    The Boston Harbor Cleanup

    Boston Harbor earned a widespread reputation as the dirtiest harbor in the nation during the 1988 presidential campaign. Well before that campaign began, though, efforts were under way to reduce the amount of pollution entering the harbor. The Massachusetts Water Resources Authority was created in 1985 to undertake a massive public works program — including construction of a 1.3 billion-gallon-per-day sewage treatment plant and a sludge fertilizer processing plant — to end the decades-old practice of dumping sewage wastes into the ocean. The program will also cause water and sewer charges to rise dramatically during a fifteen-year period. The project has raised a host of environmental and public policy issues: How should sludge by-products be disposed of or used by society? What is the proper placement of the effluent outfall for a sewage treatment plant of this magnitude discharging into Massachusetts Bay? What is the appropriate level of treatment to apply to wastewater? How can ratepayers be assured that their money is being spent on the highest environmental priorities ?     Acetylation-induced TDP-43 pathology is suppressed by an HSF1-dependent chaperone program

    TDP-43 pathology marks a spectrum of multisystem proteinopathies including amyotrophic lateral sclerosis, frontotemporal lobar degeneration, and sporadic inclusion body myositis. Surprisingly, it has been challenging to recapitulate this pathology, highlighting an incomplete understanding of TDP-43 regulatory mechanisms. Here we provide evidence supporting TDP-43 acetylation as a trigger for disease pathology. Using cultured cells and mouse skeletal muscle, we show that TDP-43 acetylation-mimics promote TDP-43 phosphorylation and ubiquitination, perturb mitochondria, and initiate degenerative inflammatory responses that resemble sporadic inclusion body myositis pathology. Analysis of functionally linked amyotrophic lateral sclerosis proteins revealed recruitment of p62, ubiquilin-2, and optineurin to TDP-43 aggregates. We demonstrate that TDP-43 acetylation-mimic pathology is potently suppressed by an HSF1-dependent mechanism that disaggregates TDP-43. Our study illustrates bidirectional TDP-43 processing in which TDP-43 aggregation is targeted by a coordinated chaperone response. Thus, activation or restoration of refolding mechanisms may alleviate TDP-43 aggregation in tissues that are uniquely susceptible to TDP-43 proteinopathies.TDP-43 aggregation is linked to various diseases including amyotrophic lateral sclerosis.     Nuclear Environments Inspection with Micro Aerial Vehicles: Algorithms and Experiments

    In this work, we address the estimation, planning, control and mapping problems to allow a small quadrotor to autonomously inspect the interior of hazardous damaged nuclear sites. These algorithms run onboard on a computationally limited CPU. We investigate the effect of varying illumination on the system performance. To the best of our knowledge, this is the first fully autonomous system of this size and scale applied to inspect the interior of a full scale mock-up of a Primary Containment Vessel (PCV). The proposed solution opens up new ways to inspect nuclear reactors and to support nuclear decommissioning, which is well known to be a dangerous, long and tedious process. Experimental results with varying illumination conditions show the ability to navigate a full scale mock-up PCV pedestal and create a map of the environment, while concurrently avoiding obstacles.Comment: 10 pages, ISER 201

    Modelflow Estimates of Stroke Volume Do Not Correlate With Doppler Ultrasound Estimates During Upright Posture

