    The effect of cognitive training on impulse control among Methamphetamine addicts in the Western Cape

    Magister Psychologiae - MPsychSubstance use addiction is a debilitating and destructive human disorder that affects millions of people worldwide. Of all the provinces in South Africa, the Western Cape has the highest rate of MA use. This highly addictive stimulant, locally known as 'tik', has multiple physiological, psychological, and social effects on the user. The effects are associated with neurocognitive deficits that include deficiencies in working memory and high rates of delay discounting. Current neuropsychopharmacology literature seems to suggest that changes in neurotransmitter functioning and particular brain areas occur that contribute to some of the addictive behaviours associated with chronic MA use. New evidence is emerging that working memory training can help to improve rates of impulsivity in those addicted to MA by strengthening cognitive control. The aim of this project was to establish whether differences in impulse control existed in a sample of 33 male patients at a Western Cape drug rehabilitation centre who received either working memory training with standard drug rehabilitation and or standard drug rehabilitation only. Data was collected with a self-report impulsivity scale (BIS – 11) and analysed using inferential statistics. The results suggest that working memory training, when paired with a standard rehabilitation program, has superior effects in decreasing self-reported rates of impulsivity when compared to standard rehabilitation only. These findings suggest that working memory training may serve as a useful addition to improving impulsivity rates in MA rehabilitation treatment. Further research on a larger scale is required to investigate the findings of this project

    Serogrouping and sulphonamide sensitivity of Neisseria meningitidis isolates from the south-western Cape

Since then, against a backdrop of a relatively low incidence with winter exacerbation, epidemic periods have occurred at 10 - IS-year intervals. During the 1978 - 1979 epidemic more than 95% of isolates at Tygerberg Hospital were of serogroup B and only 5% were resistant to sulphonamides.     Immunolocalization of KATP channel subunits in mouse and rat cardiac myocytes and the coronary vasculature.

    BACKGROUND: Electrophysiological data suggest that cardiac KATP channels consist of Kir6.2 and SUR2A subunits, but the distribution of these (and other KATP channel subunits) is poorly defined. We examined the localization of each of the KATP channel subunits in the mouse and rat heart. RESULTS: Immunohistochemistry of cardiac cryosections demonstrate Kir6.1 protein to be expressed in ventricular myocytes, as well as in the smooth muscle and endothelial cells of coronary resistance vessels. Endothelial capillaries also stained positive for Kir6.1 protein. Kir6.2 protein expression was found predominantly in ventricular myocytes and also in endothelial cells, but not in smooth muscle cells. SUR1 subunits are strongly expressed at the sarcolemmal surface of ventricular myocytes (but not in the coronary vasculature), whereas SUR2 protein was found to be localized predominantly in cardiac myocytes and coronary vessels (mostly in smaller vessels). Immunocytochemistry of isolated ventricular myocytes shows co-localization of Kir6.2 and SUR2 proteins in a striated sarcomeric pattern, suggesting t-tubular expression of these proteins. Both Kir6.1 and SUR1 subunits were found to express strongly at the sarcolemma. The role(s) of these subunits in cardiomyocytes remain to be defined and may require a reassessment of the molecular nature of ventricular KATP channels. CONCLUSIONS: Collectively, our data demonstrate unique cellular and subcellular KATP channel subunit expression patterns in the heart. These results suggest distinct roles for KATP channel subunits in diverse cardiac structures

    Carriage of Haemophilus influenzae in Cape Town children

    Little is known about the epideIDiology of Haemophilus influenzae infections in South Africa. This study was designed to determine the prevalence, serotype distribution, antimicrobial susceptibility pattern and effect of age and hospitalisation on the carriage of H. influenzae in 322 Cape Town children.The overall and type b specific carriage rates in normal children (N =107) were 45,8% and 4,7% respectively. The yield following nasopharyngeal culture was twice that following throat culture (P < 0,001). Children hospitalised with tuberculosis (N =62) had significantly greater carriage rates, 66,1% and 37,1% respectively (P =0,02). Institutionalised mentally handicapped children (N =77) and children with tuberculosis attending an outpatient clinic (N =76) had lower carriage rates (P < 0,02). Antimicrobial resistance was a major problem only in children hospitalised with tuberculosis (rifampicin 100%, penicillin 43,9%, erythromycin 85,4%, co-trimoxazole 82,9%). This universal resistance to rifampicin has not been reported previously. There was no difference in the mean age of children with positive or negative cultures, with the exception of those hospitalised with tuberculosis. In this group children infected with type b were much younger (mean 19,7 months) than those with other and non-typeableinfections (32,1 months) and the non-infected(50,1 months) (P =0,04). Duration of hospitalisation or outpatient therapy in the patients with tuberculosis did not influence carriage rates.We conclude that carriage of H. influenzae in normal children is similar to that reported from other countries and that carriage, particularly of type b, in children hospitalised with tuberculosis was of significance and probably contributed to an outbreak of multi-resistant invasive H. influenzae disease in this group

