    The field of molecular fecal source tracking in the water environment has developed rapidly since the first PCR assays for general and host-­‐specific Bacteroides 16s rRNA markers were published. Numerous host-­‐specific molecular markers and PCR assays have been developed, adding greater specificity, sensitivity and quantitative methods to the array of options. The public demand for readying methods for transfer to the commercial lab, so that they may be used to generate data for public utilities, citizen action groups and regulatory agencies, has fueled the development of an entire new research community. These methods, however plentiful, have not found community agreement and there is no consensus concerning the appropriate implementation of molecular fecal source tracking in the field. Some issues plaguing the implementation include imperfect marker specificity, environmental variability, DNA extraction variability, PCR inhibition and high cost of molecular analysis. This thesis presents an approach for locating hot spots of human fecal pollution in an urban watershed by using published methodologies for the collection of molecular fecal source tracking data along with a tiered watershed screening tool for cost reduction and two data normalization techniques which ameliorate several known sources of error and strengthen the efficacy of watershed application

    Linguistic terrioriality under stress:European perspectives

    This article revisits a well-known dichotomy (the ‘territorial’ and ‘personal’ principles) and develops a four-element classification of state approaches(from the most generous to the most menacing, from the perspective of speakers of minority languages). The article examines the implications for language policy of geographically dispersed or spatially concentrated patterns of distribution of speakers of particular languages. We begin by exploring the general literature on language policy, focusing in particular on the territorial and personal principles, the use of ‘threshold rules’ at municipal and other subnational levels, and the hybrid language regimes that are often a consequence of sociolinguistic complexity. We consider the extent to which responses to linguistic diversity across Europe may be understood by reference to these principles and categories. We explain why we have selected particular case studies (the Baltic republics, Transylvania, Switzerland, Belgium and Ireland) for further exploration. We conclude that, notwithstanding the value of the typologies we consider, real-life cases are almost invariably more complex, with states implementing policies that defy categorisation, that may change over time, and that may treat different language minorities by reference to different principles

    Prescribing 6-weeks of running training using parameters from a self-paced maximal oxygen uptake protocol

    The self-paced maximal oxygen uptake test (SPV) may offer effective training prescription metrics for athletes. This study aimed to examine whether SPV-derived data could be used for training prescription. Twenty-four recreationally active male and female runners were randomly assigned between two training groups: (1) Standardised (STND) and (2) Self-Paced (S-P). Participants completed 4 running sessions a week using a global positioning system-enabled (GPS) watch: 2 × interval sessions; 1 × recovery run; and 1 × tempo run. STND had training prescribed via graded exercise test (GXT) data, whereas S-P had training prescribed via SPV data. In STND, intervals were prescribed as 6 × 60% of the time that velocity at [Formula: see text] ([Formula: see text]) could be maintained (T ). In S-P, intervals were prescribed as 7 × 120 s at the mean velocity of rating of perceived exertion 20 ( RPE20). Both groups used 1:2 work:recovery ratio. Maximal oxygen uptake ([Formula: see text]), [Formula: see text], T RPE20, critical speed (CS), and lactate threshold (LT) were determined before and after the 6-week training. STND and S-P training significantly improved [Formula: see text] by 4 ± 8 and 6 ± 6%, CS by 7 ± 7 and 3 ± 3%; LT by 5 ± 4% and 7 ± 8%, respectively (all P < .05), with no differences observed between groups. Novel metrics obtained from the SPV can offer similar training prescription and improvement in [Formula: see text], CS and LT compared to training derived from a traditional GXT

    Punnett Square or Protein Production? The Expert-Novice Divide for Conceptions of Genes and Gene Expression

    Concepts of molecular biology and genetics are difficult for many biology undergraduate students to master yet are crucial for deep understanding of how life works. By asking students to draw their ideas, we attempted to uncover the mental models about genes and gene expression held by biology students (n = 23) and experts (n = 18) using semistructured interviews. A large divide was identified between novice and expert conceptions. While experts typically drew box-and-line representations and thought about genes as regions of DNA that were used to encode products, students typically drew whole chromosomes rather than focusing on gene structure and conflated gene expression with simple phenotypic outcomes. Experts universally described gene expression as a set of molecular processes involving transcription and translation, whereas students often associated gene expression with Punnett squares and phenotypic outcomes. Follow-up survey data containing a ranking question confirmed students’ alignment of their mental models with the images uncovered during interviews (n = 156 undergraduate biology students) and indicated that Advanced students demonstrate a shift toward expert-like thinking. An analysis of 14 commonly used biology textbooks did not show any relationship between Punnett squares and discussions of gene expression, so it is doubtful students’ ideas originate directly from textbook reading assignments. Our findings add to the literature about mechanistic reasoning abilities of learners and provide new insights into how biology students think about genes and gene expression

    Time-to-exhaustion trials (TTE) are used in a laboratory setting to measure endurance performance. However, there is some concern with their ecological validity compared with time-trials (TT). Consequently, we aimed to compare cycling performance in TTE and TT where the duration of the trials was matched. Seventeen trained male cyclists completed three TTE at 80, 100 and 105% of maximal aerobic power (MAP). On a subsequent visit they performed three TT over the same duration as the TTE. Participants were blinded to elapsed time, power output, cadence and heart rate (HR). Average TTE was 865 ± 345 s, 165 ± 98 s and 117 ± 45 s for the 80, 100 and 105% trials respectively. Average power output was higher for TTE (294 ± 44 W) compared to TT (282 ± 43 W) at 80% MAP (P0.05). There was no difference in cadence, HR, or RPE for any trial (P>0.05). Critical power (CP) was also higher when derived from TTE compared to TT (P<0.01). It is concluded that TTE results in a higher average power output compared to TT at 80% MAP. When determining CP, TTE rather than TT protocols appear superior

This study investigated the relevance of the matching hypothesis for anxiety interventions in tennis. Elite junior tennis players (N = 100; Age: M = 13.9 yr., SD = 1.8 yr.) completed the Competitive State Anxiety Inventory-2 before and after one of four randomly assigned intervention strategies approximately one hour prior to competition at a National Junior Championship. A two-factor multivariate analysis of variance (group x time) with repeated measures on the time factor gave no significant main effect by group but indicated significant reductions in somatic anxiety and cognitive anxiety and a significant increase in self-confidence following intervention. A significant group by time interaction emerged for self-confidence. The results question the need to match intervention strategy to the mode of anxiety experienced

    An exonic splicing enhancer in human IGF-I pre-mRNA mediates recognition of alternative exon 5 by the serine-arginine protein splicing factor-2/alternative splicing factor

    The human IGF-I gene has six exons, four of which are alternatively spliced. Variations in splicing involving exon 5 may occur, depending on the tissue type and hormonal environment. To study the regulation of splicing to IGF-I exon 5, we established an in vitro splicing assay, using a model pre-mRNA containing IGF-I exons 4 and 5 and part of the intervening intron. Using a series of deletion mutants, we identified an 18-nucleotide purine-rich splicing enhancer in exon 5 that increases the splicing efficiency of the upstream intron from 6 to 35%. We show that the serine-arginine protein splicing factor-2/alternative splicing factor specifically promotes splicing in cultured cells and in vitro and is recruited to the spliceosome in an enhancer-specific manner. Our findings are consistent with a role for splicing factor-2/alternative splicing factor in the regulation of splicing of IGF-I alternative exon 5 via a purine-rich exonic splicing enhancer
