolic demand, except at high ambient temperatures where quolls hyperventilated to facilitate evaporative heat loss; tiger and eastern quolls also salivated. A higher evaporative water loss for eastern quolls (1.43 ± 0.212 mg H^sub 2^O g^sup -1^ h^sup -1^) presumably reflects their more mesic distribution. The point of relative water economy was low for tiger (-1.3°C), eastern (-12.5°C) and northern (+3.3) quolls, and highest for the chuditch (+22.6°C). We suggest that these differences in water economy reflect lower expired air temperatures and hence lower respiratory evaporative water loss for the arid-zone chuditch relative to tropical and mesic quolls

    Channelling of hydrothermal fluids during the accretion and evolution of the upper oceanic crust: Sr isotope evidence from ODP Hole 1256D

    ODP Hole 1256D in the eastern equatorial Pacific is the first penetration of a complete section of fast spread ocean crust down to the dike-gabbro transition, and only the second borehole to sample in situ sheeted dikes after DSDP Hole 504B. Here a high spatial resolution record of whole rock and mineral strontium isotopic compositions from Site 1256 is combined with core observations and downhole wireline geophysical measurements to determine the extent of basalt-hydrothermal fluid reaction and to identify fluid pathways at different levels in the upper ocean crust.The volcanic sequence at Site 1256 is dominated by sheet and massive lava flows but the Sr isotope profile shows only limited exchange with seawater. However, the upper margins of two anomalously thick (>25 m) massive flow sequences are strongly hydrothermally altered with elevated Sr isotope ratios and appear to be conduits of lateral low-temperature off-axis fluid flow. Elsewhere in the lavas, high 87Sr/86Sr are restricted to breccia horizons. Mineralised hyaloclastic breccias in the Lava-Dike Transition are strongly altered to Mg-saponite, silica and pyrite, indicating alteration by mixed seawater and cooled hydrothermal fluids. In the Sheeted Dike Complex 87Sr/86Sr ratios are pervasively shifted towards hydrothermal fluid values (~0.705). Dike chilled margins display secondary mineral assemblages formed during both axial recharge and discharge and have higher 87Sr/86Sr than dike cores, indicating preferential fluid flow along dike margins. Localised increases in 87Sr/86Sr in the Dike-Gabbro Transition indicates the channelling of fluids along the sub-horizontal intrusive boundaries of the 25 to 50 m-thick gabbroic intrusions, with only minor increases in 87Sr/86Sr within the cores of the gabbro bodies.When compared to the pillow lava-dominated section from Hole 504B, the Sr isotope measurements from Site 1256 suggest that the extent of hydrothermal circulation in the upper ocean crust may be strongly dependent on the eruption style. Sheet and massive flow dominated lava sequences typical of fast spreading ridges may experience relatively restricted circulation, but there may be much more widespread circulation through pillow lava-dominated sections. In addition, the Hole 1256D sheeted dikes display a much greater extent of Sr-isotopic exchange compared to dikes from Hole 504B. Because seawater-derived hydrothermal fluids must transit the dikes during their evolution to black smoker-type fluids, the different Sr-isotope profiles for Holes 504B and 1256D suggest there are significant variations in mid-ocean ridge hydrothermal systems at fast and intermediate spreading ridges, which may impact geochemical cycles of elements mobilised by fluid-rock exchange at different temperatures

    An international comparative study of blood pressure in populations of European vs. African descent

    Background: The consistent finding of higher prevalence of hypertension in US blacks compared to whites has led to speculation that African-origin populations are particularly susceptible to this condition. Large surveys now provide new information on this issue. Methods: Using a standardized analysis strategy we examined prevalence estimates for 8 white and 3 black populations (N = 85,000 participants). Results: The range in hypertension prevalence was from 27 to 55% for whites and 14 to 44% for blacks. Conclusions: These data demonstrate that not only is there a wide variation in hypertension prevalence among both racial groups, the rates among blacks are not unusually high when viewed internationally. These data suggest that the impact of environmental factors among both populations may have been under-appreciated

    Use of a hybrid optical spectrometer for the measurement of changes in oxidized cytochrome c oxidase concentration and tissue scattering during functional activation

    We report the use of a novel dual-channel hybrid near-infrared optical spectrometer for the measurement of changes in oxidized cytochrome c oxidase (oxCCO) concentration and optical scattering in 11 healthy volunteers during functional activation induced by an anagram-solving task. A hemodynamic Changed ‘haemodynamic’ to ‘hemodynamic’ though followed UK spelling. Please check. response consistent with functional activation was seen in 9 out of 11 subjects and in 13 of the total 22 channels measured; only these channels were retained for further analysis. In each of these 13 channels the hemodynamic response was accompanied by a statistically significant change in [oxCCO], although the direction of this change was heterogenous with a significant increase seen in eight channels and a significant decrease seen in five channels. There was no significant change in the optical scattering coefficient measured at four wavelengths, and the use of differential pathlength factor (DPF) calculated in real-time using measured absorption and scattering showed a high level of agreement with a conventional algorithm. The possible reasons for the heterogeneity of the [oxCCO] response are discussed