    Orthostatic intolerance affects 6080% of astronauts returning from longduration missions, representing a significant risk to completing missioncritical tasks. While likely multifactorial, a reduction in stroke volume (SV) represents one factor contributing to orthostatic intolerance during stand and head up tilt (HUT) tests. Current measures of SV during stand or HUT tests use Doppler ultrasound and require a trained operator and specialized equipment, restricting its use in the field. BeatScope (Finapres Medical Systems BV, The Netherlands) uses a modelflow algorithm to estimate SV from continuous blood pressure waveforms in supine subjects; however, evidence supporting the use of Modelflow to estimate SV in subjects completing stand or HUT tests remain scarce. Furthermore, because the blood pressure device is held extended at heart level during HUT tests, but allowed to rest at the side during stand tests, changes in the finger arterial pressure waveform resulting from arm positioning could alter modelflow estimated SV. The purpose of this project was to compare Doppler ultrasound and BeatScope estimations of SV to determine if BeatScope can be used during stand or HUT tests. Finger photoplethysmography was used to acquire arterial pressure waveforms corrected for hydrostatic fingertoheart height using the Finometer (FM) and Portapres (PP) arterial pressure devices in 10 subjects (5 men and 5 women) during a stand test while simultaneous estimates of SV were collected using Doppler ultrasound. Measures were made after 5 minutes of supine rest and while subjects stood for 5 minutes. Next, SV estimates were reacquired while each arm was independently raised to heart level, a position similar to tilt testing. Supine SV estimates were not significantly different between all three devices (FM: 68+/-20, PP: 71+/-21, US: 73+/-21 ml/beat). Upon standing, the change in SV estimated by FM (18+/-8 ml) was not different from PP (21+/-12), but both were significantly less than US (37+/-16 ml, p<.05). Raising finger BP devices to heart level caused no significant change in SV measured with any of the devices (FM: 1.5+/-19, PP: 1.7+/-26, US: 0.5+/-6), although variability was 36x greater as assessed by both blood pressure devices compared to US. Retrospective analysis of blood pressure data to assess SV in 11 supine subjects revealed significantly different estimates between methods (FM: 95+/-17, US: 75+/-32, p<.05), but the change in SV resulting from HUT was similar between methods (FM: 37+/-9, US: 40+/-18 ml). However, the correlation coefficient determined from pairs of SV estimated by US and FM was weak (r2=0.03). These data suggest Modelflow cannot be used in lieu of Doppler ultrasound to estimate SV during stand or HUT tests. Further investigation should focus on identifying factors contributing to differences between these measurement techniques in order to make use of a simple method for assessing beatbybeat changes in SV during postural changes, especially during field testing

    Hyperspectral Perfusion Monitoring of Irradiated Breast Patients

    Studies examining acute perfusion changes (month) in irradiated fields are limited. Hyperspectral imaging (HSI) is a novel method of scanning spectroscopy that provides direct measurement of cutaneous tissue perfusion that is non-invasive. In this clinical study, we examine the ability of HSI to assess cutaneous changes in skin perfusion during the acute period following irradiation in patients. Patients undergoing external beam breast conserving radiotherapy (n=15) or post-mastectomy radiation (n=3) were enrolled. Total treatment doses ranged between 42 Gy and 50 Gy. Baseline images were obtained before irradiation for bilateral breasts in each patient and then subsequently at each dose fraction. Skin reaction assessment was also performed on the patients. In the irradiated breast, total perfusion was found to increase prior to skin reaction formation and continued to steadily increase over the first 30 days in all patients. Skin reactions included erythema and dry desquamation starting at day 11. These findings suggest that HSI can identify early changes of tissue oxygenation and perfusion in acute radiation injury and may be able to predict the severity of such injuries. Future work will look at mitigating acute injury with topical applications and studying the perfusion changes in chronically irradiated skin

    3-Methyl-1-butanol production in Escherichia coli: random mutagenesis and two-phase fermentation

    Interest in producing biofuels from renewable sources has escalated due to energy and environmental concerns. Recently, the production of higher chain alcohols from 2-keto acid pathways has shown significant progress. In this paper, we demonstrate a mutagenesis approach in developing a strain of Escherichia coli for the production of 3-methyl-1-butanol by leveraging selective pressure toward l-leucine biosynthesis and screening for increased alcohol production. Random mutagenesis and selection with 4-aza-d,l-leucine, a structural analogue to l-leucine, resulted in the development of a new strain of E. coli able to produce 4.4 g/L of 3-methyl-1-butanol. Investigation of the host’s sensitivity to 3-methyl-1-butanol directed development of a two-phase fermentation process in which titers reached 9.5 g/L of 3-methyl-1-butanol with a yield of 0.11 g/g glucose after 60 h

    Catastrophic Bushfires, Indigenous Fire Knowledge and Reframing Science in Southeast Australia.

    The catastrophic 2019/2020 Black Summer bushfires were the worst fire season in the recorded history of Southeast Australia. These bushfires were one of several recent global conflagrations across landscapes that are homelands of Indigenous peoples, homelands that were invaded and colonised by European nations over recent centuries. The subsequent suppression and cessation of Indigenous landscape management has had profound social and environmental impacts. The Black Summer bushfires have brought Indigenous cultural burning practices to the forefront as a potential management tool for mitigating climate-driven catastrophic bushfires in Australia. Here, we highlight new research that clearly demonstrates that Indigenous fire management in Southeast Australia produced radically different landscapes and fire regimes than what is presently considered “natural”. We highlight some barriers to the return of Indigenous fire management to Southeast Australian landscapes. We argue that to adequately address the potential for Indigenous fire management to inform policy and practice in managing Southeast Australian forest landscapes, scientific approaches must be decolonized and shift from post-hoc engagement with Indigenous people and perspectives to one of collaboration between Indigenous communities and scientists
