    Highlights of meiotic genes in Arabidopsis thaliana

    Meiosis is a fascinating and complex phenomenon and, despite its central role in sexual plant reproduction, little is known on the molecular mechanisms involved in this process. We review the progress made in recent years using Arabidopsis thaliana mutants for isolating meiotic genes. In particular, emphasis is given on the description of mutants affecting either the regular commitment to meiosis, or the mechanisms of synapsis, recombination, and cytokinesis. We believe that the isolation of genes affecting some crucial meiotic events may represent the first step towards the practical use of meiotic genes in plant breeding. The introduction of deviations in the meiotic pathway into sexual crops will have important implications for the exploitation of apomixis and sexual polyploidization.     Data-Driven Estimation of Heavy-Truck Residual Value at the Buy-Back

    In a context of deep transformation of the entire automotive industry, starting from pervasive and native connectivity, commercial vehicles (heavy, light, and buses) are generating and transmitting much more data than passenger cars, with a much higher expected value, motivated by the higher costs of the vehicles and their added-value related businesses, such as logistics, freight, and transportation management. This paper presents a data-driven and unsupervised methodology to provide a descriptive model assessing the residual value estimates of heavy trucks subject to buy-back. A huge amount of telematics data characterizing the actual usage of commercial vehicles is jointly analyzed with different external conditions (e.g., altimetry), affecting the truck's performance to estimate the devaluation of the vehicle at the buy-back. The proposed approach has been evaluated on a large set of real-world heavy trucks to demonstrate its effectiveness in correctly assessing the real status of wear and residual value at the end of leasing contracts, to provide a few and quantitative insights through an informative, interactive and user-friendly dashboard to make a proper decision on the next business strategies to be adopted. The proposed solution has already been deployed by a private company within its data analytics services since (1) an interpretable descriptive model of the main factors/parameters and corresponding weights affecting the residual value is provided and (2) the experimental results confirmed the promising outcomes of the proposed data-driven methodology

    Quinic acids from Aster caucasicus and from transgenic callus expressing a beta-amyrin synthase.

    Several different classes of secondary metabolites, including flavonoids, triterpenoid saponins and quinic acid derivatives, are found in Aster spp. (Fam. Asteraceae). Several Aster compounds revealed biological as well as pharmacological activities. In this work, a phytochemical investigation of A. caucasicus evidenced the presence of quinic acid derivatives, as well as the absence of triterpene saponins. To combine in one species the production of different phytochemicals, including triterpenes, an Agrobacterium-mediated transformation of A. caucasicus was set up to introduce A. sedifolius β-amyrin synthase (AsOXA1)-encoding gene under the control of the constitutive promoter CaMV35S. The quali-quantitative analysis of transgenic calli with ectopic expression of AsOXA1 showed, in one sample, a negligible amount of triterpene saponins combined with higher amount of quinic acid derivatives as compared with the wild type callus

    Appropriateness of internal digital phantoms for monitoring the stability of the UBIS 5000 quantitative ultrasound device in clinical trials

    In bone status assessment, proper quality assurance/quality control is crucial since changes due to disease or therapeutic treatment are very small, in the order of 2-5%. Unlike for dual X-ray absorptiometry, quality control procedures have not been extensively developed and validated for quantitative ultrasound technology, limiting its use in longitudinal monitoring. While the challenge of developing an ideal anthropometric phantom is still open, some manufacturers use the concept of the internal digital phantom mimicking human characteristics to check the stability of their device. The objective of the study was to develop a sensitive model of quality control suitable for the correction of QUS patient data. In order to achieve this goal, we simulated a longitudinal device lifetime with both correct and malfunctioning behaviors. Then, we verified the efficiency of digital phantoms in detecting those changes and subsequently established the in vitro/in vivo relationship. This is the first time that an attempt to validate an internal digital phantom has made, and that this type of validation approach is used. The digital phantom (DP) was designed to mimic normal bone (BUAP2) and osteoporotic bone (BUAP1) properties. The DP was studied using the UBIS 5000 ultrasound device (DMS, France). Diverse malfunctions of the UBIS-5000 were simulated. Several series of measurements were performed on both BUAP1 and 2 and on 12 volunteers at each grade of malfunction. The effect of each simulated malfunction on in vivo and in vitro results was presented graphically by plotting the average BUA values against the percentage change from baseline. The change from baseline in BUA was modeled using linear regression, and the in vivo/in vitro ratio was obtained from the model. All experimentations influenced the measure of BUAP1 and 2 as well as the measure of our 12 volunteers. However, the degree of significance varied as a function of the severity of the malfunction, and the results also differed substantially in magnitude between in vivo and in vitro. Indeed, the DP was about 10 times more sensitive to variations of the transfer function than was the in vivo measurement, which is very reassuring. The sensitivity of the digital phantoms was reliable in the determination of simulated malfunctions of the UBIS-5000. The digital phantoms provided an accurate evaluation of the acoustic performance of the scanner, including the fidelity of transducers. In light of these results, the QC approach of the UBIS-5000 will be extremely simple to implement compared with other devices.     Empowering Commercial Vehicles through Data-Driven Methodologies

    In the era of “connected vehicles,” i.e., vehicles that generate long data streams during their usage through the telematics on-board device, data-driven methodologies assume a crucial role in creating valuable insights to support the decision-making process effectively. Predictive analytics allows anticipation of vehicle issues and optimized maintenance, reducing the resulting costs. In this paper, we focus on analyzing data collected from heavy trucks during their use, a relevant task for companies due to the high commercial value of the monitored vehicle. The proposed methodology, named TETRAPAC, offers a generalizable approach to estimate vehicle health conditions based on monitored features enriched by innovative key performance indicators. We discussed performance of TETRAPAC in two real-life settings related to trucks. The obtained results in both tasks are promising and able to support the company’s decision-making process in the planning of maintenance interventions

