    Wide band observations of the X-ray burster GS 1826-238

    GS 1826-238 is a well-studied X-ray bursting neutron star in a low mass binary system. Thermal Comptonisation by a hot electron cloud is a widely accepted mechanism accounting for its high energy emission, while the nature of most of its soft X-ray output is not completely understood. A further low energy component is typically needed to model the observed spectra: pure blackbody and Comptonisation-modified blackbody radiation by a lower temperature (a few keV) electron plasma were suggested to explain the low energy data. We studied the steady emission of GS 1826-238 by means of broad band (X to soft Gamma-rays) measurements obtained by the INTEGRAL observatory in 2003 and 2006. The newly developed, up-to-date Comptonisation model CompTB is applied for the first time to study effectively the low-hard state variability of a low-luminosity neutron star in a low-mass X-ray binary system. We confirm that the 3-200 keV emission of \GS is characterised by Comptonisation of soft seed photons by a hot electron plasma. A single spectral component is sufficient to model the observed spectra. At lower energies, no direct blackbody emission is observed and there is no need to postulate a low temperature Compton region. Compared to the 2003 measurements, the plasma temperature decreased from 20 to 14 keV in 2006, together with the seed photons temperature. The source intensity was also found to be 30% lower in 2006, whilst the average recurrence frequency of the X-ray bursts significantly increased. Possible explanations for this apparent deviation from the typical limit-cycle behaviour of this burster are discussed.Comment: 6 pages, 2 figures. Accepted for publication in A&

    Diphtheria toxin-like ADP-ribosyltransferases 1 and 5 (ARTD-1, ARTD-5) are poly ADP-ribose enzymes (PARP) involved in non-homologous end-joining (NHEJ), which is the major pathway of double-strand break (DSB) repair. In addition, ARTD-5, or Tankyrase (TNKS), is a positive regulator of the WNT signaling implicated in the development and biological behavior of many neoplasms, such as Medulloblastoma (MB), in which radiotherapy is an essential part of the treatment. The use of radiosensitizing agents may improve the therapeutic index in MB patients by increasing the efficacy of radiotherapy, while reducing toxicity to the neuroaxis. ARTD-5 seems to be a good molecular target for improving the current treatment of MB. In this study, we used the small molecule XAV939, a potent ARTD-5 inhibitor with a slight affinity for ARTD-1, in different human MB cell lines. XAV939 inhibited the WNT pathway and DNA-PKcs in our MB cells, with many biological consequences. The co-administration of XAV939 and ionizing radiations (IR) inhibited MB cells proliferation and clonogenic capacity, decreased their efficacy in repairing DNA damage, and increased IR-induced cell mortality. In conclusion, our in vitro data show that XAV939 could be a very promising small molecule in MB treatment, and these results lay the basis for further in vivo studies with the aim of improving the current therapy available for MB patients

    Weighted multivariate curve resolution – alternating least squares based on sample relevance

    Alternating least squares, within the multivariate curve resolution framework has seen a lot of practical applications and shows its distinction with its relatively simple and flexible implementation. However, the limitations of least squares should be considered carefully when deviating from the standard assumed data structure. Within this work we highlight the effects of noise in the presence of minor components, and we propose a novel weighting scheme within the weighted multivariate curve-resolution-alternating least squares framework, to resolve it. Two simulated and one Raman imaging case is investigated, by comparing the novel methodology against standard multivariate curve resolution-alternating least squares and essential spectral pixel selection. A trade-off is observed between current methods, while the novel weighting scheme demonstrates a balance, where the benefits of the previous two methods are retained

    Burst-properties as a function of mass accretion rate in GX 3+1

    GX 3+1 is a low-mass X-ray binary that is persistently bright since its discovery in 1964. It was found to be an X-ray burster twenty years ago proving that the compact object in this system is a neutron star. The burst rate is so low that only 18 bursts were reported prior to 1996. The Wide Field Cameras on BeppoSAX have, through a dedicated monitoring program on the Galactic center region, increased the number of X-ray bursts from GX 3+1 by 61. Since GX 3+1 exhibits a slow (order of years) modulation in the persistent flux of about 50%, these observations opens up the unique possibility to study burst properties as a function of mass accretion rate for very low burst rates. This is the first time that bursts are detected from GX 3+1 in the high state. From the analysis we learn that all bursts are short with e-folding decay times smaller than 10 s. Therefore, all bursts are due to unstable helium burning. Furthermore, the burst rate drops sixfold in a fairly narrow range of 2-20 keV flux; we discuss possible origins for this.Comment: 9 pages and 7 figures.     Food chain traceability, identification of adulterations, and the control of labeling compliance are topic that require the evaluation of the foodstuff in its entirety: in this respect, more and more researchers are investigating the possibility of using multidimensional or hyphenated techniques for the fingerprinting of the products. However, these techniques produce data structures that are multidimensional as well and that require proper chemometric approaches for data processing (multi-way data analysis). In this Chapter, the state-of-the-art approaches for the classification of multiway data will be discussed theoretically and compared on case studies coming form the food authenticity context, such as the traceability of extra virgin olive oils of protected denomination of origin and table wines

    Acquisizione magnetica marina dell\u27alto strutturale del Mt. Vercelli (Mar Tirreno Centrale) e del golfo di Napoli: descrizione dei rilievi e primi risultati

