The application of Multivariate Curve Resolution methods (MCR-ALS and PARAFAC) into multi-wavelength fluorescence spectroscopy, molecular absorption and ultra-performance liquid chromatography coupled to mass spectrometry data was evaluated as a new and complementary approach for the initial analysis of the biosynthesis of fluorescent natural products. To demonstrate the usefulness of this methodology, violacein biosynthesis by Chromobacterium violaceum was used as an example. Violacein is a secondary metabolite that exhibits a broad spectrum of biological activities and the understanding of its biosynthetic pathway can be of great interest for drug design. Therefore, a culture of C. violaceum was grown in a bioreactor from which aliquots were collected every two hours for subsequent analysis. In the first part of this work, the excitation¿emission matrices of the samples have revealed the dynamic behavior of the fluorophores involved in this biosynthesis. The parallel factor analysis (PARAFAC) method was applied to resolve the fluorescence profiles where six pure emission and excitation spectra were obtained and their relationship with violacein biosynthesis established. In the second part, the multivariate curve resolution procedure based on alternating least squares optimization (MCR-ALS) was used for extracting information from fused data (UV-VIS absorption, fluorescence and ultra-performance liquid chromatography with mass spectrometry) gathered during the violacein biosynthesis. Five chemical components were resolved from which spectral and temporal information were obtained. Compounds with molecular masses 453, 465 and 479 u, until now unreported as intermediate of this biosynthesis, were identified. Finally, the methodology based on the application of curve resolution methods PARAFAC and MCR-ALS has proven to be a unique and powerful tool to achieve a deeper insight of the biosynthesis of natural productsDoutoradoFísico-QuímicaDoutor em Ciência

In this work, the application of multivariate curve resolution-alternating least squares (MCR-ALS) is proposed for extracting information from multitechnique fused multivariate data (UV-VIS absorption, fluorescence, and liquid chromatography-mass spectrometry) gathered during the biosynthesis of violacein pigment. Experimental data sets were pretreated and arranged in a row-wise augmented data matrix before their chemometric investigation. Five different chemical components were resolved. Kinetic and spectral information about these components were obtained and their relationship with violacein biosynthesis was established. Three new chemical compounds with molar masses of 453, 465, and 479 u, until now not reported in the literature, were identified and proposed as intermediates in the biosynthesis of other indolocarbazoles. The precursor (tryptophan), one intermediate (deoxyviolacein), and the final product (violacein) of violacein biosynthesis were identified and characterized using the proposed approach. The chemometric procedure based on the MCR-ALS method has proved to be a powerful tool to investigate violacein biosynthesis and its application can be easily extended to the study of other bioprocesses.4051293-30

Theoretical Study Of Acid-catalyzed Hydrolysis Of Epoxides.

A theoretical study of propylene oxide acid-catalyzed hydrolysis was performed by investigation of the S(N)1 and S(N)2-like mechanisms. By using chemometric tools, hierarchical cluster analysis (HCA), and principal component analysis (PCA), the MP2/6-311++G** level of theory was selected from HF, MP2, and DFT as the best method to describe the geometry of the basic skeleton (oxirane). At this level of theory, geometry optimizations, vibrational frequencies, intrinsic reaction coordinate (IRC), and other thermodynamic calculations have shown that the borderline S(N)2 mechanism is more favorable than pure S(N)2 and S(N)1 mechanisms in the gas phase. In the S(N)1 mechanism, the existence of the typical carbocation was not observed, and furthermore, the possibility of epoxide conversion to a protonated aldehyde was indicated, even in the presence of a water molecule (nucleophile). The Chelpg charge distribution of the reactants, steric hindrance, synchronous bond breaking-formation and trajectory angle of nucleophilic attack are discussed for pure and borderline S(N)2 mechanisms. Solvation effect calculations indicate that the pure S(N)2 mechanism is more favorable than borderline S(N)2 and S(N)1 mechanisms. This is discussed in terms of hydrogen bond formation.1145187-9

PARAFAC: adjustment for modeling consumer study covering probiotic and conventional yogurt

The feasibility of the parallel factors model (PARAFAC) as a modeling tool for consumer sensory acceptance data of complex food matrices was investigated. The attributes of aroma, flavor, taste, texture, and overall liking of probiotic and conventional yogurts were evaluated by 120 consumers using a 9-point hedonic hybrid scale. Six yogurt samples were used: three were prototypes supplemented with a glucose oxidase/glucose system, which is a potential oxygen scavenging system, and the other three were commercial yogurts. The yogurts supplemented with the glucose oxidase/glucose system presented similar sensory acceptance towards commercial probiotic and conventional yogurts (p > 0.05), besides having garnered lower mean scores for the sensory attributes. Appearance and overall liking were the most significant attributes for the first two components of the PARAFAC model. Finally, the advantage of using this method to explore the intrinsic nature of consumer sensory data is discussed and compared to principal component analysis (PCA)45121121
