The influence of the shape of Au nanoparticles on the catalytic current of fructose dehydrogenase
Graphite electrodes were modified with triangular (AuNTrs) or spherical (AuNPs) nanoparticles and further modified with fructose dehydrogenase (FDH). The present study reports the effect of the shape of these nanoparticles (NPs) on the catalytic current of immobilized FDH pointing out the different contributions on the mass transfer–limited and kinetically limited currents. The influence of the shape of the NPs on the mass transfer–limited and the kinetically limited current has been proved by using two different methods: a rotating disk electrode (RDE) and an electrode mounted in a wall jet flow-through electrochemical cell attached to a flow system. The advantages of using the wall jet flow system compared with the RDE system for kinetic investigations are as follows: no need to account for substrate consumption, especially in the case of desorption of enzyme, and studies of product-inhibited enzymes. The comparison reveals that virtually identical results can be obtained using either of the two techniques. The heterogeneous electron transfer (ET) rate constants (kS) were found to be 3.8 ± 0.3 s−1 and 0.9 ± 0.1 s−1, for triangular and spherical NPs, respectively. The improvement observed for the electrode modified with AuNTrs suggests a more effective enzyme-NP interaction, which can allocate a higher number of enzyme molecules on the electrode surface
SÃntesis de nanoprismas ramificados de oro con potenciales aplicaciones biotecnológicas

Este trabajo se basó en la sÃntesis de nanopartÃculas anisotrópicas de oro, utilizando vesÃculas a base de L-α-fosfatidilcolina (PC) y sal sódica de dimiristoilfosfatidilglicerol (DMPG-Na). Las partÃculas fueron purificadas por centrifugación y con la utilización de diferentes surfactantes, siendo el dodecilsulfato de sodio (SDS) el que presentó mejores resultados. Los nanotriángulos de oro (AuNTs)
purificados, fueron caracterizados por Dispersión Dinámica de la Luz (DLS, por sus siglas en inglés) y Microscopia Electrónica de Transmisión (TEM, por sus siglas en inglés). Los nanotriángulos de oro obtenidos mostraron un potencial Zeta de (-60,50 ± 2,55) mV y tamaños de partÃcula de (91,15 ± 2,79) nm. Mediante TEM se determinó que el 80 % de lasnpartÃculas corresponden a nanotriángulos y el resto a partÃculas esféricas. Los AuNTs presentan una longitud de bode entre 60 nm y 80 nm, mientras que el diámetro de las partÃculas esféricas está entre 60 nm y 70 nm. Los nanotriángulos de oro modificados con ramificaciones en su superficie fueron obtenidos mediante la incorporación de semillas (triángulos de oro) a una disolución de HAuCl4 y en presencia de diferentes aditivos como AgNO3 y ácido ascórbico. También, se evaluó la adición de haluros sobre la formación de ramificaciones. Las estructuras fueron evaluadas mediante UV-Vis-NIR y TEM. Se determinó que la adición de I- y Br- inhiben el crecimiento
de ramificaciones en la superficie de la partÃcula y mientras que el ión Cl- tiene un efecto contario. Los nanotriángulos de oro fueron modificados con un polÃmero termosensible a base de poli(isopropilacrilamida) con grupos tiol y carboxÃlico en sus extremos. Las AuNPs modificadas con el polÃmero presentan una disminución del diámetro hidrodinámico al aumentar la temperatura y un aumento del potencial Zeta en comparación con las partÃculas sin modificar. Estas partÃculas fueron además incubadas junto con la enzima β-glucuronidasa a una temperatura de 25 °C y bajo diferentes condiciones de pH y fuerza iónica. En este análisis tanto el potencial Zeta como los tamaños de partÃcula tienden a aumentar, indicando una interacción del AuNTs-polÃmero con la enzima.Currently, gold nanoparticles are an interesting alternative to be used in various applications. This work was based on the synthesis of anisotropic gold nanoparticles, using vesicles based on L-α-phosphatidylcholine (PC) and dimyristoylphosphatidylglycerol sodium salt (DMPG-Na). The particles were purified by centrifugation and with the use of different surfactants, being sodium dodecyl sulfate (SDS) the one that presented the best results. The purified gold nanotriangles (AuNTs) were characterized by Dynamic Light Scattering (DLS) and Transmission Electron Microscopy (TEM). The gold nanotriangles obtained showed a Zeta potential of (-60.50 ± 2.55) mV and particle