    Durante la Campagna Oceanografica Ver2010, effettuata nel mese di maggio 2010, sono stati acquisiti due set di dati geomagnetici marini in alta risoluzione relativi alle aree del Mt. Vercelli (Mar Tirreno Centrale) e del Golfo di Napoli. Tale attività di misura è stata resa possibile grazie alla collaborazione tecnica scientifica tra l’Unità di Progetto “Geofisica e Tecnologie Marine” dell’Istituto Nazionale di Geofisica e Vulcanologia (INGV) e l’Istituto Idrografico della Marina (IIM). La proficua collaborazione è sancita dall’accordo di cooperazione “Co.Na.Ge.M” (Coordinamento Nazionale per la Geofisica Marina). Nello specifico, i due rilievi sono stati svolti mediante l’impiego della Nave Idro-oceanografica “Aretusa”, unità navale dell’Istituto Idrografico della Marina con scafo a catamarano in vetroresina e, pertanto, particolarmente adatta a rilievi di tipo geomagnetico. L’attività di acquisizione geofisica del Monte Vercelli rientra nel Progetto TySec – Prin 2007 che coinvolge l’Università degli Studi di Genova, l’Università Politecnica delle Marche e l’Istituto Nazionale di Geofisica e Vulcanologia. L’obiettivo del progetto è quello di valutare le relazioni tra le caratteristiche morfologiche e idrodinamiche dell’area del seamount e l’ecosistema ad esso correlato. In questo lavoro proponiamo i risultati preliminari del survey geomagnetico e batimetrico della struttura sommersa del Vercelli

    A new bursting X-ray transient: SAX J1750.8-2900

    We have analysed in detail the discovery measurements of the X-ray burster SAX J1750.8-2900 by the Wide Field Cameras on board BeppoSAX in spring 1997, at a position ~1.2 degrees off the Galactic Centre. The source was in outburst on March 13th when the first observation started and showed X-ray emission for ~ 2 weeks. A total of 9 bursts were detected, with peak intensities varying from ~ 0.4 to 1.0 Crab in the 2-10 keV range. Most bursts showed a fast rise time (~ 1s), an exponential decay profile with e-folding time of ~ 5s, spectral softening during decay, and a spectrum which is consistent with few keV blackbody radiation. These features identify them as type-I X-ray bursts of thermonuclear origin. The presence of type-I bursts and the source position close to the Galactic Centre favours the classification of this object as a neutron star low mass X-ray binary. X-ray emission from SAX J1750.8-2900 was not detected in the previous and subsequent Galactic bulge monitoring, and the source was never seen bursting again.Comment: 13 pages, 3 Postscript figures, aaspp4 styl

    Transcriptomic signatures of neuronal differentiation and their association with risk genes for autism spectrum and related neuropsychiatric disorders.

    Genes for autism spectrum disorders (ASDs) are also implicated in fragile X syndrome (FXS), intellectual disabilities (ID) or schizophrenia (SCZ), and converge on neuronal function and differentiation. The SH-SY5Y neuroblastoma cell line, the most widely used system to study neurodevelopment, is currently discussed for its applicability to model cortical development. We implemented an optimal neuronal differentiation protocol of this system and evaluated neurodevelopment at the transcriptomic level using the CoNTeXT framework, a machine-learning algorithm based on human post-mortem brain data estimating developmental stage and regional identity of transcriptomic signatures. Our improved model in contrast to currently used SH-SY5Y models does capture early neurodevelopmental processes with high fidelity. We applied regression modelling, dynamic time warping analysis, parallel independent component analysis and weighted gene co-expression network analysis to identify activated gene sets and networks. Finally, we tested and compared these sets for enrichment of risk genes for neuropsychiatric disorders. We confirm a significant overlap of genes implicated in ASD with FXS, ID and SCZ. However, counterintuitive to this observation, we report that risk genes affect pathways specific for each disorder during early neurodevelopment. Genes implicated in ASD, ID, FXS and SCZ were enriched among the positive regulators, but only ID-implicated genes were also negative regulators of neuronal differentiation. ASD and ID genes were involved in dendritic branching modules, but only ASD risk genes were implicated in histone modification or axonal guidance. Only ID genes were over-represented among cell cycle modules. We conclude that the underlying signatures are disorder-specific and that the shared genetic architecture results in overlaps across disorders such as ID in ASD. Thus, adding developmental network context to genetic analyses will aid differentiating the pathophysiology of neuropsychiatric disorders

    DATA FUSION APPROACHES IN SPECTROSCOPIC CHARACTERIZATION AND CLASSIFICATION OF PDO WINE VINEGARS

    Spain is one of the major producers of high-quality wine vinegars having three protected designations of origin (a.k.a. PDOs): "Vinagre de Jerez", "Vinagre de Condado de Huelva" and "Vinagre de Montilla-Moriles". Their high prices due to their high quality and their high production costs explain the need for developing an adequate quality control technique and the interest in extensive characterization in order to capture the identity of each denomination. In this framework, methodologies based on non-targeted techniques, such as spectroscopies, are becoming popular in food authentication. Thus, for improving vinegar quality assessment, fusion of data blocks obtained from the same samples but different analytical techniques could be a good strategy, since the quantity and quality of sample knowledge could be enhanced providing new insights into the differentiation of vinegars. Therefore, the aim of this manuscript is the development of a multi-platform methodology and a model able to classify the Spanish wine vinegar PDOs. Sixty-five PDO wine vinegars were analyzed by four spectroscopic techniques: Fourier transform mid-infrared spectroscopy (MIR), near infrared spectroscopy (NIR), multidimensional fluorescence spectroscopy (EEM) and proton nuclear magnetic resonance (1H-NMR). Two different data fusion strategies were evaluated: Mid-level data fusion with different preprocessing, and Common Component and Specific Weights analysis multiblock method. Exploratory and classification analysis on the data from individual techniques were also performed and compared with data fusion models. The data fusion models improved the classification, providing a more efficient differentiation, than the models based on single methods, and supporting the approach to combine these methods to achieve synergies for an optimized PDO differentiation