    Propagation of Radiosonde Pressure Sensor Errors to Ozonesonde Measurements

    Several previous studies highlight pressure (or equivalently, pressure altitude) discrepancies between the radiosonde pressure sensor and that derived from a GPS flown with the radiosonde. The offsets vary during the ascent both in absolute and percent pressure differences. To investigate this problem further, a total of 731 radiosonde-ozonesonde launches from the Southern Hemisphere subtropics to Northern mid-latitudes are considered, with launches between 2005 - 2013 from both longer-term and campaign-based intensive stations. Five series of radiosondes from two manufacturers (International Met Systems: iMet, iMet-P, iMet-S, and Vaisala: RS80-15N and RS92-SGP) are analyzed to determine the magnitude of the pressure offset. Additionally, electrochemical concentration cell (ECC) ozonesondes from three manufacturers (Science Pump Corporation; SPC and ENSCI-Droplet Measurement Technologies; DMT) are analyzed to quantify the effects these offsets have on the calculation of ECC ozone (O3) mixing ratio profiles (O3MR) from the ozonesonde-measured partial pressure. Approximately half of all offsets are 0.6 hPa in the free troposphere, with nearly a third 1.0 hPa at 26 km, where the 1.0 hPa error represents 5 persent of the total atmospheric pressure. Pressure offsets have negligible effects on O3MR below 20 km (96 percent of launches lie within 5 percent O3MR error at 20 km). Ozone mixing ratio errors above 10 hPa (30 km), can approach greater than 10 percent ( 25 percent of launches that reach 30 km exceed this threshold). These errors cause disagreement between the integrated ozonesonde-only column O3 from the GPS and radiosonde pressure profile by an average of +6.5 DU. Comparisons of total column O3 between the GPS and radiosonde pressure profiles yield average differences of +1.1 DU when the O3 is integrated to burst with addition of the McPeters and Labow (2012) above-burst O3 column climatology. Total column differences are reduced to an average of -0.5 DU when the O3 profile is integrated to 10 hPa with subsequent addition of the O3 climatology above 10 hPa. The RS92 radiosondes are superior in performance compared to other radiosondes, with average 26 km errors of -0.12 hPa or +0.61 percent O3MR error. iMet-P radiosondes had average 26 km errors of -1.95 hPa or +8.75 percent O3MR error. Based on our analysis, we suggest that ozonesondes always be coupled with a GPS-enabled radiosonde and that pressure-dependent variables, such as O3MR, be recalculated-reprocessed using the GPS-measured altitude, especially when 26 km pressure offsets exceed 1.0 hPa 5 percent

    Impact of oral meloxicam administered alone or in combination with gabapentin on experimentally induced lameness in beef calves

    This study examined the pharmacokinetics and analgesic effect of oral meloxicam (MEL) administered alone or in combination with gabapentin (GABA) in an experimental bovine lameness model. Eighteen male British × Continental beef calves aged 4 to 6 mo and weighing 297 to 392 kg were randomly assigned to receive either 1) 0.5 mg/kg lactose monohydrate placebo (PLBO; n = 6), 2) 0.5 mg/kg MEL (n = 6), or 3) 0.5 mg/kg MEL combined with 15 mg/kg GABA (MEL-GABA; n = 6) once daily for 4 d. The first treatment was administered 4 h after a chemical synovitis/arthritis was induced with injection of 15 mg amphotericin B into the left hind lateral distal interphalangeal joint. Changes in activity were evaluated continuously with pedometers. Contact force, contact area, contact pressure, impulse, and stride length were recorded once daily with a pressure mat and visual lameness scores were determined by a masked observer using a 5-point scale. Cortisol and drug concentrations were determined daily by immunoassay and HPLC-mass spectrometry, respectively. Outcomes were compared statistically using a random effects mixed model and analysis of covariance. There was a positive association between lameness scores and serum cortisol concentrations (P = 0.02) and a negative association between lameness score and step count (P \u3c 0.0001), total force (P = 0.001), force applied to the lateral claw (P= 0.02), contact pressure (P = 0.005), and impulse of the lateral claw (P = 0.01). Step count was greater in MEL calves compared with PLBO (P = 0.008) and MEL-GABA (P = 0.04) calves. Impulse was greater in the MEL-GABA calves compared with the PLBO calves (P = 0.03). There was an inverse relationship between plasma MEL concentrations and lameness score (P = 0.02) and a positive association between MEL concentrations and force applied to the lateral claw (P = 0.03), total contact pressure (P = 0.03), and impulse on the lateral claw (P = 0.02). There was a tendency towards a positive association between GABA concentrations, total impulse, and impulse on the lateral claw (P = 0.08) and a negative associate between GABA concentrations and step count (P = 0.08). The results of this study suggest that MEL administered alone or in combination with GABA reduced the severity of lameness in calves following induction of lameness with amphotericin B. These findings have implications for developing analgesic protocols in lame calves that address both production and welfare concerns
