    How is artificial lighting affecting the catches in deep water rose shrimp trawl fishery of the Central Mediterranean Sea?

    The effect of artificial lights mounted on the headrope trawl net on the catch of deep water rose shrimp (Parapenaeus longirostris), European hake (Merluccius merluccius), and Atlantic horse mackerel (Trachurus trachurus) was tested in a survey carried out on-board a commercial trawler off the SW Sicilian coast. A total of 18 repeated nocturnal hauls, alternating without (control) and with (test) LED lights (10 green and 10 white) according to the fishers’ setup, were conducted. Overall, the test net catch rates were not significantly higher than those of the control net (Kruskal-Wallis test, p > 0.05), except for P. longirostris (p < 0.05). Conversely, the two-tailed Kolmogorov–Smirnov test revealed statistical differences in the size structure of P. longirostris, M. merluccius, and T. trachurus between the test and control nets (p < 0.05). Using generalised linear mixed models, the test net was found to yield higher catches of undersized individuals of the three species and adults of P. longirostris than the control net. Our study results are discussed in the context of the exploitation and management of Mediterranean trawl fisheries

    Interactive effects of fishing effort reduction and climate change in a central Mediterranean fishing area: Insights from bio-economic indices derived from a dynamic food-web model

    Disentangling the effects of mixed fisheries and climate change across entire food-webs requires a description of ecosystems using tools that can quantify interactive effects as well as bio-economic aspects. A calibrated dynamic model for the Sicily Channel food web, made up of 72 functional groups and including 13 fleet segments, was developed. A temporal simulation until 2050 was conducted to evaluate the bio-economic interactive effects of the reduction of bottom trawling fishing effort by exploring different scenarios that combine fishery and climate change. Our results indicate that direct and indirect effects produce a net increase in biomass of many functional groups with immediate decline of trawlers’ catches and economic incomes, followed by a long term increase mainly due to biomass rebuilding of commercial species which lasts 5-10 years after fishing reduction. Synergistic and antagonistic effects caused by changes in the fishing effort and in climate characterize a specific functional group’s response in biomass which, in turn, modulate also the catch and income of the other fleets, and especially of those sharing target resources. However, trawler’s intra-fleet competition is higher than the others fleet effects. In the medium term, the effects of fishing effort reduction are higher than those of climate change and seem to make exploitation of marine resources more sustainable over time and fishery processes more efficient by improving ecosystem health

    Distribution characteristics of pandalid shrimps (Decapoda: Caridea: Pandalidae) along the Central Mediterranean Sea

    The genus Plesionika is represented in the Mediterranean Sea by eight species, six of which,Plesionikaacanthonotus, P. antigai, P. edwardsii, P. gigliolii, P. heterocarpus and P. martia, are very common on muddy bottoms of the continental slope. During nine experimental trawl surveys a total of 29,038 individuals of these six pandalid species, was collected off the central western coasts of Italy (central Mediterranean) in order to study population structure and spatial distribution. P . antigaiinhabits the shelf break and upper slope; P. heterocarpus shows a wide bathymetric distribution, from the shelf-break to the upper slope; P. edwardsii and P. gigliolii occur in the upper slope. P. acanthonotus and P. martia occur in the deepest depths investigated. Segregation by size is revealed for the species that inhabit the same bottoms. The non-homogenous spatial distribution of Plesionikaspecies in the study area was probably related to the spatial differences in the magnitude of primary production in the area

    Identifying Persistent Hot Spot Areas of Undersized Fish and Crustaceans in Southern European Waters: Implication for Fishery Management Under the Discard Ban Regulation

    The recent establishment of the “landing obligation” under the reformed EU Common Fishery Policy has the twofold objective of reducing the excessive practice of discarding unwanted catch at sea and encouraging more selective and sustainable fisheries. Within this context, the awareness of the spatial distribution of potential unwanted catches is important for devising management measures aimed to decrease discards. This study analyzed the distribution of Hot Spot density areas of demersal fish and crustaceans below the Minimum Conservation Reference Size (MCRS) in four different southern European seas: continental Portuguese coast, Catalan Sea, South of Sicily, Liguria and northern Tyrrhenian Seas using both bottom trawl survey data and information on the spatial distribution of commercial fisheries. Critical areas for discarding were identified as zones where the highest densities of individuals below MCRS were consistently recorded throughout a series of years. Results clearly showed a patchy distribution of undersized individuals in each investigated area, highlighting the overlap between high density patches of both discards and fishing effort. The present findings provide a relevant knowledge for supporting the application of spatial-based management actions, such as the designation of Fisheries Restricted Areas (FRAs), in order to minimize the by-catch of undersized specimens and improve the sustainability of demersal fisheries

    Site-specific integration mediated by a hybrid adenovirus/adeno-associated virus vector