sizes of (91.15 ± 2.79) nm. By means of TEM, it was determined that 80% of the nparticles correspond to nanotriangles and the rest to spherical particles. The AuNTs have a bode length between 60 nm and 80 nm, while the diameter of the spherical particles is between 60 nm and 70 nm. The modified gold nanotriangles with branches on their surface were obtained by incorporating seeds (gold triangles) into a HAuCl4 solution and in the presence of different additives such as AgNO3 and ascorbic acid. Also, the addition of halides on branching was evaluated. The structures were evaluated by UV-Vis-NIR and TEM. It was determined that the addition of I- and Br- inhibit the growth of branches on the surface of the particle and while the Cl- ion has a counter effect. The gold nanotriangles were modified with a thermosensitive polymer based on poly (isopropylacrylamide) with thiol and carboxylic groups at their ends. Polymer-modified AuNPs show a decrease in hydrodynamic diameter with increasing temperature and an increase in Zeta potential compared to unmodified particles. These particles were also incubated together with the enzyme β-glucuronidase at a temperature of 25 ° C and under different conditions of pH and ionic strength. In this analysis, both the Zeta potential and the particle sizes tend to increase, indicating an interaction of the AuNTs-polymer with the enzyme.Universidad Nacional, Costa RicaEscuela de QuÃmic
Biofabrication of Gold Nanotriangles Using Liposomes as a Dual Functional Reductant and Stabilizer
Negatively charged liposomes accomplished both functions as a reducing and stabilizing agent in the synthesis of gold nanotriangles (GNTs). Liposomes are based on a mixture of phospholipids phosphatidylcholine/phosphoglycerol, and they were used as a template phase to perform the GNTs. The method was evaluated under different conditions such as temperature, reaction time, phosphoglycerol chain length, and precursor concentration. Isotropic and anisotropic gold nanoparticles are formed simultaneously during the synthesis. Therefore, by combining centrifugation and depletion flocculation strategies, the sample was concentrated in terms of GNTs from 15% crude to 80% by using sodium dodecyl sulfate (SDS). As a result, a green colored dispersion was obtained containing highly purified, well-defined, negatively charged GNTs, where the edge length of most particles is centered in the range of 60-80 nm with an average thickness of 7.8 ± 0.1 nm. By this purification process, it was possible to highly increase the yield in terms of GNTs. Other surfactants [cetyltrimethylammonium chloride (CTAC), hexadecyltrimethylammonium bromide (CTAB), Tween 20, and dodecyldimethylammonium bromide] were evaluated during the purification stage, and both CTAB and CTAC show similar results to those obtained by using SDS. These GNTs are potential candidates for future applications in molecular imaging, photothermal therapy, drug delivery, biosensing, and photodynamic therapy.Los liposomas cargados negativamente cumplÃan ambas funciones como agente reductor y estabilizador en la sÃntesis de nanotriángulos de oro (GNT). Los liposomas se basan en una mezcla de fosfolÃpidos fosfatidilcolina / fosfoglicerol, y se usaron como fase molde para realizar las GNT. El método se evaluó en diferentes condiciones, como temperatura, tiempo de reacción, longitud de la cadena de fosfoglicerol y concentración de precursor. Las nanopartÃculas de oro isotrópicas y anisotrópicas se forman simultáneamente durante la sÃntesis. Por lo tanto, al combinar las estrategias de centrifugación y floculación por agotamiento, la muestra se concentró en términos de GNT del 15% en bruto al 80% mediante el uso de dodecilsulfato de sodio (SDS). Como resultado, se obtuvo una dispersión de color verde que contenÃa GNT altamente purificados, bien definidos y cargados negativamente, donde la longitud del borde de la mayorÃa de las partÃculas se centra en el rango de 60-80 nm con un grosor promedio de 7,8 ± 0,1 nm. Mediante este proceso de purificación, fue posible aumentar considerablemente el rendimiento en términos de GNT. Otros tensioactivos [cloruro de cetiltrimetilamonio (CTAC), bromuro de hexadeciltrimetilamonio (CTAB), Tween 20 y bromuro de dodeciltrimetilamonio] fueron evaluados durante la etapa de purificación, y tanto CTAB como CTAC muestran resultados similares a los obtenidos usando SDS.
