    Surface tension driven flow of blood in a rectangular microfluidic channel: Effect of erythrocyte aggregation

    Microfluidic platforms have increasingly been explored for in vitro blood diagnostics and for studying complex microvascular processes. The perfusion of blood in such devices is typically achieved through pressure driven set-ups. Surface tension driven blood flow provides an alternative flow delivery option, and various studies in the literature have examined the behaviour of blood flow in such fluidic devices. In such flows, the influence of red blood cell (RBC) aggregation, the phenomenon majorly responsible for the non-Newtonian nature of blood, requires particular attention. In the present work, we examine differences in the surface tension driven flow of aggregating, non-aggregating RBC, and Newtonian suspensions, in a rectangular micro channel. The velocity fields were obtained using microPIV techniques. The analytical solution for blood velocity in the channel is developed utilising the power law model for blood viscosity. The results showed that RBC aggregation has an impact at the late stages of the flow, observed mainly in the bluntness of the velocity profiles. At the initial stages of the flow the shearing conditions are found moderately elevated, preventing intense RBC aggregate formation. As the flow decelerates in the channel RBC aggregation increases, affecting the flow characteristics

    Balancing locality and concurrency: solving sparse triangular systems on GPUs

    Many numerical optimisation problems rely on fast algorithms for solving sparse triangular systems of linear equations (STLs). To accelerate the solution of such equations, two types of approaches have been used: on GPUs, concurrency has been prioritised to the disadvantage of data locality, while on multi-core CPUs, data locality has been prioritised to the disadvantage of concurrency. In this paper, we discuss the interaction between data locality and concurrency in the solution of STLs on GPUs, and we present a new algorithm that balances both. We demonstrate empirically that, subject to there being enough concurrency available in the input matrix, our algorithm outperforms Nvidia’s concurrencyprioritising CUSPARSE algorithm for GPUs. Experimental results show a maximum speedup of 5.8-fold. Our solution algorithm, which we have implemented in OpenCL, requires a pre-processing phase that partitions the graph associated with the input matrix into sub-graphs, whose data can be stored in low-latency local memories. This preliminary analysis phase is expensive, but because it depends only on the input matrix, its cost can be amortised when solving for many different right-hand sides

    Quantitative study of hydration of C3S and C2S by thermal analysis. Evolution and composition of C-S-H gels formed

    This research is part of a European project (namely, CODICE project), main objective of which is modelling, at a multi-scale, the evolution of the mechanical performance of non-degraded and degraded cementitious matrices. For that, a series of experiments were planned with pure synthetic tri-calcium silicate (C3S) and bi-calcium silicate (C2S) (main components of the Portland cement clinker) to obtain different calcium–silicate–hydrate (C–S–H) gel structures during their hydration. The characterization of those C–S–H gels and matrices will provide experimental parameters for the validation of the multi-scale modelling scheme proposed. In this article, a quantitative method, based on thermal analyses, has been used for the determination of the chemical composition of the C–S–H gel together with the degree of hydration and quantitative evolution of all the components of the pastes. Besides, the microstructure and type of silicate tetrahedron and mean chain length (MCL) were studied by scanning electron microscopy (SEM) and 29Si magic-angle-spinning (MAS) NMR, respectively. The main results showed that the chemical compositions for the C–S–H gels have a CaO/SiO2 M ratio almost constant of 1.7 for both C3S and C2S compounds. Small differences were found in the gel water content: the H2O/SiO2 M ratio ranged from 2.9 ± 0.2 to 2.6 ± 0.2 for the C3S (decrease) and from 2.4 ± 0.2 to 3.2 ± 0.2 for the C2S (increase). The MCL values of the C–S–H gels, determined from 29Si MAS NMR, were 3.5 and 4 silicate tetrahedron, for the hydrated C3S and C2S, respectively, remaining almost constant at all hydration periods

    Spinal versus General Anaesthesia in Postoperative Pain Management during Transurethral Procedures

    We compared the analgesic efficacy of spinal and general anaesthesia following transurethral procedures. 97 and 47 patients underwent transurethral bladder tumour resection (TUR-B) and transurethral prostatectomy (TUR-P), respectively. Postoperative pain was recorded using an 11-point visual analogue scale (VAS). VAS score was greatest at discharge from recovery room for general anaesthesia (P = 0.027). The pattern changed significantly at 8 h and 12 h for general anaesthesia's efficacy (P = 0.017 and P = 0.007, resp.). A higher VAS score was observed in pT2 patients. Patients with resected tumour volume >10 cm3 exhibited a VAS score >3 at 8 h and 24 h (P = 0.050, P = 0.036, resp.). Multifocality of bladder tumours induced more pain overall. It seems that spinal anaesthesia is more effective during the first 2 postoperative hours, while general prevails at later stages and at larger traumatic surfaces. Finally, we incidentally found that tumour stage plays a significant role in postoperative pain, a point that requires further verification

    Social media use and impact during the holiday travel planning process

    Through an empirical study among holiday travellers, residing in the Former Soviet Union Republics, this paper presents a comprehensive view of role and impact of social media on the whole holiday travel planning process: Before, during and after the trip, providing insights on usage levels, scope of use, level of influence and trust. Findings suggest that social media are predominantly used after holidays for experience sharing. It is also shown that there is a strong correlation between perceived level of influence from social media and changes made in holiday plans prior to final decisions. Moreover, it is revealed that user-generated content is perceived as more trustworthy when compared to official tourism websites, travel agents and mass media advertising

    Expanded NK cells from umbilical cord blood and adult peripheral blood combined with daratumumab are effective against tumor cells from multiple myeloma patients

    In this study we evaluated the potential of expanded NK cells (eNKs) from two sources combined with the mAbs daratumumab and pembrolizumab to target primary multiple myeloma (MM) cells ex vivo. In order to ascertain the best source of NK cells, we expanded and activated NK cells from peripheral blood (PB) of healthy adult donors and from umbilical cord blood (UCB). The resulting expanded NK (eNK) cells express CD16, necessary for carrying out antibody-dependent cellular cytotoxicity (ADCC). Cytotoxicity assays were performed on bone marrow aspirates of 18 MM patients and 4 patients with monoclonal gammopathy of undetermined significance (MGUS). Expression levels of PD-1 on eNKs and PD-L1 on MM and MGUS cells were also quantified. Results indicate that most eNKs obtained using our expansion protocol express a low percentage of PD-1+ cells. UCB eNKs were highly cytotoxic against MM cells and addition of daratumumab or pembrolizumab did not further increase their cytotoxicity. PB eNKs, while effective against MM cells, were significantly more cytotoxic when combined with daratumumab. In a minority of cases, eNK cells showed a detectable population of PD1+ cells. This correlated with low cytotoxic activity, particularly in UCB eNKs. Addition of pembrolizumab did not restore their activity. Results indicate that UCB eNKs are to be preferentially used against MM in the absence of daratumumab while PB eNKs have significant cytotoxic advantage when combined with this mAb
