    Application of alternative cell separation systems for the harvest of mammalian cell culture processes in a fully disposable single-use facility

    In a fully disposable facility where the use of continuous disk-stack centrifuges are not preferred, harvest processes based on conventional depth filtration become more challenging with increasing single-use bioreactor (SUB) size and higher density culture. Lower filterability due to high volume and high cell density diminishes the efficiency of the depth filtration. In addition, the use of larger depth filtration systems is constrained by the facility footprint. To address the challenge described, several alternative single-use harvest technologies were evaluated. A disposable centrifuge with a small foot print, low consumable cost, and ease of operation was tested. In a study using a 1kL SUB for harvest, the disposable centrifuge was compared to a continuous disk-stack centrifuge, both followed by conventional depth filtration. The best operating conditions, as well as the edges of failures, were found for the disposable centrifuge. When compared to the disk-stack centrifuge results showed that the operation can be performed in a wide range without impacting depth filtration area requirements and less depth filter area was needed. In addition to testing a disposable centrifuge, flocculation was evaluated and compared, followed by the depth filtration. This was found to be another low-cost alternative process, while also occupying less facility space. The flocculation technology was tested with different cell lines and results showed significant improvement in filterability and reduction of depth filter area compared to full traditional depth filtration train

    Model North Atlantic Treaty Organization (NATO) consists of several committees, each of whom draft policy relevant to their respective topics of discussion. The Committees debate for the course of the conference, drafting NATO policy to solve a multitude of issues. During this time, a simulated crisis also occurs, and each committee must respond to the updates from the crisis as well as focus on their original goals. After the final committee sessions of the conference, the final resolutions passed by each committee are sent to the North Atlantic Council (NAC), who works to approve all of them unanimously. The end result is a final communique, consisting of the unanimously approved policy from the body of the entire conference.     Landsat Imagery from a CubeSat: Results and Operational Lessons from the R3 Satellite\u27s First 18 Months in Space

    R3 is a 3-U CubeSat launched on a RocketLab Electron into a 500 km circular orbit at 85° inclination on December 16th, 2018. The spacecraft flies a multispectral sensor that takes data in the six Landsat visible and near infrared bands. The R3 sensor mates a custom refractive telescope with a Materion Precision Optics Landsat filter, and an ON Semiconductor fast-framing high-sensitivity Si CMOS array, to produce 50-km wide, 44-m resolution Landsat-like image strips. Data are taken in push-broom mode and are downlinked via a 100Mbps compact lasercom system. Frames are then co-added on the ground in time-delay-integration (TDI) fashion to increase signal-to-noise ratio and create multi-spectral Earth images from the compact sensor. The system is an engineering concept demonstration of a compact multispectral sensor in CubeSat form. We describe our ConOps, flight operations, sensor focus and alignment, initial imaging check out, and initial comparisons of R3 data to Landsat-8 imagery of the same Earth locations. RGB, color infrared, and normalized differential vegetation index (NDVI) products are compared between CUMULOS and Landsat-8. Results show good multispectral image quality from the CubeSat sensor, and illustrate the ability of R3 to detect vegetation and other features in a manner similar to Landsat, as well as the challenge in perfectly exposing all 6 VIS/NIR Landsat bands using our commercial 10-bit CMOS array. We also highlight the performance of the compact laser communications system which enabled the successful performance of this mission

    Entangled Rings

    Consider a ring of N qubits in a translationally invariant quantum state. We ask to what extent each pair of nearest neighbors can be entangled. Under certain assumptions about the form of the state, we find a formula for the maximum possible nearest-neighbor entanglement. We then compare this maximum with the entanglement achieved by the ground state of an antiferromagnetic ring consisting of an even number of spin-1/2 particles. We find that, though the antiferromagnetic ground state does not maximize the nearest-neighbor entanglement relative to all other states, it does so relative to other states having zero z-component of spin.Comment: 19 pages, no figures; v2 includes new results; v3 corrects a numerical error for the case N=

    Diagnosis and management of functional tic-like phenomena

    Over the past 3 years, a global phenomenon has emerged characterized by the sudden onset and frequently rapid escalation of tics and tic-like movements and phonations. These symptoms have occurred not only in youth known to have tics or Tourette syndrome (TS), but also, and more notably, in youth with no prior history of tics. The Tourette Association of America (TAA) convened an international, multidisciplinary working group to better understand this apparent presentation of functional neurological disorder (FND) and its relationship to TS. Here, we review and summarize the literature relevant to distinguish the two, with recommendations to clinicians for diagnosis and management. Finally, we highlight areas for future emphasis and research

    STepped exercise program for patients with knee OsteoArthritis (STEP-KOA): protocol for a randomized controlled trial

    Background: Physical therapy (PT) and other exercise-based interventions are core components of care for knee osteoarthritis (OA), but both are underutilized, and some patients have limited access to PT services. This clinical trial is examining a STepped Exercise Program for patients with Knee OsteoArthritis (STEP-KOA). This model of care can help to tailor exercise-based interventions to patient needs and also conserve higher resource services (such as PT) for patients who do not make clinically relevant improvements after receiving less costly interventions. Methods / Design: Step-KOA is a randomized trial of 345 patients with symptomatic knee OA from two Department of Veterans Affairs sites. Participants are randomized to STEP-KOA and Arthritis Education (AE) Control groups with a 2:1 ratio, respectively. STEP-KOA begins with 3 months of access to an internet-based exercise program (Step 1). Participants not meeting response criteria for clinically meaningful improvement in pain and function after Step 1 progress to Step 2, which involves bi-weekly physical activity coaching calls for 3 months. Participants not meeting response criteria after Step 2 progress to in-person PT visits (Step 3). Outcomes will be assessed at baseline, 3, 6 and 9 months (primary outcome time point). The primary outcome is the Western Ontario and McMasters Universities Osteoarthritis Index (WOMAC), and secondary outcomes are objective measures of physical function. Linear mixed models will compare outcomes between the STEP-KOA and AE control groups at follow-up. We will also evaluate patient characteristics associated with treatment response and conduct a cost-effectiveness analysis of STEP-KOA. Discussion: STEP-KOA is a novel, efficient and patient-centered approach to delivering exercise-based interventions to patients with knee OA, one of the most prevalent and disabling health conditions. This trial will provide information on the effectiveness of STEP-KOA as a novel potential model of care for treatment of OA
