    The role of reactive oxygen species in antibiotic-induced cell death in Burkholderia cepacia complex bacteria

    It was recently proposed that bactericidal antibiotics, besides through specific drug-target interactions, kill bacteria by a common mechanism involving the production of reactive oxygen species (ROS). However, this mechanism involving the production of hydroxyl radicals has become the subject of a lot of debate. Since the contribution of ROS to antibiotic mediated killing most likely depends on the conditions, differences in experimental procedures are expected to be at the basis of the conflicting results. In the present study different methods (ROS specific stainings, gene-expression analyses, electron paramagnetic resonance, genetic and phenotypic experiments, detection of protein carbonylation and DNA oxidation) to measure the production of ROS upon antibiotic treatment in Burkholderia cepacia complex (Bcc) bacteria were compared. Different classes of antibiotics (tobramycin, ciprofloxacin, meropenem) were included, and both planktonic and biofilm cultures were studied. Our results indicate that some of the methods investigated were not sensitive enough to measure antibiotic induced production of ROS, including the spectrophotometric detection of protein carbonylation. Secondly, other methods were found to be useful only in specific conditions. For example, an increase in the expression of OxyR was measured in Burkholderia cenocepacia K56-2 after treatment with ciprofloxacin or meropenem (both in biofilms and planktonic cultures) but not after treatment with tobramycin. In addition results vary with the experimental conditions and the species tested. Nevertheless our data strongly suggest that ROS contribute to antibiotic mediated killing in Bcc species and that enhancing ROS production or interfering with the protection against ROS may form a novel strategy to improve antibiotic treatment

    Antimicrobial nano-silver non-woven polyethylene terephthalate fabric via an atmospheric pressure plasma deposition process

    An antimicrobial nano-silver non-woven polyethylene terephthalate (PET) fabric has been prepared in a three step process. The fabrics were first pretreated by depositing a layer of organosilicon thin film using an atmospheric pressure plasma system, then silver nano-particles (AgNPs) were incorporated into the fabrics by a dipping-dry process, and finally the nano-particles were covered by a second organosilicon layer of 10-50 nm, which acts as a barrier layer. Different surface characterization techniques like SEM and XPS have been implemented to study the morphology and the chemical composition of the nano-silver fabrics. Based on these techniques, a uniform immobilization of AgNPs in the PET matrix has been observed. The antimicrobial activity of the treated fabrics has also been tested using P. aeruginosa, S. aureus and C. albicans. It reveals that the thickness of the barrier layer has a strong effect on the bacterial reduction of the fabrics. The durability and stability of the AgNPs on the fabrics has also been investigated in a washing process. By doing so, it is confirmed that the barrier layer can effectively prevent the release of AgNPs and that the thickness of the barrier layer is an important parameter to control the silver ions release

    Surveying activity-travel behavior in Flanders: Assessing the impact of the survey design

    peer reviewedEver since car ownership and car use started to increase in Western Europe and the USA, transportation planners attempted to model people’s travel behavior. In the context of the Feathers project a dynamic activity-based travel demand framework is developed for Flanders. In this paper, the complete survey design of the data collection effort required for such dynamic activity-based model is discussed. A mixed survey design of using a PDA application on the one hand, and using traditional paper and pencil diaries on the other hand, turns out to be a very suitable way of collecting detailed information about planned and executed activity-travel behavior of households. The results show that no attrition effects are present, not on the number of out-of-home activities reported, nor on the number of trips reported. Moreover the survey mode (PDA versus paper and pencil) has no direct impact on the quantities investigated. Notwithstanding, it is essential for further analysis on the Feathers data to explicitly take into account mode effects because of two reasons. First, the effect of explanatory variables can be influenced by the survey mode. Second, the variance in the estimation of the quantity investigated can differ significantly. Heteroscedatisc linear regression models provide the required framework to explicitly take into account these mode effects

    An estimation of total vehicle travel reduction in the case of telecommuting. Detailed analysis using an activity-based modeling approach

One of the possible measures that can be adopted in TDM is the implementation of telecommuting. A significant number of studies have been conducted in the past to evaluate the effect of telecommuting on peak-period trips. However it is less studied whether telecommuting also effectively and significantly reduces total vehicle travel. For this reason, a conventional modeling approach was adopted in this paper to calculate total kilometers of travel saved in the case telecommuting would materialize in the Flanders area. In a second part, the paper also introduces the use of an activity-based modeling approach to evaluate the effect of telecommuting. By doing so, an operational activity-based framework is externally validated by means of another completely different model, both calibrated for the same application and study area

    Integration of inland waterway transport in the intermodal supply chain: a taxonomy of research challenges

    This paper identifies research opportunities which will enable the further integration of inland waterway transport in the intermodal supply chain. Intermodal transport may be interpreted as a chain of actors who supply a transport service. Inland navigation can play a crucial role in increasing supply chain service performance. A first group of research challenges lies in the evolving relationship between transport geography and logistics activities. The next set of research challenges has the objective to encourage efficient operations in IWT: development of a system wide model for IWT, integration of operational planning systems and analysis of bundling networks. A third group of research efforts is directed towards shippers and consignees who use the intermodal transport chain to send or receive their goods: further development of models that integrate intermodal transport decisions with supply chain decisions and creation of green supply chains. A fourth cluster of research challenges concerns the problem domain of external cost calculations. Finally detailed time series data on freight transport should be collected to support these future research tracks

Results of a regression analysis also indicate that in both regions uncertainty in daily distance travelled per person is strongly correlated with the inverse square root of the relevant socio-demographic population and the complexity of the classification, measured in terms of the number of possible classes

    The response of canine faecal microbiota to increased dietary protein is influenced by body condition

    Background: High protein diets shift the faecal microbiota into a more unfavourable composition in obese humans. In lean dogs, higher protein consumption is accompanied with increased production of putrefactive fermentation products, whereas obese dogs have a different gut microbiota compared to lean dogs. Still, the impact of high dietary protein on gut microbiota in obese dogs remains unclear. The aim of this study was to investigate faecal microbial changes in lean and obese dogs in response to two different levels of dietary protein. Six healthy lean and six obese Beagles were fed a high protein diet (HP) and a low protein diet (LP) for 28 days each in a crossover design. Denaturing gradient gel electrophoresis and quantitative PCR were performed on faecal samples for microbial profiling. Plasma acylcarnitine and fermentation metabolites were measured. Results: Dogs fed HP had higher concentrations of protein fermentation metabolites including faecal ammonia, isovalerate, isobutyrate, phenol, indole, serum indoxyl sulphate and plasma 3-OH isovalerylcarnitine compared to dogs fed LP, whereas no changes in faecal concentrations of acetate and butyrate were observed. The abundances of clostridial clusters IV and XIVa, covering the majority of butyrate-producing bacteria, and of the butyrate kinase gene, one of the terminal genes of the butyrate synthesis pathway were higher in dogs on HP compared to LP. Significant interactions between diet and body condition were found for the abundance of Firmicutes, Lactobacillus and clostridial cluster I. The similarity coefficient of faecal microbiota between the two diets was smaller in obese dogs than in lean dogs. Conclusions: High protein diet increased the abundance and activity of butyrate-producing bacteria in Beagles independent of the body condition. In addition, increasing dietary protein content had a greater overall impact on faecal microbiota in obese compared to lean dogs