    Adenovirus (Ad) and adeno-associated virus(AAV) have attractive and complementary properties that canbe exploited for gene transfer purposes. Ad vectors are probablythe most efficient vehicles to deliver foreign genes both invitro and in vivo. AAV exhibits the unique ability to establishlatency by efficiently integrating at a specific locus of humanchromosome 19 (AAVS1). Two viral elements are necessaryfor the integration at AAVS1: Rep68y78 and the invertedterminal repeats (AAV-ITRs). In this study, we report thedevelopment of two helper-dependent adenoviral (HD) vectors,one carrying the Rep78 gene, the other an AAV-ITRflankedtransgene. Although Rep proteins have been demonstratedto interfere with Ad replication, HD Rep78 vector wassuccessfully amplified on serial passages in 293CRE4 cellswith a yield of 50–100 transducing units per cell. DNAintegration at the AAVS1 site also was demonstrated inhepatoma cells coinfected with the HD-expressing Rep78 andwith the second HD vector carrying a transgene flanked byAAV-ITRs. The high transduction efficiency, large cloningcapacity, and high titer of the HD, combined with the sitespecificintegration machinery provided by AAV-derived components,make the AdyAAV hybrid viruses a promising vehiclefor gene therap

    Mirroring everyday clinical practice in clinical trial design: a new concept to improve the external validity of randomized double-blind placebo-controlled trials in the pharmacological treatment of major depression

    Background: Randomized, double-blind, placebo-controlled trials constitute the gold standard in clinical research when testing the efficacy of new psychopharmacological interventions in the treatment of major depression. However, the blinded use of placebo has been found to influence clinical trial outcomes and may bias patient selection. Discussion: To improve clinical trial design in major depression so as to reflect clinical practice more closely we propose to present patients with a balanced view of the benefits of study participation irrespective of their assignment to placebo or active treatment. In addition every participant should be given the option to finally receive the active medication. A research agenda is outlined to evaluate the impact of the proposed changes on the efficacy of the drug to be evaluated and on the demographic and clinical characteristics of the enrollment fraction with regard to its representativeness of the eligible population. Summary: We propose a list of measures to be taken to improve the external validity of double-blind, placebocontrolled trials in major depression. The recommended changes to clinical trial design may also be relevant for other psychiatric as well as medical disorders in which expectations regarding treatment outcome may affect the outcome itself

    Il campionamento biologico delle catture/sbarcati commerciali, sezione C – Relative al mestiere ed E – Relative agli stock (Campbiol), nell’ambito del Programma Nazionale per la Raccolta Dati Alieutici (PNRDA) (Reg. Ce. N°199/2008; N°665/2008 e decisione della commissione N°949/2008) risulta di grande importanza per conoscere come agisce il prelievo dei diversi ”metiers” sulle diverse specie, in termini di variazioni dell’abbondanza e struttura demografica delle risorse da pesca. Il campionamento biologico risponde, quindi, principalmente alle seguenti esigenze: 1. Ricostruire la struttura demografica delle catture commerciali/sbarchi di ogni specie (in taglia/etĂ ), considerando tutti i ”mĂ©tiers” che incidono significativamente sull’ammontare globale delle catture. 2. Ricostruire il pattern di sfruttamento dei diversi ”mĂ©tiers” per le diverse specie. 3. Consentire lo studio dei fenomeni biologici rilevanti, quali la crescita ed il ciclo sessuale, che variano nel corso dell’anno. 4. Acquisire informazioni sulla struttura demografica con specifico riferimento allo stadio di maturazione gonadica (maturitĂ /taglia-etĂ ) ed alla relazione fra lunghezza e peso corporeo (taglia-peso/etĂ ).     Different impact of definitions of sarcopenia in defining frailty status in a population of older women with early breast cancer

    Sarcopenia is a geriatric syndrome characterized by losses of quantity and quality of skeletal muscle, which is associated with negative outcomes in older adults and in cancer patients. Different definitions of sarcopenia have been used, with quantitative data more frequently used in oncology, while functional measures have been advocated in the geriatric literature. Little is known about the correlation between frailty status as assessed by comprehensive geriatric assessment (CGA) and sarcopenia in cancer patients. We retrospectively analyzed data from 96 older women with early breast cancer who underwent CGAs and Dual X-ray Absorptiometry (DXA) scans for muscle mass assessment before cancer treatment at a single cancer center from 2016 to 2019 to explore the correlation between frailty status as assessed by CGA and sarcopenia using different definitions. Based on the results of the CGA, 35 patients (36.5%) were defined as frail. Using DXA Appendicular Skeletal Mass (ASM) or the Skeletal Muscle Index (SMI=ASM/heightˆ2), 41 patients were found to be sarcopenic (42.7%), with no significant difference in prevalence between frail and nonfrail subjects. Using the European Working Group on Sarcopenia in Older People (EWGSOP2) definition of sarcopenia (where both muscle function and mass are required), 58 patients were classified as “probably” sarcopenic; among these, 25 were sarcopenic and 17 “severely” sarcopenic. Only 13 patients satisfied both the requirements for being defined as sarcopenic and frail. Grade 3-4 treatment-related toxicities (according to Common Terminology Criteria for Adverse Events) were more common in sarcopenic and frail sarcopenic patients. Our data support the use of a definition of sarcopenia that includes both quantitative and functional data in order to identify frail patients who need tailored treatment