    Genome-wide analysis of histone modifiers in tomato: gaining an insight into their developmental roles

    BACKGROUND: Histone post-translational modifications (HPTMs) including acetylation and methylation have been recognized as playing a crucial role in epigenetic regulation of plant growth and development. Although Solanum lycopersicum is a dicot model plant as well as an important crop, systematic analysis and expression profiling of histone modifier genes (HMs) in tomato are sketchy. RESULTS: Based on recently released tomato whole-genome sequences, we identified in silico 32 histone acetyltransferases (HATs), 15 histone deacetylases (HDACs), 52 histone methytransferases (HMTs) and 26 histone demethylases (HDMs), and compared them with those detected in Arabidopsis (Arabidopsis thaliana), maize (Zea mays) and rice (Oryza sativa) orthologs. Comprehensive analysis of the protein domain architecture and phylogeny revealed the presence of non-canonical motifs and new domain combinations, thereby suggesting for HATs the existence of a new family in plants. Due to species-specific diversification during evolutionary history tomato has fewer HMs than Arabidopsis. The transcription profiles of HMs within tomato organs revealed a broad functional role for some HMs and a more specific activity for others, suggesting key HM regulators in tomato development. Finally, we explored S. pennellii introgression lines (ILs) and integrated the map position of HMs, their expression profiles and the phenotype of ILs. We thereby proved that the strategy was useful to identify HM candidates involved in carotenoid biosynthesis in tomato fruits. CONCLUSIONS: In this study, we reveal the structure, phylogeny and spatial expression of members belonging to the classical families of HMs in tomato. We provide a framework for gene discovery and functional investigation of HMs in other Solanaceae species

    RAAD: LIGHT-1 CubeSat's Payload for the Detection of Terrestrial Gamma-Ray Flashes

    The Rapid Acquisition Atmospheric Detector (RAAD), onboard the LIGHT-1 3U CubeSat, detects photons between hard X-rays and soft gamma-rays, in order to identify and characterize Terrestrial Gamma Ray Flashes (TGFs). Three detector configurations are tested, making use of Cerium Bromide and Lanthanum BromoChloride scintillating crystals coupled to photomultiplier tubes or Multi-Pixel Photon Counters, in order to identify the optimal combination for TGF detection. High timing resolution, a short trigger window, and the short decay time of its electronics allow RAAD to perform accurate measurements of prompt, transient events. Here we describe the overview of the detection concept, the development of the front-end acquisition electronics, as well as the ground testing and simulation the payload underwent prior to its launch on December 21st, 2021. We further present an analysis of the detector's in-orbit system behavior and some preliminary results.Comment: 19 pages, 15 figure

    A multipurpose X-ray fluorescence scanner developed for in situ analysis

    Abstract: Over the time, instrument transportability has become more and more important, especially in Cultural Heritage, as often artworks cannot be moved from their site, either because of the size or due to problems with permission issues, or simply because moving them to a laboratory is physically impossible, as e.g. in the case of mural paintings. For this reason, the INFN-CHNet, the network for Cultural Heritage studies of the Italian National Institute of Nuclear Physics (INFN), has developed an XRF scanner for in situ analyses. The instrument is the result of a wide collaboration, where different units of the network have been developing the diverse parts, then merged in a single system. The XRF scanner has been designed to be a four-season and green instrument. The control/acquisition/analysis software has been fully developed by our group, using only open-source software. Other strong points of the system are easiness of use, high portability, good performances and ultra-low radiation dispersion, which allows us to use even when the public can be present. It can run both with mains or on batteries, in the latter case with a maximum runtime longer than 10 h. It has a very low cost, when compared to commercial systems with equivalent performances, and easily replaceable components, which makes it accessible for a much wider portion of the interested community. The system has been thought and designed as an open system, suitable for further development/improvements, that can result interesting for non-conventional XRF analysis. The CHNet XRF scanner has proved to be really very well suited for applications in the Cultural Heritage field, as testified by the many recent applications.     Whole-genome re-sequencing of two Italian tomato landraces reveals sequence variations in genes associated with stress tolerance, fruit quality and long shelf-life traits

    Tomato is a high value crop and the primary model for fleshy fruit development and ripening. Breeding priorities include increased fruit quality, shelf life and tolerance to stresses. To contribute towards this goal, we re-sequenced the genomes of Corbarino (COR) and Lucariello (LUC) landraces, which both possess the traits of plant adaptation to water deficit, prolonged fruit shelf-life and good fruit quality. Through the newly developed pipeline Reconstructor, we generated the genome sequences of COR and LUC using datasets of 65.8M and 56.4M of 30–150bp paired-end reads, respectively. New contigs including reads that could not be mapped to the tomato reference genome were assembled, and a total of 43, 054 and 44, 579 gene loci were annotated in COR and LUC. Both genomes showed novel regions with similarity to Solanum pimpinellifolium and Solanum pennellii. In addition to small deletions and insertions, 2, 000 and 1, 700 single nucleotide polymorphisms (SNPs) could exert potentially disruptive effects on 1, 371 and 1, 201 genes in COR and LUC, respectively. A detailed survey of the SNPs occurring in fruit quality, shelf life and stress tolerance related-genes identified several candidates of potential relevance.
